    Cartilage canals in newborn dogs : histochemical and immunohistochemical findings

    Cartilage canals (CCs) are microscopic structures involved in secondary ossification centers (SOCs) development. The features of CCs were investigated in the humeral and femoral proximal epiphyses of small-sized newborn dogs (from premature to 28 days after birth) with histochemical and immunohistochemical approaches. Masson's Trichrome revealed a ring-shaped area around CCs, which changes in colour from green (immature collagen) to red (mature collagen) as ossification progresses; perichondrium staining always matched the ring colour. Safranin-O was always negative. Immunohistochemical analysis revealed immunopositivity for both collagen type I and V around the CCs; collagen type II was negative. CCs count showed a tendency to be higher in the humerus than in the femur. This work enlightened for the first time changes in composition of CCs surrounding matrix during SOCs development in dogs, paving the way to further investigations

    Meniscus Matrix Remodeling in Response to Compressive Forces in Dogs

    Joint motion and postnatal stress of weight bearing are the principal factors that determine the phenotypical and architectural changes that characterize the maturation process of the meniscus. In this study, the effect of compressive forces on the meniscus will be evaluated in a litter of 12 Dobermann Pinschers, of approximately 2 months of age, euthanized as affected by the quadriceps contracture muscle syndrome of a single limb focusing on extracellular matrix remodeling and cell-extracellular matrix interaction (i.e., meniscal cells maturation, collagen fibers typology and arrangement). The affected limbs were considered as models of continuous compression while the physiologic loaded limbs were considered as controls. The results of this study suggest that a compressive continuous force, applied to the native meniscal cells, triggers an early maturation of the cellular phenotype, at the expense of the proper organization of collagen fibers. Nevertheless, an application of a compressive force could be useful in the engineering process of meniscal tissue in order to induce a faster achievement of the mature cellular phenotype and, consequently, the earlier production of the fundamental extracellular matrix (ECM), in order to improve cellular viability and adhesion of the cells within a hypothetical synthetic scaffold

    Cervical cystic lymphangioma in a young dog: CT findings

    Lymphangioma is a rare lymphatic disorder; in veterinary medicine it is still considered a benign tumour, while in human medicine has been recently classified as a Lymphatic Malformation (LM), in fact it probably origins from a failure in development of connections between lympatic and venous system. Reported localizations of canine lymphangioma include skin, subcutaneous and fascial tissue of axilla, limbs, inguinal and mammary regions; lymph nodes; retroperitoneal space; nasopharynx. In the present report we describe the computed tomographic (CT) features of a cervical cystic lymphangioma in a young dog. A 1-year-old intact male Italian Shepherd dog was referred to the primary care veterinarian with a 1-month history of left ventrolateral neck swelling. No other clinical signs were present. Ultrasonographic (US) examination revealed a mass with hyperechoic thick wall, hypoechoic content with hyperechoic fluctuating areas. Fine needle aspiration biopsy (FNAB) revealed a cloudy pinkish fluid, citologically referable to serous-hematic fluid with chronic inflamation. For better assessment of the morphology and of the margins of the lesion, the dog was referred for CT examination. Pre- and post-contrast CT scan of head, neck and thorax were made. A mass located between the muscles of the caudal neck and thoracic/axillary regions (from the level of C4 to the level of T2) was found, which partially occupied the left visceral space of the neck and bulged into the thoracic inlet. The mass was ellipsoid-shaped (40x45x140 mm), with well-defined margins and heterogeneous soft tissue attenuation. It was apparently capsulated, with fluid-like content and soft tissue attenuating septa and small areas within the fluid. Adjacent to the mass three areas of soft tissue mineralization, smoothly marginated, were found. It was responsible for mild mass effect, without significant compression on the surrounding structures. Mild left axillary and left medial retropharyngeal lymphadenomegaly was noted, with normal shape and attenuation of the limph nodes. Post-contrast images showed moderate enhancement of the mass wall and the soft-tissue-attenuating areas/septa within it; no enhancement of the fluid was noted. No other abnormalities were found. The mass was completely surgically excised and submitted for histopathologic analysis, with a definitive diagnosis of cystic lymphangioma. In human literature CT features of lymphangioma are widely described, while in veterinary literature there are no reports about its CT appearance; moreover, human lymphangioma is described as preferentially located in the neck and axillary regions, while, to our knowledge, cervical localization has never been reported before for canine lymphangioma

