    An update on the status of wet forest stream-dwelling frogs of the Eungella region

    Eungella’s wet forests are home to a number of stream-breeding frogs including three species endemic to the Eungella region: the Eungella dayfrog (Taudactylus eungellensis), Eungella tinkerfrog (T. liemi), and northern gastric brooding frog (Rheobatrachus vitellinus). During the mid-1980s, T. eungellensis and R. vitellinus suffered dramatic population declines attributable to amphibian chytridiomycosis, a disease caused by the amphibian chytrid fungus (Batrachochytrium dendrobatidis or Bd). While surveys in the late 1980s failed to locate T. eungellensis or R. vitellinus, populations of the former were located on a handful of streams surveyed by researchers in the mid-to-late 1990s. Between January 2000 and November 2015, additional surveys targeting these and other wet forest frog species were conducted at 114 sites within Eungella National Park and adjoining areas of State Forest. During these surveys, we located T. eungellensis at many more sites than surveys in the 1990s. Abundances of T. eungellensis at these sites were typically low, however, and well below abundance levels prior to declines in the mid-1980s. As with surveys in the 1990s, T. eungellensis was scarce at high-elevation sites above 600 metres altitude. Numbers of this species do not appear to have increased significantly since the mid-1990s, suggesting recovery of T. eungellensis populations is occurring slowly, at best. In contrast with T. eungellensis, T. liemi was frequently recorded at high-elevation sites, albeit at low densities. As with previous surveys, surveys during 2000–2015 were unsuccessful in locating R. vitellinus. Further frog surveys and monitoring (including disease surveillance) are needed to better assess the status of stream frogs at Eungella, and to understand the influence of Bd on the abundance and distribution of threatened stream-dwelling frogs at Eungella

    Maternal obesity is associated with the formation of small dense LDL and hypoadiponectinemia in the third trimester

    Context: Maternal obesity is associated with high plasma triglyceride, poor vascular function, and an increased risk for pregnancy complications. In normal-weight pregnant women, higher triglyceride is associated with increased small, dense low-density lipoprotein (LDL). Hypothesis: In obese pregnancy, increased plasma triglyceride concentrations result in triglyceride enrichment of very low-density lipoprotein-1 particles and formation of small dense LDL via lipoprotein lipase. Design: Women (n = 55) of body mass index of 18–46 kg/m2 were sampled longitudinally at 12, 26, and 35 weeks' gestation and 4 months postnatally. Setting: Women were recruited at hospital antenatal appointments, and study visits were in a clinical research suite. Outcome Measures: Plasma concentrations of lipids, triglyceride-rich lipoproteins, lipoprotein lipase mass, estradiol, steroid hormone binding globulin, insulin, glucose, leptin, and adiponectin were determined. Results: Obese women commenced pregnancy with higher plasma triglyceride, reached the same maximum, and then returned to higher postnatal levels than normal-weight women. Estradiol response to pregnancy (trimester 1–3 incremental area under the curve) was positively associated with plasma triglyceride response (r2 adjusted 25%, P < .001). In the third trimester, the proportion of small, dense LDL was 2-fold higher in obese women than normal-weight women [mean (SD) 40.7 (18.8) vs 21.9 (10.9)%, P = .014], and 35% of obese, 14% of overweight, and none of the normal-weight women displayed an atherogenic LDL subfraction phenotype. The small, dense LDL mass response to pregnancy was inversely associated with adiponectin response (17%, P = .013). Conclusions: Maternal obesity is associated with an atherogenic LDL subfraction phenotype and may provide a mechanistic link to poor vascular function and adverse pregnancy outcome

    Obesity and excessive lipolysis are implicated in preeclampsia (PE). Intrauterine growth restriction is associated with low maternal body mass index and decreased lipolysis. Our aim was to assess how maternal and offspring fatty acid metabolism is altered in mothers in the third trimester of pregnancy with PE (n=62) or intrauterine growth restriction (n=23) compared with healthy pregnancies (n=164). Markers of lipid metabolism and erythrocyte fatty acid concentrations were measured. Maternal adipose tissue fatty acid composition and mRNA expression of adipose tissue fatty acid–metabolizing enzymes and placental fatty acid transporters were compared. Mothers with PE had higher plasma triglyceride (21%, P<0.001) and nonesterified fatty acid (50%, P<0.001) concentrations than controls. Concentrations of major n−6 and n−3 long-chain polyunsaturated fatty acids in erythrocytes were 23% to 60% lower (all P<0.005) in PE and intrauterine growth restriction mothers and offspring compared with controls. Subcutaneous adipose tissue Δ−5 and Δ−6 desaturase and very long-chain fatty acid elongase mRNA expression was lower in PE than controls (respectively, mean [SD] control 3.38 [2.96] versus PE 1.83 [1.91], P=0.030; 3.33 [2.25] versus 1.03 [0.96], P<0.001; 0.40 [0.81] versus 0.00 [0.00], P=0.038 expression relative to control gene [square root]). Low maternal and fetal long-chain polyunsaturated fatty acid concentrations in PE may be the result of decreased maternal synthesis

