    Endothelial Differentiation of Human Stem Cells Seeded onto Electrospun Polyhydroxybutyrate/Polyhydroxybutyrate-Co-Hydroxyvalerate Fiber Mesh

    Tissue engineering is based on the association of cultured cells with structural matrices and the incorporation of signaling molecules for inducing tissue regeneration. Despite its enormous potential, tissue engineering faces a major challenge concerning the maintenance of cell viability after the implantation of the constructs. The lack of a functional vasculature within the implant compromises the delivery of nutrients to and removal of metabolites from the cells, which can lead to implant failure. In this sense, our investigation aims to develop a new strategy for enhancing vascularization in tissue engineering constructs. This study's aim was to establish a culture of human adipose tissue-derived stem cells (hASCs) to evaluate the biocompatibility of electrospun fiber mesh made of polyhydroxybutyrate (PHB) and its copolymer poly-3-hydroxybutyrate-co-3-hydroxyvalerate (PHB-HV) and to promote the differentiation of hASCs into the endothelial lineage. Fiber mesh was produced by blending 30% PHB with 70% PHB-HV and its physical characterization was conducted using scanning electron microscopy analysis (SEM). Using electrospinning, fiber mesh was obtained with diameters ranging 300 nm to 1.3 µm. To assess the biological performance, hASCs were extracted, cultured, characterized by flow cytometry, expanded and seeded onto electrospun PHB/PHB-HV fiber mesh. Various aspects of the cells were analyzed in vitro using SEM, MTT assay and Calcein-AM staining. The in vitro evaluation demonstrated good adhesion and a normal morphology of the hASCs. After 7, 14 and 21 days of seeding hASCs onto electrospun PHB/PHB-HV fiber mesh, the cells remained viable and proliferative. Moreover, when cultured with endothelial differentiation medium (i.e., medium containing VEGF and bFGF), the hASCs expressed endothelial markers such as VE-Cadherin and the vWF factor. Therefore, the electrospun PHB/PHB-HV fiber mesh appears to be a suitable material that can be used in combination with endothelial-differentiated cells to improve vascularization in engineered bone tissues

    Neste trabalho, buscou-se determinar o perfil de indústrias farmacêuticas dedicadas à fabricação e fracionamento de medicamentos fitoterápicos e oficinais no Estado do Rio de Janeiro por informações do CVS-RJ. Foram identificadas 48 empresas com atividades relacionadas à fabricação e/ou fracionamento e distribuição de medicamentos oficinais e fitoterápicos e analisadas no grau de adequação à legislação sanitária vigente, cumprimento de boas práticas de fabricação vigentes, bem como às questões relacionadas ao registro de produtos; as principais irregularidades foram o fluxo de pessoal e controle de qualidade adequado. Constatou-se que há atualmente empresas em situação: satisfatória (29,2%), satisfatórias com restrições (10,4%), insatisfatórias (6,2%), interditadas (39,6%), e solicitantes do cancelamento do processo por não terem condições para o cumprimento da RDC nº 210/03 (14,6%). Segundo o quadro atual, ainda é grande o número de empresas com atividades relacionadas à fabricação e/ou fracionamento e distribuição de medicamentos fitoterápicos e oficinais em fase de adequação ou em condições não adequadas no Rio de Janeiro. Ao mesmo tempo, é pouco factível a adequação, frente ao tipo de produto e porte financeiro, apontando um prognóstico desfavorável para o setor em questão.<br>In this study we evaluated the current profile of manufacturers of officinal medicines and phytotherapeutics in relation to the Brazilian regulatory legislation. Forty-eight industries involved in manufacture and distribution of officinal medicines and phytotherapeutics were identified in one hundred current administrative processes of the National Health Surveillance Agency in Brazil. The analysis of the inspection reports of these companies considered the first nine months after the implantation of Resolution RDC 210/03 and revealed five specific company profiles: satisfactory (29.2%), satisfactory with restrictions (10.4%), unsatisfactory (6.2%), interdicted (39.6%), and applying for cancellation due to lack of conditions for complying with Resolution RDC 210/03 (14.6%). The main irregularities found in these companies involved the operational flow, quality control and product registration issues. Our results revealed a great number of companies whose activities are related to the production of officinal medicines and phytotherapeutics but that are still not totally adequate to the industrial park of Rio de Janeiro. The perspectives for the future of this category of industries are thus not favorable because of their difficulties to meet the current requirements

    A osteoporose é uma doença cada vez mais diagnosticada em mulheres e homens de todo o mundo. Embora os esteróides sexuais sejam importantes na gênese da osteoporose, a inatividade física constitui um fator de risco. O exercício físico atua no osso por efeito direto, via força mecânica, ou indireto, mediado por fatores hormonais. Mas os mecanismos pelos quais a atividade física melhora a massa óssea ainda não são totalmente conhecidos. Baseando-se nos resultados que demonstram os efeitos benéficos da atividade física no tecido ósseo, a prática de esportes vem sendo cada vez mais indicada na prevenção e até mesmo no tratamento da osteoporose. O objetivo desta revisão é descrever os efeitos da atividade física no tecido ósseo normal e na prevenção e tratamento da osteoporose.<br>La osteoporosis es una enfermedad que cada vez más se diagnostica en mujeres y hombres de todo el mundo. Aunque los esteroides sexuales sean importantes en la génesis de la osteoporosis, la inactividad física constituye un factor de riesgo. Sin embargo la patogénesis por la que la actividad física mejora la masa ósea todavía no es totalmente conocida. Con base en los resultados que demuestran los efectos benéficos de la actividad física en el tejido óseo, la práctica de deportes viene siendo indicada cada vez más como medio de prevención y hasta incluso como tratamiento de la osteoporosis. Although the sexual steroids are important in the genesis of human osteoporosis, it is believed that the lack of physical activity constitutes a risk factor. Physical activity acts on the bone by direct effect via mechanical force, or indirect effect through hormonal factors. However, the mechanism through which physical activity improves the bone mass is not completely known. Sports practice has been increasingly recommended for prevention and even treatment of osteoporosis based on the results that have demonstrated the beneficial effects of physical activity on the bone tissue.     Soft object manipulation has recently gained popularity within the robotics community due to its potential applications in many economically important areas. Although great progress has been recently achieved in these types of tasks, most state-of-the-art methods are case-specific; They can only be used to perform a single deformation task (e.g. bending), as their shape representation algorithms typically rely on "hard-coded" features. In this paper, we present LaSeSOM, a new feedback latent representation framework for semantic soft object manipulation. Our new method introduces internal latent representation layers between low-level geometric feature extraction and high-level semantic shape analysis; This allows the identification of each compressed semantic function and the formation of a valid shape classifier from different feature extraction levels. The proposed latent framework makes soft object representation more generic (independent from the object's geometry and its mechanical properties) and scalable (it can work with 1D/2D/3D tasks). Its high-level semantic layer enables to perform (quasi) shape planning tasks with soft objects, a valuable and underexplored capability in many soft manipulation tasks. To validate this new methodology, we report a detailed experimental study with robotic manipulators.</p