    Planar bone scintigraphy and CT findings in dogs with forelimb lameness

    Scintigraphy has been used for many years in veterinary medicine, due to its high sensitivity, for the localization of lameness of unknown origin in horses and for the assessment of thyroid/skeletal neoplasia in dogs. In the last few years bone scintigraphy (BS) has become increasingly used in dogs for the localization of occult lameness, when clinical examination and radiographic exam are inconclusive [1]. This study describes BS and computed tomographic (CT) findings in dogs referred for monolateral forelimb obscure lameness, for which a precise localization had not been found by clinical examination nor radiographic exam (no abnormalities at all, mild radiological abnormalities which could not be related to the grade of lameness or symmetrical bilateral alterations). Eight dogs matched inclusion criteria: 3 mixed breed, 1 Bernese mountain dog, 1 Amstaff, 1 Labrador retriever, 1 Australian shepherd and 1 Boxer. BS images showed intense IRU (Increased Radiopharmaceutical Uptake) of elbow joint in 6 cases; these findings coincided to CT alterations of proximal ulna in 5 dogs (mostly located in the medial coronoid process - MCP region: bone density alterations, evidence of fragmentation, new bone formation). In one of these cases, an intense IRU was observed in correspondence of the region of the flexors attachment, with no concurrent abnormalities on CT examination. These findings were suggestive of an obscure form of flexor enthesopathy. In one case we observed diffuse and intense IRU of the carpus joint; this coincided with arthrosis and the presence of a subchondral cyst. One dog showed only mild IRU of the elbow joint, not compatible with the degree of lameness. Because of lack of significant IRU, CT and MRI examination were performed and revealed the presence of an expansive lesion in correspondence of the brachial plexus roots compatible with PNST (Peripheral Nerve Sheath Tumor). BS' high sensitivity allowed the localization of the lameness thanks to the assessment of functional bone state, as already stated in literature [2]. However, its low specificity required additional imaging (CT, MRI), targeted on the region identified on scintigraphic examination. In our experience, the combined use of functional and morphologic diagnostic imaging techniques (bone scintigraphy and computed tomography- magnetic resonance) has been helpful to reach a definitive diagnosis. Further studies, with an increased sample size, are needed to evaluate whether there is a correlation between bone density changes and grade of IRU in limbs affected by different pathologies

    Effects of stocking density on reared Siberian sturgeon (Acipenser baerii) larval growth, muscle development and fatty acids composition in a recirculating aquaculture system

    This study evaluated the effects of rearing density on muscle growth and development in Siberian sturgeon (Acipenser baerii) larvae. Three different stocking densities were tested: low (LD, 30 larvae/l), mid (MD, 80 larvae/l) and high (HD, 150 larvae/l) in a recirculating aquaculture system. Larvae were sampled at hatching (T0), schooling (T1) and complete yolk-sac absorption (T2) stage and were weighed and processed for muscle tissue histometrical analyses and for qualitative morphological study analyses; fatty acid profile was also determined by Gas Chromatography\u2014Flame Ionization Detector analysis. Low-density larvae presented a higher weight than MD or HD at T2 (p < 0.05). Histometrical analysis revealed that total muscle area was similar at T1 and T2, but higher than T0, while it was lower at HD at schooling (p < 0.05). The fatty acid profile revealed no differences between densities while, during development, there was a selective consumption: sparing or increasing of essential fatty acids to the detriment of their precursors. Our study suggests that lower densities appear to be more suitable to rear Siberian sturgeon in this particular stage of development. Indeed, larvae reared at the lower density were heavier and longer while larvae reared at the higher density showed lower muscle proliferation rate. As a consequence, LD larvae may exert an increase of potential growth at a mid-long term

