    Epithelial lining of the endometrium during the luteal phase in patients under controlled ovarian hyperstimulation

    The endometrium is receptive for the embryo and presents an implantation window for a limited time. This study is aimed at highlighting an alternation in pinopod expression and to provide more analysisof the structural characteristics of epithelial lining of the endometrium during luteal phase in patients undergoing controlled ovarian hyperstimmulation (COH). Twelve oocyte donors were used. Theyunderwent two endometrial biopsies 2-7 days after human chronic gonadotrophin (HCG) administration. Endometrial epithelial surface appearance was evaluated by scanning electron microscopy. Microvillibecame more uniformly distributed as the luteal phase progressed. Also the central aspects of the cells appeared to protrude more into the endometrial lumen as the cycle progressed. Apical protrusions(pinopods) were noted to develop and regress during the midluteal phase after HCG administration. Pinopods began development in the region of the glandular orifices to become much denser at the glandular orifices than in regions further away from the glands. In conclusion, epithelial lining ofendometrium in stimulated cycle during the luteal phase progresses in an orderly manner. This advanced development may result in an alteration of the window of implantation between the developing endometrium and the developing blastocyst and affect pregnancy rates in women undergoing controlled ovarian hyperstimmulation

    Taxonomic Studies on Three Marine Ciliates from China, Including a New Species (Ciliophora, Cyrtophorida)

    The present work investigates the living morphology and infraciliature of three marine cyrtophorid ciliates, which were isolated from Qingdao, China. Compared with its congeners, Orthotrochilia sinica spec. nov. can be distinguished by a combination of features: body slender and elliptical in outline, size about 50–60 Γ— 20–25 ΞΌm in vivo, 18–21 somatic kineties, the length of the left perioral kinety treble the length of the right one, two ventrally located contractile vacuoles on the right side, and 25–32 nematodesmal rods. Based on current observations and the previous description, the diagnosis of Trochilioides tenuis (Deroux, 1976) Chen et al. 2011 is improved: cell size 30–40 Γ— 20–35 ΞΌm in vivo, oval shaped in outline; consistently three right kineties, four left kineties and seven postoral kineties; a single contractile vacuole; marine habitat. A second species of Trochilioides, T. recta (Kahl, 1923) Chen et al. 2011 is re-described based on a Chinese population. Furthermore, a key to the identification of species of the genus Trochilioides whose infraciliature data are available is supplied, and Chlamydonyx trivialis (Fenchel, 1965) comb. nov. [basionym: Trochilioides trivialis Fenchel, 1965] is suggested

    This paper investigates the morphology and morphogenesis during binary fission of a Chinese population ofΒ Euplotes amietiΒ Dragesco, 1970, a fresh water form which has previously not been well defined. This organism is morphologically very similar to the well-knownΒ Euplotes eurystomusΒ but differs from the latter both in the number of dorsal kineties and the molecular data. According to the information obtained, it is characterized by a combination of features including nine frontoventral cirri, ca. 60 membranelles, 12–15Β dorsal kineties, a macronucleus in the shape of the number 3, and a β€˜double-eurystomus’ type of silverline system. Its morphogenesis proceeds broadly in the same pattern as in its congeners.Β In this study, the SSU rRNA gene was sequenced for the first time, and phylogenetic analyses indicated that it is closely related to theΒ eurystomus-aediculatus-woodruffi-Β complex. Considering the extreme similarities in morphology betweenΒ E.Β amietiΒ andΒ E.eurystomus, we believe that the four sequences (four isolates) under the name ofΒ Euplotes eurystomusΒ (No. FR873716; FR873717; EF193250; AJ310491 deposited in GenBank) are very likely from misidentified material; that is, they represent different populations ofΒ Euplotes amieti

    Redescriptions of two Marine Scuticociliates from China, with notes on Stomatogenesis in Parauronema longum (Ciliophora, Scuticociliatida)

    The morphology and infraciliature of two marine scuticociliates, Pleuronema puytoraci Grolière and Detcheva, 1974, and Parauronema longum Song, 1995, collected from China, were investigated using live observation and protargol impregnation methods. Based on the data obtained for the China population, new information of the living morphology of Pleuronema puytoraci is documented and details of the complete infraciliature is available for the first time. The stomatogenesis of Parauronema longum is basically similar to that of its congeners and can be summarized as follows: membranelle 1, membranelle 2 and the scutica of the opisthe originate from the parental paroral membrane, whereas membranelle 3 of the opisthe develops from the parental scutica; the paroral membrane originates from the parental paroral membrane

    Tea and coffee consumption in relation to vitamin D and calcium levels in Saudi adolescents

    Background Coffee and tea consumption was hypothesized to interact with variants of vitamin D-receptor polymorphisms, but limited evidence exists. Here we determine for the first time whether increased coffee and tea consumption affects circulating levels of 25-hydroxyvitamin D in a cohort of Saudi adolescents. Methods A total of 330 randomly selected Saudi adolescents were included. Anthropometrics were recorded and fasting blood samples were analyzed for routine analysis of fasting glucose, lipid levels, calcium, albumin and phosphorous. Frequency of coffee and tea intake was noted. 25-hydroxyvitamin D levels were measured using enzyme-linked immunosorbent assays. Results Improved lipid profiles were observed in both boys and girls, as demonstrated by increased levels of HDL-cholesterol, even after controlling for age and BMI, among those consuming 9–12 cups of coffee/week. Vitamin D levels were significantly highest among those consuming 9–12 cups of tea/week in all subjects (p-value 0.009) independent of age, gender, BMI, physical activity and sun exposure. Conclusion This study suggests a link between tea consumption and vitamin D levels in a cohort of Saudi adolescents, independent of age, BMI, gender, physical activity and sun exposure. These findings should be confirmed prospectively

