    Stem Cell Therapy on Animal Model (Rattus Norvegicus) End Gestational TIME Exposed to Carbon Black as Observed From Caspase-3 Expression

    Background: Air pollution in the form of Diesel Exhaust Particles emerging from motor vehicles are harmful to health that have adverse reproductive health impacts, especially during pregnancy. The use of stem cells in treating white mice (Rattus norvegicus) exposed to carbon black can reveal the potential for treatment of placental impairment during pregnancy. Purpose: to demonstrate the effectivity of Rat Bone Marrow Mesenchymal Stem Cell therapy on rats (Rattus norvegicus) exposed to carbon black as observed from caspase-3 expression. Methods: This research uses a completely randomized design with factorial pattern. Forty-eight gravid female rats were divided into six treatment groups. Result: caspase-3 expression in each treatment showed no significant differences in the groups treated with RBMMSC in each gravid groups treatment (therapy GD 11 and GD 17) were exposed to carbon black and not treated with RBMMSC. The same are indicated by the normal trophoblast cells (cytotrophoblast and syncytiotrophoblast cells) in the RBMMSC treated group showed no significant difference with the group exposed to carbon black only. Conclusion: this research indicate that Rat Bone Marrow Mesenchymal Stem Cell therapy in Rattus norvegicus exposed to carbon black have not been able to reduce expression of caspase-3

    Profile of Crude Protein Tyrosine Kinase on Plasma Membrane of Merino Sheep Spermatozoa Using the Method of SDS-Page (Sodium Dodecyl Sulphate-Polyacrilamide Gel Electrophoresis)

    The research aims to find out profile of crude protein tyrosine kinase on plasma membrane of Merino sheep spermatozoa. The research used fresh semen sample collected using artificial vagina technique, next it was centrifuged to separate spermatozoa from seminal plasma. Then, spermatozoa (pellet) was isolated. After obtaining spermatozoa isolate, analysis on protein tyrosine kinase using SDS-PAGE method was conducted. The method to analyze protein using SDS-PAGE to separate protein based on molecular weight. Analysis on molecular weight was conducted by comparing result of isolate tapes with protein marker tapes. Running on SDS-PAGE gel isolate protein plasma membrane of Merino sheep spermatozoa in the research resulted in 13 protein tapes with average molecular weights of 115.44 kDa; 95.78 kDa; 78.22 kDa; 69.02 kDa; 62.33 kDa; 53.72 kDa; 40.56 kDa; 30.74 kDa; 21.16 kDa; 13.67 kDa; 10.85 kDa; 9.49 and 8.07 kDa kDa. From this result, the second tape with the molecular weight of 95.78 kDa was believed to be protein tyrosine kinase tape however it needs to be confirmed further using Western Blot method

    Detection Gene of Luteinizing Hormone Receptor (RLH) in Madrasin Cattle Using Polymerase Chain Reaction

    The aim of this study was to determine the profile of the Luteinizing Hormone Receptor (rLH) gene in Madrasin cross breed cattle. The sample of this study used blood samples from 3 breeds of cattle. There were Madrasin from Bangkalan City, Madura from Bangkalan City and Limousin from BET Cipelang Bogor. The sample criteria were used female cattle around 2,5 years old and ever partus (once). The genomic DNA was extracted for polymerase chain reaction (PCR), followed by sequencing with software BioEdit Vers. 8.0 to determine the sequence of nucleotide bases. The results of this study were that three samples detected fragments in the form of DNA with nucleotide base of 373 bp. The conclusion, gene rLH in Madrasin Cattle can detected using PCR

