    Major role of organic anion transporter 3 in the transport of indoxyl sulfate in the kidney.BackgroundIndoxyl sulfate is a uremic toxin that accumulates in the body because of the patient's inability to excrete it and it induces a number of uremic symptoms and leads to chronic renal failure. The functional failure of the excretion system for indoxyl sulfate causes its accumulation in blood. The purpose of the present study was to characterize the transport mechanism responsible for the renal excretion of indoxyl sulfate.MethodsThe [3H]indoxyl sulfate transport mechanism was investigated using an in vivo tissue-sampling single-injection technique, the kidney uptake index (KUI) method. Rat organic anion transporter 3 (rOAT3)-expressing Xenopus laevis oocyte system was used for measuring [3H]indoxyl sulfate uptake activity.ResultsProbenecid showed a concentration-dependent inhibitory effect on the uptake of [3H]indoxyl sulfate using the KUI method, and uptake was inhibited by organic anions such as para-aminohippuric acid (PAH) and benzylpenicillin, by weak base such as cimetidine, and by uremic toxins, such as 3-carboxy-4-methyl-5-propyl-2-furanpropanoic acid (CMPF) and hippuric acid (HA). However, salicylic acid, indomethacin, 3,5,3′-triiodo-L-thyronine and indole acetic acid (IA) had no effect on the uptake. rOAT3-expressing oocytes exhibited uptake of [3H]indoxyl sulfate by rOAT3 (Km = 158 μmol/L). Moreover, a number of uremic toxins inhibited the uptake of [3H]indoxyl sulfate by rOAT3.ConclusionsThese results suggest that rOAT3 is responsible for the renal uptake of indoxyl sulfate, and uremic toxins share the transport mechanism for indoxyl sulfate. Mutual inhibition of these uremic toxins via OAT3 may accelerate their accumulation in the body and, thereby, the progression of nephrotoxicity in uremia

    Celiomesenteric anomaly with concurrent aneurysm

    AbstractWe describe a rare case of a celiomesenteric anomaly with concurrent aneurysm. The patient, a 53-year-old man, had no abdominal pain or discomfort. The presence of a celiac artery aneurysm was suspected on the basis of the results of abdominal computerized tomographic scanning and echo ultrasound scanning performed because of proteinuria. Intra-arterial digital subtraction angiographic results showed the anomaly and aneurysm. Because of the risk of rupture of the aneurysm, the lesion was repaired surgically, with the placement of an interpositional prosthetic graft. We found no previous reports of celiomesenteric anomaly with concurrent aneurysm repaired with prosthetic graft. (J Vasc Surg 1999;29:711-4.

    Nongyrotropic electron velocity distribution functions near the lunar surface

    We have analyzed nongyrotropic electron velocity distribution functions (VDFs) obtained near the lunar surface. Electron VDFs, measured at ∼10–100 km altitude by Kaguya in both the solar wind and the Earth's magnetosphere, exhibit nongyrotropic empty regions associated with the ‘gyroloss’ effect; i.e., electron absorption by the lunar surface combined with electron gyromotion. Particle-trace calculations allow us to derive theoretical forbidden regions in the electron VDFs, thereby taking into account the modifications due to nonuniform magnetic fields caused by diamagnetic-current systems, lunar-surface charging, and electric fields perpendicular to the magnetic field. Comparison between the observed empty regions with the theoretically derived forbidden regions suggests that various components modify the characteristics of the nongyrotropic electron VDFs depending on the ambient-plasma conditions. On the lunar nightside in the magnetotail lobes, negative surface potentials slightly reduce the size of the forbidden regions, but there are no distinct effects of either the diamagnetic current or perpendicular electric fields. On the dayside in the solar wind, the observations suggest the presence of either the diamagnetic-current or solar wind convection electric field effects, or both. In the terrestrial plasma sheet, all three mechanisms can substantially modify the characteristics of the forbidden regions. The observations imply the presence of a local electric field of at least 5 mV/m although the mechanism responsible for production of such a strong electric field is unknown. Analysis of nongyrotropic VDFs associated with the gyroloss effect near solid surfaces can promote a better understanding of the near-surface plasma environment and of plasma–solid-surface interactions

