ErbB2, EphrinB1, Src Kinase and PTPN13 Signaling Complex Regulates MAP Kinase Signaling in Human Cancers
In non-cancerous cells, phosphorylated proteins exist transiently, becoming de-phosphorylated by specific phosphatases that terminate propagation of signaling pathways. In cancers, compromised phosphatase activity and/or expression occur and contribute to tumor phenotype. The non-receptor phosphatase, PTPN13, has recently been dubbed a putative tumor suppressor. It decreased expression in breast cancer correlates with decreased overall survival. Here we show that PTPN13 regulates a new signaling complex in breast cancer consisting of ErbB2, Src, and EphrinB1. To our knowledge, this signaling complex has not been previously described. Co-immunoprecipitation and localization studies demonstrate that EphrinB1, a PTPN13 substrate, interacts with ErbB2. In addition, the oncogenic V660E ErbB2 mutation enhances this interaction, while Src kinase mediates EphrinB1 phosphorylation and subsequent MAP Kinase signaling. Decreased PTPN13 function further enhances signaling. The association of oncogene kinases (ErbB2, Src), a signaling transmembrane ligand (EphrinB1) and a phosphatase tumor suppressor (PTPN13) suggest that EphrinB1 may be a relevant therapeutic target in breast cancers harboring ErbB2-activating mutations and decreased PTPN13 expression

Relationships between the functionality of the interchange node and planning of connections in the transport network
Punktem wyjścia rozważań są wymagania poprawy funkcjonalności
węzła przesiadkowego w naziemnym miejskim transporcie
publicznym (integrującego tramwaje, autobusy, ewentualnie trolejbusy).
Poprawa funkcjonalności ma tu dotyczyć przede wszystkim zwiększenia
przepustowości węzła (umożliwienia obsługi większej liczby kursów/
pojazdów) i ograniczenia strat czasu poprzez redukcję zatrzymań
i nieuzasadnionych postojów, przy zachowaniu jego zwartości i wygody
podróżnych. Ta część artykułu bazuje na analizie funkcjonujących węzłów
oraz publikacji ich dotyczących (w tym autorskich). Sformułowane
są zalecenia odnośnie pożądanej formy węzła (jako rozwiązanie docelowe).
Wskazuje się także możliwe kierunki i sposoby przekształceń
istniejących obiektów. Jednym z zaleceń jest uproszczenie liczby relacji
w ramach węzła, co ma wpływ na planowanie całej sieci transportu
publicznego w skali miasta czy nawet aglomeracji. Dotyczy to układu
linii, liczby kursów, lokalizacji i rozplanowania węzłów itp. Te aspekty
wpływu zmian formy poszczególnych węzłów na planowanie i modelowanie
przewozów w całej sieci są główną częścią artykułu. W artykule
przedstawiono uwagi ogólne oraz przykład konkretnych przekształceń
wybranego węzła we Wrocławiu. Celem proponowanych działań (rozwiązań)
jest umożliwienie zwiększenia liczby połączeń w sieci, redukcja
strat czasu i, w konsekwencji, pozyskanie większej liczby pasażerów
(zmiany w podziale zadań przewozowych). W końcowej części artykułu
zainicjowana jest dyskusja nad tym, jak wyżej opisane uwarunkowania
można uwzględnić w modelowaniu podróży.The requirements to improve the functionality of the interchange
node in above-ground urban public transport (integrating
trams, buses or trolleybuses) are the starting point for the considerations.