    Evaluation of in vivo response of three biphasic scaffolds for osteochondral tissue regeneration in a sheep model

    Osteochondral defects are a common problem in both human medicine and veterinary practice although with important limits concerning the cartilaginous tissue regeneration. Interest in the subchondral bone has grown, as it is now considered a key element in the osteochondral defect healing. The aim of this work was to generate and to evaluate the architecture of three cell-free scaffolds made of collagen, magnesium/hydroxyapatite and collagen hydroxyapatite/wollastonite to be implanted in a sheep animal model. Scaffolds were designed in a bilayer configuration and a novel "Honey" configuration, where columns of hydroxyapatite were inserted within the collagen matrix. The use of different types of scaffolds allowed us to identify the best scaffold in terms of integration and tissue regeneration. The animals included were divided into four groups: three were treated using different types of scaffold while one was left untreated and represented the control group. Evaluations were made at 3 months through CT analysis. The novel "Honey" configuration of the scaffold with hydroxyapatite seems to allow for a better reparative process, although we are still far from obtaining a complete restoration of the defect at this time point of follow-up

    Swine cortical and cancellous bone: histomorphometric and densitometric characterisation

    Introduction: Swine bone morphology, composition and remodelling are similar to humans\u2019, therefore they are considered good models in bone-related research. They have been used for several studies involving bone growth, bone and cartilage fractures and femoral head osteonecrosis. Nevertheless, the literature about pig normal bone features is incomplete. This work aims to fill the literature gaps on the microarchitecture and Bone Mineral Density (BMD) of swine femoral diaphysis and distal epiphysis and tibial plateau and diaphysis. Materials and methods: Five hind limbs were collected from slaughtered 80-100 kg pigs. Microscopic analysis of cortical and cancellous bone from middle/distal femur and proximal/middle tibia was performed to determine basic histomorphometric parameters at different sites. Dual-energy X-Rays Absorptiometry was also employed to evaluate BMD. ANOVA and correlation between BMD, bone area (BA) and cortical thickness were performed. Results and discussion: Diaphyseal cortical bone was mostly plexiform both in the tibia and the femur; primary/secondary osteons without clear organization were also found. Mean values for bone area, bone perimeter, trabecular width, number and separation and BMD at different anatomical sites were defined. No significant difference was found for these values at different anatomical sites. BMD proved to be positively correlated with cortical thickness (r=0,80; p<0,01). Despite the small sample size, these results seem homogeneous. They could therefore represent reference values for normal bone parameters in pigs. Applied anatomy and regenerative medicine, in fact, demand very precise information about bone micromorphology, composition and density to provide reliable indication in bone substitutes building. Moreover, since the interpretation of bone abnormalities is based on mastering normal bone characteristics, the definition of reference parameters is mandatory to avoid misinterpretation and allow comparative evaluation. Conclusion: The results of this study, although preliminary, may be considered a dependable starting point for the definition of normal bone features in pigs

    Peribulbar block in equine isolated heads : development of a single needle technique and tomographic evaluation

