    Characterization of arsenic-resistant endophytic Priestia megaterium R2.5.2 isolated from ferns in an arsenic-contaminated multi-metal mine in Vietnam

    Bioremediation is a biological process to remove or neutralize environmental pollutants. This study was carried out to investing at the efficacy of arsenic resistant endophytic bacteria isolated from Pteris vittata, Pityrogramma calomelanos, Blenchum orientale, and Nephrolepis exaltata, which grow in a highly arsenic (As) contamination mining site in Vietnam. Their segmented roots, stems, and leaves were homogenized separately and inoculated on LB agar plates containing 5mM As(III) and As(V). A total of 31 arsenic resistant endophytic strains were selected, in which strain R2.5.2 isolated from the root of P. calomelanos had the highest arsenic resistant capability. Strain R2.5.2 tolerated up to 320 mM and 160 mM of arsenate and arsenite, respectively. The strain developed well on a media of 0.1 5% NaCl, at 20-40ÂșC and pH 5 9, and actively utilized most of the sugar sources. It had a high IAA biosynthesis capacity with an average concentration of 19.14 mg/L, tolerated to 0.5-16 mM concentration of Ag+, Hg2+, Co2+, Ni2+, Cu2+, Cr4+, and reduced As(V). Based on 16s rDNA, R2.5.2 was identified as Priestia megaterium. The ars C gene coding for arsenate reductase catalyzing reduction of As(V) was successfully amplified in P. megaterium R2.5.2.  The selected strain may have potential use for bioremediation practice

    Anticaries activity of mangiferin isolated from Mangifera indica leaves in Vietnam

    Mangiferin (1) was isolated from Vietnamese Mangifera indica leaves. Its antimicrobial activities against the oral bacterium Streptococcus mutans GS-5 were evaluated in terms of inhibition of acid production and biofilm formation. The obtained results indicated that mangiferin at the concentration of 0.5 mM clearly inhibited acid production by this organism with a final pH value of 5.2 compared to 3.9 of the control. The reduction in biofilm biomass was found up to 92 % when it was treated with 1.0 mM mangiferin. Thus, mangiferin possesses moderate anticaries activity against S. mutans GS-5. Keywords.     Assessing decentralised policy implementation in Vietnam : The case of land recovery and resettlement in the Vung Ang Economic Zone

    From 2006 plans were implemented to create a deep-sea water port linked to an Economic Zone in the coastal Province of Ha Tinh, located in north central Vietnam. The multi-purpose Zone entitled ‘Vung Ang’, was to attract foreign investors, while the port would provide a link to nearby Laos and Thailand. The project obviously had large implications for the administrations at various levels of governance from Hanoi to the coastal communes and villages, but even more serious impacts on the people living in the affected areas. A large area of about 23,000 hectares was to be cleared, affecting the people of 9 communes, in some of which all inhabitants had to leave their houses and homesteads, to be relocated to completely new settlements about 10 miles inland. These tightly knit communities were not too happy with the prospect to leave their homes and land, the burial places of their ancestors, and the long term comforts of community support networks. While initial decision making process started at the highest levels of Vietnam Governance, the implementation of port and industrial park construction and the related relocation policy was delegated to Ha Tinh province, which is consistent with current decentralisation policies in Vietnam. Actual implementation was carried out by the affected District and Commune level officials – with support from the Communist Party led Mass Organisations – who were in charge of the planning and implementation of the relocation process. This entailed a complex and sensitive series of steps to inform affected households, prepare relocation areas and allocate compensation and alternative housing. This paper describes the implementation dynamics of relocation by depicting and assessing the roles of all stakeholders involved, including the impacts - for better or for worse – of the relocated households. It brings out the way local authorities dealt with affected people, including efforts linked to the ideal of grass-roots democracy. Key areas of contestation are uncovered, such as inadequate infrastructure and low compensation rates. The paper has a second objective to assess the degree to which decentralisation in Vietnam has been actually implemented, and how this affects policy making processes such as the Vung Ang port/industrial zone project. The paper concludes that the relocation policy was implemented in a fairly efficient and harmonious way – with a very intensive engagement of the entire provincial administrative machinery, but that it is too early to assess the livelihood opportunities of the relocated households

    Elimination of chemically synthesized pesticides, such as fungicides and nematicides, in agricultural products is a key to successful practice of the Vietnamese agriculture. We describe here the route for developing successful biostimulants based on members of the Bacillus subtilis species complex. A number of endospore-forming Gram-positive bacterial strains with antagonistic action against plant pathogens were isolated from Vietnamese crop plants. Based on their draft genome sequence, thirty of them were assigned to the Bacillus subtilis species complex. Most of them were assigned to the species Bacillus velezensis. Whole genome sequencing of strains BT2.4 and BP1.2A corroborated their close relatedness to B. velezensis FZB42, the model strain for Gram-positive plant growth-promoting bacteria. Genome mining revealed that at least 15 natural product biosynthesis gene clusters (BGCs) are well conserved in all B. velezensis strains. In total, 36 different BGCs were identified in the genomes of the strains representing B. velezensis, B. subtilis, Bacillus tequilensis, and Bacillus. altitudinis. In vitro and in vivo assays demonstrated the potential of the B. velezensis strains to enhance plant growth and to suppress phytopathogenic fungi and nematodes. Due to their promising potential to stimulate plant growth and to support plant health, the B. velezensis strains TL7 and S1 were selected as starting material for the development of novel biostimulants, and biocontrol agents efficient in protecting the important Vietnamese crop plants black pepper and coffee against phytopathogens. The results of the large-scale field trials performed in the Central Highlands in Vietnam corroborated that TL7 and S1 are efficient in stimulating plant growth and protecting plant health in large-scale applications. It was shown that treatment with both bioformulations resulted in prevention of the pathogenic pressure exerted by nematodes, fungi, and oomycetes, and increased harvest yield in coffee, and pepper.Peer Reviewe