    New thiosemicarbazone (TSC) ligands, AMT-C[dbnd]N-TSCH (L1), AMT-C[dbnd]N-TSC(CH3) (L2), CIN-C[dbnd]N-TSCH (L3) and CIN-C[dbnd]N-TSC(CH3) (L4) (AMT = 2-acetyl-5-methylthiophene, TSCH = thiosemicarbazide, TSC(CH3) = 4-methyl-3-thiosemicarbazide, CIN = cinnamaldehyde) were synthesized by condensation reaction. The reaction of [PdCODCl2] with the ligands L1, L2 and L4in the ratio 1:1 yielded complexes [Pd(L1)Cl] (C1), [Pd(L2)Cl2] (C2), and [Pd(L4)2Cl2] (C4) respectively (COD = 1,5-cyclooctadiene). The reaction of [Pd(COD)Cl2] with CIN-C[dbnd]N-TSCH (L3) in 1:2 metal to ligand ratio yielded the complex [Pd(L3)2] (C3). The ligands and the complexes were characterized by UV–Vis, FT-IR, NMR, elemental analysis and conductivity measurements. The ligand L1 coordinated to the metal in a tridentate fashion while L2, L3 and L4 coordinated in a bidentate fashion. The ligands L1 and L3 coordinated to the metal as thiol anions while ligand L4 coordinated via the thione sulphur. Conductivity measurements of the metal complexes in dimethyl sulfoxide showed that the chloride ions complexes C1, C2 and C4 are within the coordination spheres

    Association of physical activity with adiposity in preschoolers using different clinical adiposity measures: a cross-sectional study.

    More research is needed about the association between physical activity (PA), sedentary behaviour (SB), and adiposity in preschoolers, particularly using more direct clinical measures of adiposity. Therefore, the main objective of this study was to investigate the association between objectively measured PA and different clinical adiposity measures in a large sample of preschoolers. Four hundred sixty-three predominantly normal-weight (77%) 2-6-year-old preschool children participated in the Swiss Preschoolers' Health Study (SPLASHY). Physical activity was measured using accelerometers and was analyzed using 15-s (uni-axial) epoch length using validated cut-offs. Adiposity measures included body mass index (BMI), the sum of four skinfolds, and waist circumference (WC). Multilevel linear regression modeling, adjusted for age, sex and wear time, was used to assess the association between PA and SB with BMI, WC, and skinfold thickness. Total PA and different PA intensities were positively and SB was inversely associated with BMI in the total sample and in the normal-weight children (p < 0.05). Total PA was inversely associated with skinfold thickness in overweight and obese children (p < 0.05), while there was only a weak association for vigorous PA (p > 0.05). Moderate and moderate-to-vigorous PA were positively, and SB was negatively associated with WC in the total sample and in the normal-weight children (p < 0.05). Additional adjustment for potential sociocultural and biological confounding variables attenuated some of the results. In this very young and predominantly normal-weight population, PA is positively related to BMI and WC, but this relationship is not observed in overweight and obese children. In this latter population, PA is inversely, and SB is positively related to skinfold thickness. Skinfold thickness could represent a useful and simple clinical measure of body fat in preschoolers. The role of vigorous PA in the prevention of early childhood obesity should be further investigated in future studies. ISRCTN ISRCTN41045021 . Retrospectively registered 06 May 2014

    Cross-cultural adaptation of an innovative approach to learning about difficult conversations in healthcare

    Background:\u2003The Program to Enhance Relational and Communication Skills (PERCS) was developed at a large hospital in the United States to enhance clinicians\u2019 preparedness to engage in difficult conversations. Aim:\u2003To describe the implementation of PERCS in an Italian hospital and assess the program's efficacy. Methods:\u2003The Italian PERCS program featured 4-h experiential workshops enrolling 10\u201315 interdisciplinary participants. The workshops were organized around the enactment and debriefing of realistic case scenarios portrayed by actors and volunteer clinicians. Before and after the workshop, participants rated their perceived preparation, communication and relational skills, confidence, and anxiety on 5-point Likert scales. Open-ended questions explored their reflections on the learning. T-tests and content analysis were used to analyze the quantitative and qualitative data, respectively. Results:\u2003146 clinicians attended 13 workshops. Participants reported better preparation, confidence, and communication skills (p\u2009<\u20090.001) after the workshops. The program had a different impact depending on the discipline. Participants valued the emphasis on group feedback, experiential and interdisciplinary learning, and the patient's perspective, and acquired: new communication skills, self-reflective attitude, reframed perspective, and interdisciplinary teamwork. Conclusion:\u2003PERCS proved culturally adaptable to the Italian context and effective in improving participants\u2019 sense of preparation, communication skills, and confidence
