Monitoring of Hot Corrosion Behaviors for Alloys and Aluminide Coatings under Molten Sulfate Film with Thermal Cycles
In this study, a thin molten sulfate film was formed on the metallic samples and their corrosion processes have been monitored by electrochemical impedance technique under thermal cyclic condition. Inconel 600, nickel and aluminide diffusion coatings were used as samples. 50 mol%Na2SO4- 50 mol%Li2SO4 was used for preparing the molten sulfate film. Two kinds of specially designed electrochemical cells have been used in this study. One is three-electrode configuration and another is two-electrode configuration. Electrochemical impedance measurements were used for monitoring the degradation processes for all samples tested. It was found that both corrosion resistance and the change of molten sulfate film has been monitored successfully during 6 thermal cycles. In every thermal cycle, the corrosion resistance increased when the temperature shifted to lower values and decreased with the rising of temperature. Also, A drastic increase in corrosion resistance and the resistance of the molten salt film was observed when the temperature decreased below the melting point of the mixed molten sulfate film. Although both three-electrode and two-electrode configurations were usable for electrochemical measurements under molten sulfate film, it was concluded that the newly designed two-electrode configuration is more suitable under thermal cyclic condition
Contribution of collagen-binding protein Cnm of Streptococcus mutans to induced IgA nephropathy-like nephritis in rats
IgA nephropathy (IgAN), the most common primary glomerulonephritis, is considered an intractable disease with unknown pathogenic factors. In our previous study, Streptococcus mutans, the major causative bacteria of dental caries, which expresses Cnm, was related to the induction of IgAN-like nephritis. In the present study, the Cnm-positive S. mutans parental strain, a Cnm-defective isogenic mutant strain, its complementation strain, and recombinant Cnm (rCnm) protein were administered intravenously to Sprague Dawley rats, and the condition of their kidneys was evaluated focusing on the pathogenicity of Cnm. Rats treated with parental and complement bacterial strains and rCnm protein developed IgAN-like nephritis with mesangial proliferation and IgA and C3 mesangial deposition. Scanning immunoelectron microscopy revealed that rCnm was present in the electron-dense deposition area of the mesangial region in the rCnm protein group. These results demonstrated that the Cnm protein itself is an important factor in the induction of IgAN in rats

Very low-density lipoprotein-apoprotein CI is increased in diabetic nephropathy: Comparison with apoprotein CIII.BackgroundRecent studies have suggested that apoprotein (apo) CI in very low-density lipoprotein (VLDL) plays an important role in causing hypertriglyceridemia independent of apo CIII, which is associated with coronary heart disease (CHD). Because the incidence of CHD is increased in diabetic patients and is even higher when diabetic nephropathy is developed, we measured apo CI levels in VLDL from type 2 diabetic patients, with various degree of nephropathy, and compared the results with those for healthy controls or nondiabetic patients with chronic renal failure (CRF).MethodsThis study enrolled healthy control subjects, type 2 diabetic patients with normoalbuminuria, microalbuminuria, overt proteinuria, and CRF on hemodialysis and nondiabetic hemodialyis patients. VLDL (density <1.006) was separated by ultracentrifugation. Then the apo CI, CIII, and B concentrations in VLDL were measured by enzyme-linked immunosorbent assay (ELISA).ResultsThe apo CI, CIII, and B concentrations in VLDL were respectively 3-, 2-, and 2-fold higher, respectively, in diabetic patients with overt proteinuria than in controls. Hemodialysis patients with diabetic nephropathy had levels of apo CI, CIII, and B in VLDL that were 2.6-, 2.7- and 2-fold higher, respectively, than those in controls. Nondiabetic hemodialysis patients also had a 2.7-fold higher level of VLDL apo CIII, whereas VLDL apo CI and VLDL apo B were not significantly increased. VLDL apo CI was significantly correlated with VLDL apo B independently of VLDL apo CIII level.ConclusionAn increase of VLDL apo CIII is a prominent feature of dyslipidemia in CRF patients, regardless of whether they are diabetic or nondiabetic, whereas an increase of VLDL apo CI is more specific to diabetic nephropathy and is closely associated with an increase of VLDL particle numbers, a new risk factor for CHD
Cnm of Streptococcus mutans is important for cell surface structure and membrane permeability
Streptococcus mutans, a Gram-positive facultative anaerobic bacterium, is a major pathogen of dental caries. The protein Cnm of S. mutans is involved in collagen binding, but its other biological functions are unknown. In this study, a Cnm-deficient isogenic mutant and a complementation strain were generated from a Cnm-positive S. mutans strain to help determine the properties of Cnm. Initially, comparison of the cell surface structure was performed by electron microscopy, which demonstrated that Cnm appears to be localized on the cell surface and associated with a protruding cell surface structure. Deep RNA sequencing of the strains revealed that the defect in Cnm caused upregulated expression of many genes related to ABC transporters and cell-surface proteins, while a few genes were downregulated. The amount of biofilm formed by the Cnm-defective strain increased compared with the parental and complemented strains, but the biofilm structure was thinner because of elevated expression of genes encoding glucan synthesis enzymes, leading to increased production of extracellular polysaccharides. Particular antibiotics, including bacitracin and chloramphenicol, had a lower minimum inhibitory concentration for the Cnm-defective strain than particular antibiotics, including bacitracin and chloramphenicol, compared with the parental and complemented strains. Our results suggest that S. mutans Cnm is located on the cell surface, gives rise to the observed protruding cell surface, and is associated with several biological properties related to membrane permeability