    Discovery of novel class of histone deacetylase inhibitors as potential anticancer agents

    Selective inhibition of histone deacetylases (HDACs) is an important strategy in the field of anticancer drug discovery. However, lack of inhibitors that possess high selectivity toward certain HDACs isozymes is associated with adverse side effects that limits their clinical applications. We have initiated a collaborative initiatives between multi-institutions aimed at the discovery of novel and selective HDACs inhibitors. To this end, a phenotypic screening of an in-house pilot library of about 70 small molecules against various HDAC isozymes led to the discovery of five compounds that displayed varying degrees of HDAC isozyme selectivity. The anticancer activities of these molecules were validated using various biological assays including transcriptomic studies. Compounds 15, 14, and 19 possessed selective inhibitory activity against HDAC5, while 28 displayed selective inhibition of HDAC1 and HDAC2. Compound 22 was found to be a selective inhibitor for HDAC3 and HDAC9. Importantly, we discovered a none-hydroxamate based HDAC inhibitor, compound 28, representing a distinct chemical probe of HDAC inhibitors. It contains a trifluoromethyloxadiazolyl moiety (TFMO) as a non-chelating metal-binding group. The new compounds showed potent anti-proliferative activity when tested against MCF7 breast cancer cell line, as well as increased acetylation of histones and induce cells apoptosis. The new compounds apoptotic effects were validated through the upregulation of proapoptotic proteins caspases3 and 7 and downregulation of the antiapoptotic biomarkers C-MYC, BCL2, BCL3 and NFΔΈB genes. Furthermore, the new compounds arrested cell cycle at different phases, which was confirmed through downregulation of the CDK1, 2, 4, 6, E2F1 and RB1 proteins. Taken together, our findings provide the foundation for the development of new chemical probes as potential lead drug candidates for the treatment of cancer

    Malignancy risk analysis in patients with inadequate fine needle aspiration cytology (FNAC) of the thyroid

    Background Thyroid fine needle aspiration cytology (FNAC) is the standard diagnostic modality for thyroid nodules. However, it has limitations among which is the incidence of non-diagnostic results (Thy1). Management of cases with repeatedly non-diagnostic FNAC ranges from simple observation to surgical intervention. We aim to evaluate the incidence of malignancy in non-diagnostic FNAC, and the success rate of repeated FNAC. We also aim to evaluate risk factors for malignancy in patients with non-diagnostic FNAC. Materials and Methods Retrospective analyses of consecutive cases with thyroid non diagnostic FNAC results were included. Results Out of total 1657 thyroid FNAC done during the study period, there were 264 (15.9%) non-diagnostic FNAC on the first attempt. On repeating those, the rate of a non-diagnostic result on second FNAC was 61.8% and on third FNAC was 47.2%. The overall malignancy rate in Thy1 FNAC was 4.5% (42% papillary, 42% follicular and 8% anaplastic), and the yield of malignancy decreased considerably with successive non-diagnostic FNAC. Ultrasound guidance by an experienced head neck radiologist produced the lowest non-diagnostic rate (38%) on repetition compared to US guidance by a generalist radiologist (65%) and by non US guidance (90%). Conclusions There is a low risk of malignancy in patients with a non-diagnostic FNAC result, commensurate to the risk of any nodule. The yield of malignancy decreased considerably with successive non-diagnostic FNAC

    Transcriptome of tumor-Infiltrating T cells in colorectal cancer patients uncovered a unique gene signature in CD4 + T cells associated with poor disease-specific survival

    Colorectal cancer (CRC) is influenced by infiltration of immune cell populations in the tumor microenvironment. While elevated levels of cytotoxic T cells are associated with improved prognosis, limited studies have reported associations between CD4+ T cells and disease outcomes. We recently performed transcriptomic profiling and comparative analyses of sorted CD4+ and CD8+ tumor-infiltrating lymphocytes (TILs) from bulk tumors of CRC patients with varying disease stages. In this study, we compared the transcriptomes of CD4+ with CD8+ TILs. Functional annotation pathway analyses revealed the downregulation of inflammatory response-related genes, while T cell activation and angiogenesis-related genes were upregulated in CD4+ TILs. The top 200 deregulated genes in CD4+ TILs were aligned with the cancer genome atlas (TCGA) CRC dataset to identify a unique gene signature associated with poor prognosis. Moreover, 69 upregulated and 20 downregulated genes showed similar trends of up/downregulation in the TCGA dataset and were used to calculate β€œpoor prognosis score” (ppScore), which was significantly associated with disease-specific survival. High ppScore patients showed lower expression of Treg-, Th1-, and Th17-related genes, and higher expression of Th2-related genes. Our data highlight the significance of T cells within the TME and identify a unique candidate prognostic gene signature for CD4+ TILs in CRC patients
