    Differential Responses of Human Regulatory T Cells (Treg) and Effector T Cells to Rapamycin

    Background: The immunosuppressive drug rapamycin (RAPA) promotes the expansion of CD4+ CD25highFoxp3+ regulatory\ud T cells via mechanisms that remain unknown. Here, we studied expansion, IL-2R-c chain signaling, survival pathways and resistance to apoptosis in human Treg responding to RAPA.\ud Methodology/Principal Findings: CD4+CD25+ and CD4+CD25neg T cells were isolated from PBMC of normal controls (n = 21)\ud using AutoMACS. These T cell subsets were cultured in the presence of anti-CD3/CD28 antibodies and 1000 IU/mL IL-2 for 3 to 6 weeks. RAPA (1–100 nM) was added to half of the cultures. After harvest, the cell phenotype, signaling via the PI3K/ mTOR and STAT pathways, expression of survival proteins and Annexin V binding were determined and compared to values obtained with freshly-separated CD4+CD25high and CD4+CD25neg T cells. Suppressor function was tested in co-cultures with autologous CFSE-labeled CD4+CD25neg or CD8+CD25neg T-cell responders. The frequency and suppressor activity of Treg were increased after culture of CD4+CD25+ T cells in the presence of 1–100 nM RAPA (p,0.001). RAPA-expanded Treg were largely CD4+CD25highFoxp3+ cells and were resistant to apoptosis, while CD4+CD25neg T cells were sensitive. Only Treg upregulated anti-apoptotic and down-regulated pro-apoptotic proteins. Treg expressed higher levels of the PTEN protein than CD4+CD25neg cells. Activated Treg6RAPA preferentially phosphorylated STAT5 and STAT3 and did not utilize the PI3K/ mTOR pathway.\ud Conclusions/Significance: RAPA favors Treg expansion and survival by differentially regulating signaling, proliferation and sensitivity to apoptosis of human effector T cells and Treg after TCR/IL-2 activation

    Medial Temporal Lobe Activity for Recognition of Recent and Remote Famous Names: an Event-Related fMRI Study

    Previous neuroimaging studies examining recognition of famous faces have identified activation of an extensive bilateral neural network [Gorno Tempini, M. L., Price, C. One model of hippocampal functioning in autobiographical, episodic memory retrieval argues that the hippocampal complex remains active in retrieval tasks regardless of time or age of memory (multiple trace theory, MTT), whereas another proposal posits that the hippocampal complex plays a time-limited role in retrieval of autobiographical memories. The current event-related fMRI study focused on the medial temporal lobe and its response to recognition judgments of famous names from two distinct time epochs (1990s and 1950s) in 15 right-handed healthy older adults (mean age = 70 years). A pilot study with an independent sample of young and older subjects ensured that the stimuli were representative of a recent and remote time period. Increased MR signal activity was observed on a bilateral basis for both the hippocampus and parahippocampal gyrus (PHG) during recognition of familiar names from both the recent and remote time periods when compared to non-famous names. However, the impulse response functions in the right hippocampus and right PHG demonstrated a differential response to stimuli from different time epochs, with the 1990s names showing the greatest MR signal intensity change, followed by the 1950s names, followed by foils. The finding that recognition of famous names produced significant bilateral MTL activation regardless of time epoch relative to foils provides support for the MTT model. However, the finding of a temporal gradient in the right MTL also provides support for the HC model, given the greater MTL response associated with recently famous names relative to remotely famous names

    Temporally Graded Activation of Neocortical Regions in Response to Memories of Different Ages

    The temporally graded memory impairment seen in many neurobehavioral disorders implies different neuroanatomical pathways and/or cognitive mechanisms involved in storage and retrieval of memories of different ages. A dynamic interaction between medial-temporal and neocortical brain regions has been proposed to account for memory\u27s greater permanence with time. Despite considerable debate concerning its time-dependent role in memory retrieval, medial-temporal lobe activity has been well studied. However, the relative participation of neocortical regions in recent and remote memory retrieval has received much less attention. Using functional magnetic resonance imaging, we demonstrate robust, temporally graded signal differences in posterior cingulate, right middle frontal, right fusiform, and left middle temporal regions in healthy older adults during famous name identification from two disparate time epochs. Importantly, no neocortical regions demonstrated greater response to older than to recent stimuli. Our results suggest a possible role of these neocortical regions in temporally dating items in memory and in establishing and maintaining memory traces throughout the lifespan. Theoretical implications of these findings for the two dominant models of remote memory functioning (Consolidation Theory and Multiple Trace Theory) are discussed

    Age-Related Functional Recruitment for Famous Name Recognition: An Event-Related fMRI Study