    Background: One of the partnerships of PT. Greenfields Indonesia is located on the slopes of Mount Kawi, precisely in Pijiombo, Ngadirenggo Village, Wlingi District, Blitar Regency. The tropical situation in this area is very humid and has high rainfall. It causes dairy cattle to be infected with helminths. Therefore, observations of the incidence rate of gastrointestinal helminthiasis in dairy cattle are needed. Purpose: To determine the prevalence of gastrointestinal helminths in dairy cattle in the partnership of PT. Greenfields Indonesia. Method: Fecal samples from 36 dairy cattle were collected and examined using native, sedimentation, and flotation techniques. Result: Indicated that the prevalence of gastrointestinal helminths in the dairy cattle was about 66,66%. There were 24 dairy cattle positively and 12 cattle negatively. Helminth species found included class Nematoda (Haemonchus contortus, Oesophagostomum radiatum, Bunostomum phlebotomum, Mecistocirrus digitatus, and Gaigeria pachyscelis) and Cestoda (Moniezia expansa). Conclusion: The incidence rate of helminthiasis was quite high dairy cattle in the partnership of PT. Greenfields Indonesia

    Etawa Goat Estrus Quality with Estrus Synchronization

    Background: Estrus synchronization is a way to get a herd of cattle estrus at the same time. This method can increase the reproductive success of livestock, thereby increasing production and population. Purpose: To determine the estrus quality from estrus synchronized Boerja goats, in Trawas District, Mojokerto Regency. Method: Estrus synchronization using an intra-vaginal sponge contains hormone progesterone, combined with the injection of the hormone GnRH. an intra-vaginal progesterone sponge was inserted for 14 days. After 14 days, the goats were injected with the GnRH hormone, then placed in a cage with the male and observed for estrus quality. Result: the average onset of estrus was 40.6 hours, the average color of the goat's vulvar mucosa when estrus was 2.3 (dark pink), the average presence of mucus was 1.9 (excess vulvar mucus), the average mucus color is 2.8 (white), and the average temperature increase of the vulva when the goat is in estrus is 0.4 ° C. Conclusion: The observations showed that the estrus quality of the goats was very diverse and not all goats were able to show the maximum estrus quality.     The Steroid Hormone Profile in Etawah Crossbreed Goat While Ovulation Induced Using the Selectsynch Method

    This study aimed to provide alternative information and solutions in an effort to increase reproductive productivity in etawah crossbreed goats (PE). The sample used in this study was 10 female PE with an average age of 2.5 - 3 years and primiparous at least . Ovulation induction was performed using 0.1 mg intra-muscular Gonadotropin-Releasing Hormone (GnRH), after seven days injection of Prostaglandin F2α (PGF2α) was given as much as 2.5 mg submucosa of the vulva, followed by a second injection of GnRH as much as 0,2 mg intramuscular in samples that have really experienced heat and selected selectively according to the signs of natural heat shown by the sample and followed by insemination as much as 2 doses or 0.50 ml of frozen semen. Blood sampling was performed at H0, H7th, H14th and H21th after insemination. All blood samples were collected and progesterone and estrogen hormone profiles were examined using the ELISA method. From the results of the ELISA test, the mean progesterone hormone profile H0 = 4.798 ng / ml, H7th = 4.887 ng / ml, H14th = 4.824 ng / ml, H21th = 5.148 ng / ml. The profile of the hormone estrogen at H0 = 19,461 pg / ml, H7th = 17,457 pg / ml, H14th = 18,248 pg / ml, H21th = 17,515 pg / ml. This study showed an increase in the levels of the progesterone hormone at H0 to H7th, then slightly decreased in H14th and a significant increase in H21th. For the estrogen hormone, there is a decrease from H0 to H7th then there is a slight increase in H14th and decreases again in H21th

    Efektifitas Daya Pisah Electric Separating Sperm (ESS) terhadap Spermatozoa Kromosom X dan Y pada Kambing Sapera