    Peribulbar block (PPB) has been used in humans as a safer alternative to retrobulbar block (RBB). PBB, depends on the diffusion of anaesthetic solution into the muscle across the connective tissue and it is performed introducing the needle within the extraconal space. The advantages are fewer complications and palpebral akinesia. In Veterinary Medicine few studies describe this technique in dogs and cats (Shilo- Benjamini et al., 2013). The aim of the study is to determinate, in equine specimens, feasibility of inferior PBB with single needle injection, by using contrast medium (CM), and to evaluate thought Computed Tomography (CT) the distribution of the injected volume and regional anaesthesia likelihood. PBB was performed in 10 orbits. The mixture injected consisted of 20 ml of physiological solution and iodinated CM at 25%. Each periorbital area underwent three CT scans. A basal acquisition to assess the needle position before the injection, a second and third scan were performed immediately after injection, and after application of pressure on the periorbital surface area to promote CM diffusion. The injectate distribution at the base and within the extraocular muscle cone (EOMC) and around the optic nerve was evaluated and scored based on Shilo-Benjamini\u2019s work of 2017. The mean minimum distance between the tip of the needle and the optic was 2,23 mm \ub10,2. The mean volume distribution before pressure application was 23.56 cm3 \ub1 2.58 and after pressure application was 27.56 cm3 \ub1 4.8. The CM median distribution around the optic nerve at the base of the EOMC was of 117\ub0 prior pressure and 189\ub0 after pressure. The CM distribution within the EOMC was present in 1 orbit prior pressure and in 3 orbits after pressure. The CM distribution at the base of EOMC was considered unlikely to provide regional anaesthesia in 2 orbits, possible in 3 orbits and likely in 5. In the present study, intraconal distribution was not consistent. For this reason, the likelihood of achieving regional anaesthesia was evaluated at the EOMC base where through the optic foramen the oculomotor, trochlear nerve, ophthalmic branch of the trigeminal nerve, and the abducens travel to reach the orbit together with the optic nerve. Whereas the maxillary branch of the trigeminal nerve passes through the foramen rotundum (Carastro 2004). Therefore, despite the lack of intraconal distribution if the EOMC base had good distribution then it was considered likely to provide regional anaesthesia. This approach needs to be evaluated in clinical trials to assess its feasibility and effectiveness in locoregional anaesthesia; moreover, further investigations on equine PBB are mandatory with higher volumes of injectate and different approaches

    Advances in the anatomic study of the interscapular region of the cat

    New clinical oriented approaches are supported by the integration of advanced imaging techniques, e.g. computed tomography and magnetic resonance, with gross anatomy imaging. The interscapular region of the cat is a typical site of a highly invasive infiltrative pathology, i.e. Feline Injection-Site Sarcoma. Even if chemotherapy and radiotherapy have been considered as pre-surgical therapies, extensive surgery is still the recommended treatment. Evidence suggested that the relationships between muscles, infiltrative mass and adjacent musculoskeletal structures could change according to the forelimb positions: a fact to be duly considered while planning the surgical approach. Anatomic and tomographic atlases provide only images of the interscapular region from cats positioned with their forelimbs extended cranially, which means that, they do not record musculoskeletal modifications due to the forelimb movements. Aim of this study was to provide detailed images of the changes occurring in the musculoskeletal structures of the interscapular region of cats according to their forelimb position by comparing cross-sectional gross anatomy, computed tomography and magnetic resonance imaging

Computed Tomography (CT) and Magnetic Resonance Imaging (MRI) proved to be very accurate and valuable tools in estimating body and carcass composition in farm animals.1 CT has been successfully used for the characterization of food Italian products such as salami, providing a precise evaluation of fat percentage, also assessing its spatial distribution.2 Manzocco and colleagues demonstrated that MRI has great potential in monitoring the evolution of dry curing in S. Daniele hams.3In our experience, helical CT proved to be a fast tool in the classification of different meat cuts, deriving from adult cow and destined for the preparation of aircured products as \u201clean meat\u201d or \u201cfat meat\u201d, both in fresh and frozen samples. Histological studies confirmed that CT clearly distinguishes adipose and connective tissue infiltration within muscles and that semi-quantitative analysis of infiltration degree can be achieved. These data were further supported by the chemical analysis of meat samples corresponding to the same region of interest observed in both CT and histologic investigations; dry matter, crude proteins, crude fat and ash contents, calculated following standard international methods,4 varied in fact depending on the fat infiltrated extent, according with CT images. We finally observed that CT could be used in the evaluation of the same products at the end of ripening, without removing the outer envelope. These results are important for beef and meat industrial processing sector, suggesting that CT could be employed as an on-line instrument in abattoir and dry-cured meat industry in classifying the products at the beginning of the manufacturing even they are frozen. Moreover, it might represent a rapid and non-invasive technique for quality check at the end of the production line and for the assignment of the most appropriate nutritional and commercial value to different products.
