    Retrograde trafficking of Argonaute 2 acts as a rate-limiting step for de novo miRNP formation on endoplasmic reticulum–attached polysomes in mammalian cells

    microRNAs are short regulatory RNAs in metazoan cells. Regulation of miRNA activity and abundance is evident in human cells where availability of target messages can influence miRNA biogenesis by augmenting the Dicer1-dependent processing of precursors to mature microRNAs. Requirement of subcellular compartmentalization of Ago2, the key component of miRNA repression machineries, for the controlled biogenesis of miRNPs is reported here. The process predominantly happens on the polysomes attached with the endoplasmic reticulum for which the subcellular Ago2 trafficking is found to be essential. Mitochondrial tethering of endoplasmic reticulum and its interaction with endosomes controls Ago2 availability. In cells with depolarized mitochondria, miRNA biogenesis gets impaired, which results in lowering of de novo–formed mature miRNA levels and accumulation of miRNA-free Ago2 on endosomes that fails to interact with Dicer1 and to traffic back to endoplasmic reticulum for de novo miRNA loading. Thus, mitochondria by sensing the cellular context regulates Ago2 trafficking at the subcellular level, which acts as a rate-limiting step in miRNA biogenesis process in mammalian cells

    Dynamic Routing Framework for Wireless Sensor Networks

    Numerous routing protocols have been proposed for wireless sensor networks. Each such protocol carries with it a set of assumptions about the trafï¬c type that it caters to, and hence has limited interoperability. Also, most protocols are validated over workloads which only form a fraction of an actual deployment’s requirement. Most real world and commercial deployments, however, would generate multiple trafï¬c types simultaneously throughout the lifetime of the network. For example, most deployments would want all of the following to happen concurrently from the network: periodic reliable sense and disseminate, real time streams, patched updates, network reprogramming, query-response dialogs, mission critical alerts and so on. Naturally, no one routing protocol can completely cater to all of a deployments requirements. This chapter presents a routing framework that captures the communication intent of an application by using just three bits. The traditional routing layer is replaced with a collection of routing components that can cater to various communication patterns. The framework dynamically switches routing component for every packet in question. Data structure requirements of component protocols are regularized, and core protocol features are distilled to build a highly composable collection of routing modules. This creates a framework for developing, testing, integrating, and validating protocols that are highly portable from one deployment to another. Communication patterns can be easily described to lower layer protocols using this framework. One such real world application scenario is also investigated: that of predictive maintenance (PdM). The requirements of a large scale PdM are used to generate a fairly complete and realistic trafï¬c workload to drive an evaluation of such a framework

    Investigation of the AODV and the SDWCA QoS handling at different utilisation levels in adaptive clustering environments

    A simulation study using NS2 simulator using two main routing protocols with specific design parameters has been carried out to investigate the QoS main parameters such as throughput, delay, Jitter, Control Overhead, Number of packets, number of packets dropped and the rating overheads. The traffic is made of CBR slow video traffic. From the result it is noted that the SDWCA routing protocol outperforms the AODV routing protocols in the throughput, the delay and the jitter issues at different loading levels

    Dynamic Spectrum Access In Cognitive Radio Based Tactical Networks

    In this paper, we investigate how cognitive radio (CR) enabled devices can self-organize to form a tactical mesh network and operate on non-dedicated (secondary) spectrum. Each node in the network constantly senses the environment and maintains an up-to-date spectrum usage report. This report is used by a central controller (CC) to initialize the network formation. Then the other CR nodes gradually join the mesh network in a repeated, distributed manner. We provide the detailed steps for the mesh creation and also propose some refinements. We also compute the spectral efficiency that is achieved through our algorithm. Through simulation experiments, we study the effectiveness of the proposed schemes on mesh initialization latency, control signaling, collision rate during network initialization, and spectrum utilization. © 2009 IEEE