    Penelitian ini bertujuan untuk mengetahui efektifitas pemisahan alat Electric Separating Sperm (ESS) terhadap spermatozoa kromosom X dan Y pada sisi anoda dan katoda dengan variabel meliputi presentase viabilitas, motilitas, abnormalitas dan morfometri spermatozoa. Metode pengambilan semen menggunakan vagina buatan, kemudian dilakukan pemeriksaan makroskopis dan mikroskopis, apabila menunjukkan hasil yang baik dilanjutkan dengan pengenceran semen dengan Tris Kuning Telur dan dilakukan pemisahan. Terdapat tiga perlakuan pada penelitian ini yaitu P1 (3 menit), P2 (7 menit), P3 (10 menit) dengan enam kali pengulangan pada sisi anoda dan katoda. Data pada tiap variabel (viabilitas, motilitas dan abnormalitas) diperiksa menggunakan analisis statistik ANOVA dan untuk melihat adanya pemisahan spermatozoa kromosom X dan Y dilakukan pengukuran terhadap kepala menggunakan morfometri. Hasil pemisahan tertinggi yaitu pada P3 dengan perbandingan pemisahan X : Y sejumlah 65,00:35,00 pada sisi katoda, kemudian pada sisi anoda persentase perbandingan X : Y yaitu 59,00:41,00. Hasil kualitas spermatozoa setelah pemisahahn menunjukkan presentase viabilitas terbaik terdapat pada P1 yaitu 75,00a±2,00 pada sisi anoda dan 76,50a±2,17 pada sisi katoda. Motilitas spermatozoa P1 menunjukkan motilitas progresif spermatozoa dengan rataan 60,83a±2,04 pada anoda dan 61,67a±2,58 pada sisi katoda. Presentase abnormalitas tidak menunjukkan perbedaan yang signifikan antar perlakuan

    Penelitian ini merupakan penelitian deskriptif dengan menggunakan 20 itik Alabio betina siap bertelur dengan rentang usia 6 bulan dan untuk pemeriksaan kualitas telur untuk masing-masing kelompok 10 sampel. Pengamatan tingkat produksi telur dilakukan selama tiga bulan dengan 10 pengulangan, kualitas telur (ketebalan kulit telur, persentase kulit telur, persentase kulit telur, persentase albumin, persentase kuning telur, huagh unit, warna kuning telur, kadar protein dan lemak kandungan). Data produksi telur dan kualitas telur diuji menggunakan uji Anova. Hasil pengamatan rata-rata produksi harian telur 15.45 ± 0.12, 14.72 ± 0.10, 14.91 ± 0.09; produksi berat harian (kg/hari) 925.27 ± 7.50, 875.40 ± 6.32, 884 ± 5.03; hasil dari ketebalan kulit telur kualitas 0.33 ± 0.01, 0.33 ± 0.12, 0.33 ± 0.12; persentase kulit telur 11.32 ± 0.93, 11.28 ± 0.10, 11.36 ± 0.05; persentase albumin 55.70 ± 1.08, 56.66 ± 1.22, 56.12 ± 1.00; persen kuning telur 32.07 ± 1.24, 31.55 ± 1.48, 31.64 ± 1.20; Haugh unit 88.70 ± 6.21, 91.41 ± 6.70, 94.51 ± 5.06.     Persentase Motilitas dan Viabilitas Spermatozoa Kambing Sapera dalam Pengencer Tris Kuning Telur dan Susu Skim Kuning Telur Before Freezing

    Penelitian ini bertujuan untuk mengetahui perbedaan persentase motilitas dan viabilitas semen kambing Sapera terhadap evaluasi semen segar dan before freezing dengan penambahan pengencer tris kuning telur dan susu skim kuning telur. Metode penelitian ini adalah pengambilan semen, evaluasi semen segar (pemeriksaan makroskopis dan mikroskopis), pengenceran semen, gliserolisasi, ekuilibrasi dan evaluasi before freezing. Penelitian ini terdiri dari P0, P1 dan P2, masing-masing perlakuan terdiri dari sembilan ulangan. P0 adalah semen segar kambing Sapera, P1 adalah semen kambing Sapera+pengencer tris kuning telur dan P2 adalah semen kambing Sapera+pengencer susu skim kuning telur. Data yang diperoleh dengan Uji T2 sampel dependent, membandingkan P0 dan P1 serta P0 dan P2. Data menunjukkan perbedaan sangat nyata (p<0,01) pada perlakuan P0 dan P1 persentase motilitas 76,11%; 65,56% dan viabilitas 82,33%;72,22%. P0 dan P2 persentase motilitas 76,11%;61,11% dan viabilitas 82,33%;67,78%. Hasil tersebut menunjukkan bahwa penelitian persentase motilitas dan viabilitas spermatozoa kambing Sapera mengalami penurunan dari semen segar pada tris kuning telur dan susu skim kuning telur before freezing