    Digging up bulk band dispersion buried under a passivation layer

    Atomically controlled crystal growth of thin films has established foundations of nanotechnology aimed at the development of advanced functional devices. Crystallization under non-equilibrium conditions allows engineering of new materials with their atomically-flat interfaces in the heterostructures exhibiting novel physical properties. From a fundamental point of view, knowledge of the electronic structures of thin films and their interfaces is indispensable to understand the origins of their functionality which further evolves into realistic device application. In view of extreme surface sensitivity of the conventional vacuum-ultraviolet (VUV) angle-resolved photoemission spectroscopy (ARPES), with a probing depth of several angstroms, experiments on thin films have to use sophisticated in-situ sample transfer systems to avoid surface contamination. In this Letter, we put forward a method to circumvent these difficulties using soft X-ray (SX) ARPES. A GaAs:Be thin film in our samples was protected by an amorphous As layer with an thickness of 1\sim 1 nm exceeding the probing depth of the VUV photoemission with photon energy hνh\nu around 100 eV. The increase of the probing depth with increasing hνh\nu towards the SX region has clearly exposed the bulk band dispersion without any surface treatment. Any contributions from potential interface states between the thin film and the amorphous capping layer has been below the detection limit. Our results demonstrate that SX-ARPES enables the observation of coherent three-dimensional band dispersion of buried heterostructure layers through an amorphous capping layer, breaking through the necessity of surface cleaning of thin film samples. Thereby, this opens new frontiers in diagnostics of authentic momentum-resolved electronic structure of protected thin-film heterostructures.Comment: 5 pages, 3 figure

    Aseptic Meningoencephalitis Complicated by Retrobulbar Neuritis

    A 25-year-old man was admitted to our hospital for testing and follow-up of aseptic meningoencephalitis. After admission to our hospital, the patient suddenly complained of visual field disorder and a decrease of visual acuity in the right eye. We diagnosed aseptic meningitis complicated by retrobulbar neuritis using MRI. We immediately initiated weekly steroid pulse therapy, and eventually, marked improvement in visual acuity and the visual field disorder was observed without any late effects

    Establishment of Rat Embryonic Stem Cells and Making of Chimera Rats

    The rat is a reference animal model for physiological studies and for the analysis of multigenic human diseases such as hypertension, diabetes, neurological disorders, and cancer. The rats have long been used in extensive chemical carcinogenesis studies. Thus, the rat embryonic stem (rES) cell is an important resource for the study of disease models. Attempts to derive ES cells from various mammals, including the rat, have not succeeded. Here we have established two independent rES cells from Wister rat blastocysts that have undifferentiated characters such as Nanog and Oct3/4 genes expression and they have stage-specific embryonic antigen (SSEA) -1, -3, -4, and TRA-1-81 expression. The cells were successfully cultured in an undifferentiated state and can be possible over 18 passages with maintaining more than 40% of normal karyotype. Their pluripotent potential was confirmed by the differentiation into derivatives of the endoderm, mesoderm, and ectoderm. Most importantly, the rES cells are capable of producing chimera rats. Therefore, we established pluripotent rES cell lines that are widely used to produce genetically modified experimental rats for study of human diseases

    Impact of Chronic Kidney Disease on the Presence and Severity of Aortic Stenosis in Patients at High Risk for Coronary Artery Disease

    <p>Abstract</p> <p>Objective</p> <p>We evaluated the impact of chronic kidney disease (CKD) on the presence and severity of aortic stenosis (AS) in patients at high risk for coronary artery disease (CAD).</p> <p>Methods</p> <p>One hundred and twenty consecutive patients who underwent invasive coronary angiography were enrolled. Aortic valve area (AVA) was calculated by the continuity equation using transthoracic echocardiography, and was normalized by body surface area (AVA index).</p> <p>Results</p> <p>Among all 120 patients, 78% had CAD, 55% had CKD (stage 3: 81%; stage 4: 19%), and 34% had AS (AVA < 2.0cm<sup>2</sup>). Patients with AS were older, more often female, and had a higher frequency of CKD than those without AS, but the prevalence of CAD and most other coexisting conventional risk factors was similar between patients with and without AS. Multivariate linear regression analysis indicated that only CKD and CAD were independent determinants of AVA index with standardized coefficients of -0.37 and -0.28, respectively. When patients were divided into 3 groups (group 1: absence of CKD and CAD, n = 16; group 2: presence of either CKD or CAD, n = 51; and group 3: presence of both CKD and CAD, n = 53), group 3 had the smallest AVA index (1.19 ± 0.30*# cm<sup>2</sup>/m<sup>2</sup>, *p < 0.05 vs. group 1: 1.65 ± 0.32 cm<sup>2</sup>/m<sup>2</sup>, and #p < 0.05 vs. group 2: 1.43 ± 0.29* cm<sup>2</sup>/m<sup>2</sup>) and the highest peak velocity across the aortic valve (1.53 ± 0.41*# m/sec; *p < 0.05 vs. group 1: 1.28 ± 0.29 m/sec, and #p < 0.05 vs. group 2: 1.35 ± 0.27 m/sec).</p> <p>Conclusion</p> <p>CKD, even pre-stage 5 CKD, has a more powerful impact on the presence and severity of AS than other conventional risk factors for atherosclerosis in patients at high risk for CAD.</p