The improvement of functionality is to be mainly related to
increasing the capacity of the node (enabling the servicing of a larger
number of courses / vehicles) and limiting delays by reducing detentions
and unjustified stops, while maintaining its compactness
and convenience for travelers. This part of the article is based on the
analysis of functioning nodes and publications regarding them (including
author’s). Recommendations are formulated regarding the
desired form of the node (as a target solution). The possible directions
and ways of transforming existing objects are also indicated. One of
the recommendations is to simplify the number of relations within
the node, which affects the planning of the entire public transport
network in the scale of the city or even the agglomeration. This applies
to the layout of the line, the number of courses, the location
and layout of nodes, etc. These aspects of the impact of changing
the form of individual nodes on planning and modeling of transport
throughout the network are the main part of the article. General
comments and an example of specific transformations of the selected
node in Wrocław are presented. The goal of the proposed activities
(solutions) is to enable an increase in the number of connections in
the network, reduction of delays and, as a consequence, acquisition
of a larger number of passengers (changes in the modal split). The
discussion on how the conditions described above can be taken into
account in travel modeling is initiated in the final part of the article

Intelligent transport systems and mobility policy
Intelligent Transport System (ITS) in the city and the metropolitan area should be linked to the Mobility Policy (MP) formulated and implemented for this area. On the one side, the implementation of the ITS is one of the MP tools. On the other side, the specific parameters of the ITS (algorithms, tools, efficiency measures) associated with the elements of MP such as its objectives, zoning, priorities. On the basis of the review of model components of MP and IT’S THE common elements and significant correlation are summarized. The theoretical considerations are supplemented with examples from Wroclaw. This article is a continuation of the author’s work and thoughts published under the title of both issues. At their beginning the basic elements of MP and ITS are defined. Then, the ITS is described as a tool in the context of MP’s objectives, zoning and priorities. Next, the ITS parameters such as algorithms, tools, efficiency measures are defined in terms of the MP. Later, the author summarizes the common characteristics and significant correlation between ITS and MP. This allows to provide dedicated solutions to Wroclaw. The summary of the article are desirable directions of integration between MP and ITS. The key issue is here the implementation of integration of management and development among MP and ITS
The proposal of introduction of an underground sections in the Wroclaw’s public transport
Artykuł przedstawia autorską propozycję wprowadzenia
odcinków podziemnych do systemu transportu zbiorowego
Wrocławia. Przesłanką do takich planów jest potencjalny brak
przepustowości istniejących tras w centrum wynikający z koncepcji
rozwoju transportu szynowego miasta i aglomeracji. Oznacza to
między innymi intensyfikację liczby linii oraz kursów. Wspomnianą
koncepcję rozwoju, również sformułowaną przez autora niniejszej
publikacji, cytuje się w artykule, opisując najważniejsze jej elementy
w postaci liczby kursów na godzinę w kluczowych odcinkach sieci.
Formułowane są dwa rozwiązania tunelowe, jedno dla kolei aglomeracyjnej
dla połączenia dwóch dworców i przybliżenia linii do
Starego Miasta, drugie dla linii tramwajowych łączących średnicowo
osiedla zachodnie i północno-wschodnie. Specyfiką tunelu tramwajowego
jest jego sytuowanie w nasypie Fosy Miejskiej z odkryciem na
taflę wody. Tunel kolejowy zatem byłby lokalizowany głębiej i budowany
metodami górniczymi, a tunel tramwajowy, przy płytkim
prowadzeniu, mógłby zostać wykonany metodą odkrywkową. Obie
inwestycje są porównywane z podobnymi, niedawno zrealizowanymi
lub planowanymi, w miastach o wielkości podobnej do Wrocławia
(kolej: Lipsk, Łódź; tramwaj: Poznań). Pozwala to wskazać na realność
sfinansowania potencjalnej budowy. Zarysowane są także alternatywne
opcje, niewymagające budowy tuneli wraz z podaniem wad
i zalet tych alternatyw. W podsumowaniu, autor odwołuje się do
Wrocławskiej Polityki Mobilności, uzasadniając proponowane rozwiązania
zapisami tego dokumentu. Konieczność rozwoju transportu
zbiorowego wynika z zapisów o docelowych poziomach podziału
zadań przewozowych (modal split). Decyzja o wdrożeniu prezentowanych
koncepcji powinna być podparta modelowaniem ruchu w skali
całej aglomeracji.The article presents an original proposal to introduce an
underground sections in public transport system of Wroclaw. The idea
is caused by potential lack of capacity of existing routes in the city
center due to the concept of development of rail transport of the city
and agglomeration. This development means, among other things,
increasing of number of lines and courses. Two tunnel-solutions are
planned. The first one would be designed for the suburban railway network
connecting two stations and bringing the line near to the Old
Town. The second one would be designed for the tram lines connecting
diametrically settlements located in West and North-East part of
the city. The tram tunnel is characterized by its location in the scarp of
the City Moat with the outdoor on the surface of the water. Therefore
the railway tunnel would be located deeper and constructed with the
mining methods. The tram tunnel, at a shallow location, could be
constructed with open-pit mining. Both investments are comparable
with similar, recently completed or planned tunnels in cities of
similar size to Wroclaw (railway in Leipzig, tram in Poznan). This
enables to state that this potential contruction is possible to finance.
Also alternatives that do not require tunneling are mentioned, with
description of their advantages and disadvantages. In conclusion, the
author refers to the Wroclaw Mobility Policy justifying the proposed
solutions in the reference to this document. The need to develop of
public transport system should be a result of the assumed target levels
of modal split. The decision to implement the concepts should
be supported by the modeling of traffic in the whole agglomeration
Development of tram network in Poland as a background to the Wrocław Tram Program
Artykuł omawia dwa tematy: historię i stan obecny sieci
tramwajowych w Polsce oraz plany rozwoju sieci we Wrocławiu formułowane
pod postacią wieloletniego programu inwestycyjnego. Historia
rozwoju pokazuje kolejne etapy: pionierski, dominacji tramwajów i intensywnego
rozwoju, likwidacji sieci i stagnacji rozbudowy, wreszcie „renesans”
jako echo europejskich i światowych tendencji. Zestawiono nie
tylko sieci funkcjonujące, ale także opisano miasta, w których zlikwidowano
tramwaje. Przybliżono sieć w Olsztynie – jak dotąd jedyną reaktywowaną
po 50 latach nieistnienia. Podano dane statystyczne odnośnie
długości sieci, okresu funkcjonowania w poszczególnych miastach i aglomeracjach.