    Chemical Components of Essential Oils From the Leaves of Seven Species Belonging to Rutaceae Family from Binh Chau-Phuoc Buu Nature Reserve, Vietnam

    Several plant species of the Rutaceae family are medicinal plants, oil bearing and food crops. To provide more information for utilization of some species of this family in Binh Chau-Phuoc Buu Nature Reserve, we extracted essential oils from the leaves of seven species of the Rutaceae family: Acronychia pedunculata (L.) Miq., Atalantia citroides Pierre ex Guillaumin, Clausena excavata Burm.f., Glycosmis pentaphylla (Retz.) DC., Luvunga scandens (Roxb.) Buch.-Ham. ex Wight & Arn, Melicope pteleifolia (Champ. ex Benth.) T.G. Hartley, and Micromelum sp., via hydrodistillation, and identified their components using GC/MS analysis. A total of 60 compounds were identified from essential oils of seven species. The main components of the essential oils isolated from five species, including A. pedunculata, C. excavata, M. pteleifolia, G. pentaphylla, and Micromelum sp., were caryophyllene (57.63% and 55.41% in A. pedunculata and C. excavata, respectively), 1,9-decadiyne (32.59%, M. pteleifolia), ÎČ-ocimene (23.10%, G. pentaphylla), and 3-carene (58.03%, Micromelum sp.). Additionally, this study revealed the chemical composition of essential oils of L. scandens and A. citroides for the first time. The main constituent of A. citroides was 7-oxabicyclo[4.1.0] heptane, 3-oxiranyl- (53.91%) and that of L. scandens was caryophyllene (34.66%). These findings provide the basis for further application of these species in medicine

    Prospects for Food Fermentation in South-East Asia, Topics From the Tropical Fermentation and Biotechnology Network at the End of the AsiFood Erasmus+Project

    Fermentation has been used for centuries to produce food in South-East Asia and some foods of this region are famous in the whole world. However, in the twenty first century, issues like food safety and quality must be addressed in a world changing from local business to globalization. In Western countries, the answer to these questions has been made through hygienisation, generalization of the use of starters, specialization of agriculture and use of long-distance transportation. This may have resulted in a loss in the taste and typicity of the products, in an extensive use of antibiotics and other chemicals and eventually, in a loss in the confidence of consumers to the products. The challenges awaiting fermentation in South-East Asia are thus to improve safety and quality in a sustainable system producing tasty and typical fermented products and valorising by-products. At the end of the “AsiFood Erasmus+ project” (www.asifood.org), the goal of this paper is to present and discuss these challenges as addressed by the Tropical Fermentation Network, a group of researchers from universities, research centers and companies in Asia and Europe. This paper presents current actions and prospects on hygienic, environmental, sensorial and nutritional qualities of traditional fermented food including screening of functional bacteria and starters, food safety strategies, research for new antimicrobial compounds, development of more sustainable fermentations and valorisation of by-products. A specificity of this network is also the multidisciplinary approach dealing with microbiology, food, chemical, sensorial, and genetic analyses, biotechnology, food supply chain, consumers and ethnology

    Awareness and preparedness of healthcare workers against the first wave of the COVID-19 pandemic: A cross-sectional survey across 57 countries.

    BACKGROUND: Since the COVID-19 pandemic began, there have been concerns related to the preparedness of healthcare workers (HCWs). This study aimed to describe the level of awareness and preparedness of hospital HCWs at the time of the first wave. METHODS: This multinational, multicenter, cross-sectional survey was conducted among hospital HCWs from February to May 2020. We used a hierarchical logistic regression multivariate analysis to adjust the influence of variables based on awareness and preparedness. We then used association rule mining to identify relationships between HCW confidence in handling suspected COVID-19 patients and prior COVID-19 case-management training. RESULTS: We surveyed 24,653 HCWs from 371 hospitals across 57 countries and received 17,302 responses from 70.2% HCWs overall. The median COVID-19 preparedness score was 11.0 (interquartile range [IQR] = 6.0-14.0) and the median awareness score was 29.6 (IQR = 26.6-32.6). HCWs at COVID-19 designated facilities with previous outbreak experience, or HCWs who were trained for dealing with the SARS-CoV-2 outbreak, had significantly higher levels of preparedness and awareness (p<0.001). Association rule mining suggests that nurses and doctors who had a 'great-extent-of-confidence' in handling suspected COVID-19 patients had participated in COVID-19 training courses. Male participants (mean difference = 0.34; 95% CI = 0.22, 0.46; p<0.001) and nurses (mean difference = 0.67; 95% CI = 0.53, 0.81; p<0.001) had higher preparedness scores compared to women participants and doctors. INTERPRETATION: There was an unsurprising high level of awareness and preparedness among HCWs who participated in COVID-19 training courses. However, disparity existed along the lines of gender and type of HCW. It is unknown whether the difference in COVID-19 preparedness that we detected early in the pandemic may have translated into disproportionate SARS-CoV-2 burden of disease by gender or HCW type