The Amelioration of Renal Damage in Skp2-Deficient Mice Canceled by p27 Kip1 Deficiency in Skp2β/β p27β/β Mice
SCF-Skp2 E3 ubiquitin ligase (Skp2 hereafter) targets several cell cycle regulatory proteins for degradation via the ubiquitin-dependent pathway. However, the target-specific physiological functions of Skp2 have not been fully elucidated in kidney diseases. We previously reported an increase in Skp2 in progressive nephropathy and amelioration of unilateral ureteral obstruction (UUO) renal injury associated with renal accumulation of p27 in Skp2β/β mice. However, it remains unclear whether the amelioration of renal injury in Skp2β/β mice is solely caused by p27 accumulation, since Skp2 targets several other proteins. Using Skp2β/βp27β/β mice, we investigated whether Skp2 specifically targets p27 in the progressive nephropathy mediated by UUO. In contrast to the marked suppression of UUO renal injury in Skp2β/β mice, progression of tubular dilatation associated with tubular epithelial cell proliferation and tubulointerstitial fibrosis with increased expression of collagen and Ξ±-smooth muscle actin were observed in the obstructed kidneys in Skp2β/βp27β/β mice. No significant increases in other Skp2 target proteins including p57, p130, TOB1, cyclin A and cyclin D1 were noted in the UUO kidney in Skp2β/β mice, while p21, c-Myc, b-Myb and cyclin E were slightly increased. Contrary to the ameliorated UUO renal injure by Skp2-deficiency, the amelioration was canceled by the additional p27-deficiency in Skp2β/βp27β/β mice. These findings suggest a pathogenic role of the reduction in p27 targeted by Skp2 in the progression of nephropathy in UUO mice
Low-Oxygen Responses of Cut Carnation Flowers Associated with Modified Atmosphere Packaging
Gaseous factors affect post-harvest physiological processes in horticultural crops, including ornamental flowers. However, the molecular responses of cut flowers to the low-oxygen conditions associated with modified atmosphere packaging (MAP) have not yet been elucidated. Here, we show that storage of cut carnation flowers in a sealed polypropylene bag decreased the oxygen concentration in the bag to 3-5% and slowed flower opening. The vase life of carnation flowers after storage for seven days under MAP conditions was comparable to that without storage and was improved by the application of a commercial-quality preservative. The adenylate energy charge (AEC) was maintained at high levels in petals from florets stored under MAP conditions. This was accompanied by the upregulation of four hypoxia-related genes, among which the HYPOXIA-RESPONSIVE ETHYLENE RESPONSE FACTOR and PHYTOGLOBIN genes (DcERF19 and DcPGB1) were newly identified. These results suggest that hypoxia-responsive genes contribute to the maintenance of the energy status in carnation flowers stored under MAP conditions, making this gas-controlling technique potentially effective for maintaining cut flower quality without cooling
Possible link between dental diseases and arteriosclerosis in patients on hemodialysis.
BACKGROUND:Patients on hemodialysis must undergo this procedure at a hospital three times weekly and might be unable to visit a dentist. In addition, dentists might hesitate to provide oral care because such patients tend to bleed because they are medicated with anticoagulants, are susceptible to bacterial infections, and might have unusual drug reactions. We postulated that patients on hemodialysis have worse oral status than healthy people, which in turn might predispose such patients to systemic complications. METHODS:We compared the status of dental caries and periodontal diseases among 80 patients on hemodialysis and 76 healthy individuals (controls) using the decayed, missing, or filled teeth (DMFT) index, total number of C4 teeth (destruction of the entire tooth crown), and periodontal pocket depth. Clinical data were analyzed after all patients on hemodialysis and controls provided written, informed consent to participate in the study. RESULTS:Total number of C4 teeth (p = 0.021), missing teeth (MT) index (p = 0.0302), and DMFT index score β₯ 24 (p = 0.017) were significantly higher in patients on hemodialysis than controls. Pulse pressure (p = 0.0042) and the prevalence of a history of heart disease such as angina pectoris and acute myocardial infarction (p = 0.029) were higher in patients on hemodialysis with higher (β₯ 24) than lower (< 24) DMFT index scores. Periodontal pocket depth was not significantly different between these two groups. CONCLUSION:Worse status of dental caries is possibly associated with arteriosclerosis among patients on hemodialysis
Estimation of Nearshore Wind Conditions Using Onshore Observation Data with Computational Fluid Dynamic and Mesoscale Models
This study aimed to establish numerical models to replicate wind conditions for nearshore waters, sensitive to onshore topography, and to compare the characteristics of computational fluid dynamic (CFD) and mesoscale models. Vertical Doppler light detection and ranging (LiDAR) observation data were measured at an onshore site, which showed that wind conditions were affected by thermodynamic phenomena, such as land and sea breeze, and dynamical effects from neighboring onshore topography. The estimation accuracy of the CFD model depended on the height of the LiDAR data input. A height close to the target, such as the hub height of wind turbines, seemed appropriate as input data, considering that the accuracy of the wind speed shear replicated in a CFD numerical model may be uncertain. The mesoscale model replicated the wind through the thermodynamic effect and reliably estimated wind speed over nearshore waters without observation correction. Larger estimation errors were detected in the CFD model than in the mesoscale model, as the former could not account for thermodynamic effects. Wind conditions in water areas near complex coastlines may also be formed by thermodynamic factors, making analysis using a mesoscale model advantageous