Ograniczono się do trakcji elektrycznej dla miast w obecnych
granicach Polski. Wyróżniono 28 lokalizacji (w tym ośrodki składające
się z wielu miejscowości, jak GOP czy aglomeracja Łodzi). Opis uzupełniają
tabele oraz mapa z lokalizacją sieci. Tło historyczne pozwala pełniej
ogarnąć zamierzenia wrocławskie. Pod nazwą Wrocławski Program
Tramwajowy ogłoszono pod koniec roku 2016 wizję znacznej rozbudowy
sieci tramwajowej. W przeciągu 20–30 lat może nastąpić niemal podwojenie
długości sieci. Wskazano na zadania pierwszoplanowe wpisane do
Planu Transportowego uchwalonego na lata 2017–2022 oraz inwestycje
do realizacji w kolejnych latach. Ostateczny horyzont planu rozbudowy
sieci tramwajowej określono na rok 2042. Oprócz nowych tras wskazano
na węzły przesiadkowe (na kolej aglomeracyjną oraz w systemie Park and
Ride). Na mapach pokazano sieć planowaną na rok 2022 oraz na rok
2042. Jako podsumowanie artykułu sformułowano dylematy związane ze
skuteczną realizacją programu.The article discusses two topics: first, the history and
current state of tram network in Poland, second, the network
growth plans in Wrocław formulated in the form of longterm investment
program. The history of development shows the sequential
steps of: a pioneering, trams domination and intensive growth,
elimination of network expansion and stagnation, and finally the
"renaissance" as an echo of European and global trends. Statistical
data regarding the length of the network, the period of operation
in the individual cities and agglomerations have been given. They
are limited to electric traction for cities in the current Polish borders.
Twenty eight locations have been included. The description
is supplemented by tables and a map of the network. Historical
background can more fully grasp the intentions of Wrocław.
Under the name Wrocław Tram Program was announced the vision
of a large expansion of the tram network at the end of 2016.
Within 20–30 years the length of the network can be almost doubled.
Foreground tasks included in the Transport Plan adopted for
the years 2017–2022 have been identified, as well as investments
to be implemented in subsequent years. The network planned for
the years: 2022 and 2042 are shown on maps. Dilemmas related
to the effective implementation of the program was formulated as
a summary of the article
Modal split – in a problem approach
W literaturze przedmiotu (teoria) modeluje się wybór środka lokomocji (parametr „podział zadań przewozowych”) z uwzględnianiem odmiennych motywacji podróży, kategorii osób, czasu i trasy przejazdu. Tymczasem w dokumentach o charakterze użytkowym (praktyka) podział zadań przewozowych traktowany jest jako prosty wskaźnik charakteryzujący na przykład cele projektu (polityki) bez różnicowania go w wyżej wymienionych aspektach. Autor zwraca uwagę na problemy interpretacji podawanych wartości podziału zadań przewozowych między innymi w kontekście oceny efektywności wdrażanych rozwiązań względem zakładanych celów oraz proponuje uszczegółowienie zasad stosowania tego parametru.In the literature (theory) the choice of transport means (parameter "modal split") is modeled from mainstreaming different trip motivation, a category of people, travel time and route. While, in the utilitarian documents (practice), the modal split is treated as a simple indicator characterizing, for example, the objectives of the project (policy) without differentiating it in the above-mentioned aspects. Author draws the attention to the problems related to the interpretation of declared values of the modal split, inter alia, in the context of assessment of the effectiveness of implemented solutions in relation to its objectives and proposes a clarification of the rules for applying this parameter
Modelling of travels to agglomeration’s central city from surrounding villages
W artykule przedstawiono uwarunkowania modelowania podróży pomiędzy centralnym miastem aglomeracji a otaczającymi go miejscowościami. Na tle stosowanych metod i elementów modeli (przegląd literatury) wyspecyfikowano aktualne uwarunkowania nawiązujące do badań przeprowadzonych dla aglomeracji wrocławskiej. Autor wskazał istotne wyniki prowadzonych przez siebie badań oraz, posiłkując się „studium przypadku” dla wybranych miejscowości, omówił proponowane przez siebie elementy modelu.The paper presents the conditions to modelling of travels between the central city and the surrounding villages. Against the background of methods and elements of models (literature review), the current conditions referring to research carried out for the agglomeration of Wrocław are specified. The author pointed out important results of their own research and discussed the proposed elements of the model with use of a “case study” for the selected village