    Recent neuroimaging research shows that older adults exhibit recruitment, or increased activation on various cognitive tasks. The current study evaluated whether a similar pattern also occurs in semantic memory by evaluating age-related differences during recognition of Recent (since the 1990s) and Enduring (1950s to present) famous names. Fifteen healthy older and 15 healthy younger adults performed the name recognition task with a high and comparable degree of accuracy, although older adults had slower reaction time in response to Recent famous names. Event-related functional MRI showed extensive networks of activation in the two groups including posterior cingulate, right hippocampus, temporal lobe and left prefrontal regions. The Recent condition produced more extensive activation than the Enduring condition. Older adults had more extensive and greater magnitude of activation in 15 of 20 regions, particularly for the Recent condition (15 of 15; 7 of 15 also differed for Enduring); young adults did not show greater activation magnitude in any region. There were no group differences for non-famous names, indicating that age differences are task-specific. The results support and extend the existing literature to semantic memory tasks, indicating that older adult brains use functional recruitment to support task performance, even when task performance accuracy is high

    City project by river fronts on behalf of university international research

    Universities are dealing with strategies in order to improve their performances in terms of ranking classification, struggling for a good position. At the spatial planning and urban design domains the river fronts are very challenging territories to deal with, because they are sensitive areas, not only in environmental terms but also considering the requirements of the urban continuity. Urban projects for the places of contact in between the urban fabric and rivers are mainly focused on building issues and urban development questions, rather than of ecological approaches encouraging the contact of inhabitants with nature. Therefore, this research presents solutions of rethinking the territories of contact in between the urban fabric and the river water fronts, having as pivotal concern the ecological solutions. These urban projects are focused on the requalification solutions in order to ensure a better urban continuity from the buildings to the water front. The strategy is driven by the river activities, nautical sports, low density buildings, preservation of local heritage and paths for pedestrians. The case study is a quiet town in the central region of Portugal, well known by its thermal baths, which are coming from the roman times. Every year, the city of SΓ£o Pedro do Sul is visited by many tourists, looking for a very pleasant place, allowing them to improve their health condition going to the thermal baths, or the contact with nature going to the Vouga river water front. This is a city plenty of greenery, high natural potential, no pollution, and where the majority of dwellings are single houses. The main architectural landmark of the case study area is the old train station, a building from the 19 th century. In fact, since a long time ago, there are no more trains arriving to this city. Recently, the train track line was converted into an ecological corridor for pedestrians and bicycles. Besides of this effort, the place remains waiting for an urban rehabilitation opportunity as a whole, with several urban voids, buildings in bad status of conservation, car parking with no planning or a lack of functional diversity. Its huge potential regarding the presence of the river water front is, consequently, not preserved nor enhanced. In this sense, there was an international research, on behalf of the Urban Planning Unit at the Master Degree in Architecture at the University of Beira Interior, resulting from an agreement with the Municipality of SΓ£o Pedro do Sul. The urban project solutions were proposed in this academic environment comprising several teams of international students, not only coming from Portugal, but also from Greece, Brazil, Slovakia, Lithuania and Poland. There was a competition and monetary prizes to the best urban projects, sponsored by the city hall. This was a motivation for students along to the project process. The conclusions show that all the solutions are proposing strategies to preserve and to enhance the ecological identity of the place. Thus, this article will present the contents, options and solutions of the second best urban project, which was designed by a team of international students from Poland.     Regulation of Ras–MAPK pathway mitogenic activity by restricting nuclear entry of activated MAPK in endoderm differentiation of embryonic carcinoma and stem cells

    In response to retinoic acid, embryonic stem and carcinoma cells undergo differentiation to embryonic primitive endoderm cells, accompanied by a reduction in cell proliferation. Differentiation does not reduce the activation of cellular MAPK/Erk, but does uncouple mitogen-activated protein kinase (MAPK) activation from phosphorylation/activation of Elk-1 and results in inhibition of c-Fos expression, whereas phosphorylation of the cytoplasmic substrate p90RSK remains unaltered. Cell fractionation and confocal immunofluorescence microscopy demonstrated that activated MAPK is restricted to the cytoplasmic compartment after differentiation. An intact actin and microtubule cytoskeleton appears to be required for the restriction of MAPK nuclear entry induced by retinoic acid treatment because the cytoskeletal disrupting agents nocodazole, colchicine, and cytochalasin D are able to revert the suppression of c-Fos expression. Thus, suppression of cell proliferation after retinoic acid–induced endoderm differentiation of embryonic stem and carcinoma cells is achieved by restricting nuclear entry of activated MAPK, and an intact cytoskeleton is required for the restraint
