    A study on trypsin, Aspergillus flavus and Bacillus sp. protease inhibitory activity in Cassia tora (L.) syn Senna tora (L.) Roxb. seed extract

    <p>Abstract</p> <p>Background</p> <p>Proteases play an important role in virulence of many human, plant and insect pathogens. The proteinaceous protease inhibitors of plant origin have been reported widely from many plant species. The inhibitors may potentially be used for multiple therapeutic applications in viral, bacterial, fungal diseases and physiological disorders. In traditional Indian medicine system, <it>Cassia tora </it>(<it>Senna tora</it>) is reportedly effective in treatment of skin and gastrointestinal disorders. The present study explores the protease inhibitory activity of the above plant seeds against trypsin, <it>Aspergillus flavus </it>and <it>Bacillus </it>sp. proteases.</p> <p>Methods</p> <p>The crushed seeds of <it>Cassia tora </it>were washed thoroughly with acetone and hexane for depigmentation and defatting. The proteins were fractionated by ammonium sulphate (0-30, 30-60, 60-90%) followed by dialysis and size exclusion chromatography (SEC). The inhibitory potential of crude seed extract and most active dialyzed fraction against trypsin and proteases was established by spot test using unprocessed x-ray film and casein digestion methods, respectively. Electrophoretic analysis of most active fraction (30-60%) and SEC elutes were carried employing Sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) and Gelatin SDS-PAGE. Inhibition of fungal spore germination was studied in the presence of dialyzed active inhibitor fraction. Standard deviation (SD) and ANOVA were employed as statistical tools.</p> <p>Results</p> <p>The crude seeds' extract displayed strong antitryptic, bacterial and fungal protease inhibitory activity on x-ray film. The seed protein fraction 30-60% was found most active for trypsin inhibition in caseinolytic assay (P < 0.001). The inhibition of caseinolytic activity of the proteases increased with increasing ratio of seed extract. The residual activity of trypsin, <it>Aspergillus flavus </it>and <it>Bacillus </it>sp. proteases remained only 4, 7 and 3.1%, respectively when proteases were incubated with 3 mg ml<sup>-1 </sup>seed protein extract for 60 min. The inhibitory activity was evident in gelatin SDS-PAGE where a major band (~17-19 kD) of protease inhibitor (PI) was detected in dialyzed and SEC elute. The conidial germination of <it>Aspergillus flavus </it>was moderately inhibited (30%) by the dialyzed seed extract.</p> <p>Conclusions</p> <p><it>Cassia tora </it>seed extract has strong protease inhibitory activity against trypsin, <it>Aspergillus flavus </it>and <it>Bacillus </it>sp. proteases. The inhibitor in <it>Cassia tora </it>may attenuate microbial proteases and also might be used as phytoprotecting agent.</p

    Indicators of psychoses or psychoses as indicators: the relationship between Indigenous social disadvantage and serious mental illness

    Objective: To explore the relationship between Indigenous social disadvantage and serious mental illness.\ud \ud Conclusions: Rapidly changing patterns of mental disorders in Indigenous populations indicate the importance of social determinants. Canadian research on Native American suicide has demonstrated a clear link between social control factors and one mental health issue – completed suicide – a finding with major social policy implications. This work has not been replicable in Australia, reflecting the particular political and social circumstances of Aboriginal and Torres Strait Islander populations. Recent research motivated by clinicians' observations of an increase in psychotic disorders in the Indigenous populations of Cape York and the Torres Strait has demonstrated that the prevalence is high and that there are within-population differences. Given similar exposure to social disadvantage, these findings raise the possibility of utilising Indigenous psychosis prevalence as a metric to inform a more nuanced understanding of the predictors of wider vulnerability and resilience at a setting level, and as a policy and service development lever

    Três ensaios biológicos foram realizados com o objetivo de determinar o conteúdo de energia metabolizável e do coeficientes de digestibilidade do extrato etéreo da soja integral processada por diferentes métodos (extrusão e tostagem) e do farelo de soja reconstituído com óleo. No primeiro e segundo ensaios, foram utilizadas a metodologia tradicional de coleta total de excretas, utilizando pintos no primeiro ensaio e galos no segundo. No terceiro ensaio, empregou-se a metodologia de alimentação forçada (Sibbald, 1976) com galos adultos. De acordo com os resultados obtidos, as sojas testadas apresentaram composições bromatológicas semelhantes. Os processamentos usados para a soja integral conferiram diferenças na digestibilidade dos nutrientes da soja e os coeficientes de digestibilidade do extrato etéreo obtidos para a soja extrusada e para o farelo de soja com adição de óleo foram superiores aos obtidos para soja tostada pelo vapor. Os valores de energia metabolizável obtidos para a soja extrusada foram superiores aos encontrados para a soja tostada pelo vapor e para o farelo de soja com adição de óleo, que foram semelhantes entre si.<br>Three-assay birds were conducted to evaluate the contents of metabolizable energy and ether extract digestibility of processed full-fat soybeans by extrusion, by toast and of the soybean meal plus oil. Two assays were carried out by using the traditional total excreta collection by chicks and by rooster, and the other assay was developed by using the force-feeding adult rooster method (Sibbald, 1976). The results showed that the feed composition of the soybean-tested were similar. The different processing caused differences on digestibility of the nutrients of the soybeans tested and the value of coefficients of the nutrients in extruded soybean and soybean meal plus oil were higher than that heated soybean.
