Genome-wide association study of hepatocellular carcinoma in Southern Chinese patients with chronic hepatitis B virus infection
One of the most relevant risk factors for hepatocellular carcinoma (HCC) development is chronic hepatitis B virus (HBV) infection, but only a fraction of chronic HBV carriers develop HCC, indicating that complex interactions among viral, environmental and genetic factors lead to HCC in HBV-infected patients. So far, host genetic factors have incompletely been characterized. Therefore, we performed a genome-wide association (GWA) study in a Southern Chinese cohort consisting of 95 HBV-infected HCC patients (cases) and 97 HBV-infected patients without HCC (controls) using the Illumina Human610-Quad BeadChips. The top single nucleotide polymorphisms (SNPs) were then validated in an independent cohort of 500 cases and 728 controls. 4 SNPs (rs12682266, rs7821974, rs2275959, rs1573266) at chromosome 8p12 showed consistent association in both the GWA and replication phases (OR combined = 1.31-1.39; p combined = 2.71×10 -5-5.19×10 -4; PAR combined = 26-31%). We found a 2.3-kb expressed sequence tag (EST) in the region using in-silico data mining and verified the existence of the full-length EST experimentally. The expression level of the EST was significantly reduced in human HCC tumors in comparison to the corresponding non-tumorous liver tissues (P<0.001). Results from sequence analysis and in-vitro protein translation study suggest that the transcript might function as a long non-coding RNA. In summary, our study suggests that variations at chromosome 8p12 may promote HCC in patients with HBV. Further functional studies of this region may help understand HBV-associated hepatocarcinogenesis. © 2011 Chan et al.published_or_final_versio

No triangles on the moduli space of maximally supersymmetric gauge theory
Maximally supersymmetric gauge theory in four dimensions has a remarkably
simple S-matrix at the origin of its moduli space at both tree and loop level.
This leads to the question what, if any, of this structure survives at the
complement of this one point. Here this question is studied in detail at one
loop for the branch of the moduli space parameterized by a vacuum expectation
value for one complex scalar. Motivated by the parallel D-brane picture of
spontaneous symmetry breaking a simple relation is demonstrated between the
Lagrangian of broken super Yang-Mills theory and that of its higher dimensional
unbroken cousin. Using this relation it is proven both through an on- as well
as an off-shell method there are no so-called triangle coefficients in the
natural basis of one-loop functions at any finite point of the moduli space for
the theory under study. The off-shell method yields in addition absence of
rational terms in a class of theories on the Coulomb branch which includes the
special case of maximal supersymmetry. The results in this article provide
direct field theory evidence for a recently proposed exact dual conformal
symmetry motivated by the AdS/CFT correspondence.Comment: 39 pages, 4 figure

The SKA and "High-Resolution" Science
"High-resolution", or "long-baseline", science with the SKA and its
precursors covers a broad range of topics in astrophysics. In several research
areas, the coupling between improved brightness sensitivity of the SKA and a
sub-arcsecond resolution would uncover truly unique avenues and opportunities
for studying extreme states of matter, vicinity of compact relativistic
objects, and complex processes in astrophysical plasmas. At the same time, long
baselines would secure excellent positional and astrometric measurements with
the SKA and critically enhance SKA image fidelity at all scales. The latter
aspect may also have a substantial impact on the survey speed of the SKA, thus

Distributed Synthesis in Continuous Time
We introduce a formalism modelling communication of distributed agents
strictly in continuous-time. Within this framework, we study the problem of
synthesising local strategies for individual agents such that a specified set
of goal states is reached, or reached with at least a given probability. The
flow of time is modelled explicitly based on continuous-time randomness, with
two natural implications: First, the non-determinism stemming from interleaving
disappears. Second, when we restrict to a subclass of non-urgent models, the
quantitative value problem for two players can be solved in EXPTIME. Indeed,
the explicit continuous time enables players to communicate their states by
delaying synchronisation (which is unrestricted for non-urgent models). In
general, the problems are undecidable already for two players in the
quantitative case and three players in the qualitative case. The qualitative
undecidability is shown by a reduction to decentralized POMDPs for which we
provide the strongest (and rather surprising) undecidability result so far
Prevalence of occult hepatitis B infection in a highly endemic area for chronic hepatitis B: A study of a large blood donor population
Background and aims: The aim of the present study was to determine the population prevalence of occult hepatitis B (OHB) infection and its clinical profile in a highly endemic area of chronic hepatitis B virus disease. Methods: OHB was first identified by individual sample testing for hepatitis B surface antigen (HBsAg) followed by nucleic acid testing (NAT) and vice versa for 3044 (cohort 1, stored sera from donation within 1 year) and 9990 (cohort 2, prospective study) blood donors, respectively. OHB was confirmed meticulously by ≥2 out of 3 tests with detectable hepatitis B virus (HBV) DNA using a sensitive standardised assay. Detailed serology and viral load in the serum and liver were studied. Results: The prevalence of OHB was 0.13% (4/3044) and 0.11% (11/9967) for cohort 1 and 2, respectively. In cohort 2, 10 out of 11 OHB samples were positive for anti-HBc (hepatitis B core antigen) antibody (all were immunoglobulin G). Seven had detectable anti-HBs. The serum HBV DNA levels were extremely low (highest 14.1 IU/ml). Of the six donors who underwent liver biopsies, all had normal liver biochemistry, extremely low liver HBV DNA (highest 6.21 copies/cell) and nearly normal liver histology. For those with viral sequence generation, none had the common HBsAg mutant G145R. Conclusions: The prevalence of OHB in a highly endemic area of chronic HBV was very low, thus implying a low impact on transfusion services. To implement universal screening, the high cost of NAT should be taken into account. OHB blood donors had very low HBV replication, and normal liver biochemistry and histology, conferring a favourable prognosis.published_or_final_versio
Pneumococcal carriage in sub-Saharan Africa--a systematic review.
BACKGROUND: Pneumococcal epidemiology varies geographically and few data are available from the African continent. We assess pneumococcal carriage from studies conducted in sub-Saharan Africa (sSA) before and after the pneumococcal conjugate vaccine (PCV) era. METHODS: A search for pneumococcal carriage studies published before 2012 was conducted to describe carriage in sSA. The review also describes pneumococcal serotypes and assesses the impact of vaccination on carriage in this region. RESULTS: Fifty-seven studies were included in this review with the majority (40.3%) from South Africa. There was considerable variability in the prevalence of carriage between studies (I-squared statistic = 99%). Carriage was higher in children and decreased with increasing age, 63.2% (95% CI: 55.6-70.8) in children less than 5 years, 42.6% (95% CI: 29.9-55.4) in children 5-15 years and 28.0% (95% CI: 19.0-37.0) in adults older than 15 years. There was no difference in the prevalence of carriage between males and females in 9/11 studies. Serotypes 19F, 6B, 6A, 14 and 23F were the five most common isolates. A meta-analysis of four randomized trials of PCV vaccination in children aged 9-24 months showed that carriage of vaccine type (VT) serotypes decreased with PCV vaccination; however, overall carriage remained the same because of a concomitant increase in non-vaccine type (NVT) serotypes. CONCLUSION: Pneumococcal carriage is generally high in the African continent, particularly in young children. The five most common serotypes in sSA are among the top seven serotypes that cause invasive pneumococcal disease in children globally. These serotypes are covered by the two PCVs recommended for routine childhood immunization by the WHO. The distribution of serotypes found in the nasopharynx is altered by PCV vaccination
