    Tracking of Salmonella through the Pork Slaughter Process

    End of project reportTo help address the problem of salmonellosis in the Republic of Ireland (RoI), a national Salmonella control programme was introduced in 1997 with a view to reducing the prevalence of Salmonella in pigs on the farm and on pig carcasses. The primary objective of this present study was to determine the correlation between the Salmonella serological and bacteriological status of pigs presented for slaughter and the Salmonella status of pork cuts following slaughter, dressing and chilling. Two additional studies investigated the prevalence and numbers of Salmonella spp. in the boning halls of four commercial pork abattoirs and at retail level in butcher shops and supermarkets in the RoI. The results indicated that categorisation of pig herds on the basis of a historical serological test for Salmonella was not a good predictor of the bacteriological Salmonella status of individual pigs at time of slaughter. However, it is acknowledged that serological testing does help in giving a rough estimate of the overall Salmonella status of a pig herd. There was a linear correlation between prevalence of Salmonella in caecal contents and on pork cuts at factory level; therefore, if the number of herds presented for slaughter with high levels of Salmonella (category 3) was reduced, there would be less potential for contamination of the lairage, equipment etc. and so less likelihood of Salmonella contamination on pork. The impact of crosscontamination during transport, lairage, processing and distribution cannot be ignored and measures to diminish this would significantly reduce the dissemination of Salmonella in the chain and the consequent risk posed.     Wild record of an apple snail in the Waikato River, Hamilton, New Zealand and their incidence in freshwater aquaria

    We report the discovery of a single specimen of a live apple snail Pomacea diffusa Blume 1957 (Ampullariidae: Prosobranchia), from the Waikato River, Hamilton city, central North Island, New Zealand. This species, along with the congeneric P. insularum, is imported for the aquarium trade, and its occurrence in the river likely stemmed from an aquarium release. A survey of 55 aquaria belonging to 43 hobbyists revealed 27 apple snails, with one owner having 22 snails. Assessment of environmental tolerances and impacts of P. diffusa, based largely on studies of the closely related and commonly confused congener P. bridgesii, suggests that direct habitat impacts by this species are likely to be minor. However, there could be indirect influences on native biodiversity through predation on eggs or competition for food supplies with other detritivorous species if densities were to become high. Water temperatures in the Waikato River below Hamilton (10-23˚C in 2009) may enable released individuals to persist for an extended period, and over summer may exceed the threshold required to enable breeding. However, population establishment would be most likely in locations where water is heated through geothermal influences or industrial cooling water discharges

    The influence of goal-state access cost on planning during problem solving

    Two problem-solving experiments investigated the relationship between planning and the cost of accessing goal-state information using the theoretical framework of the soft constraints hypothesis (Gray & Fu, 2004; Gray, Simms, Fu, & Schoelles, 2006). In Experiment 1, 36 participants were allocated to low, medium, and high access cost conditions and completed a problem-solving version of the Blocks World Task. Both the nature of planning (memory based or display based) and its timing (before or during action) changed with high goal-state access cost (a mouse movement and a 2.5-s delay). In this condition more planning before action was observed, with less planning during action, evidenced by longer first-move latencies, more moves per goal-state inspection, and more short (≤0.8 s) and long (>8 s) “preplanned” intermove latencies. Experiment 2 used an eight-puzzle-like transformation task and replicated the effect of goal-state access cost when more complex planning was required, also confirmed by sampled protocol data. Planning before an episode of move making increased with higher goal-state access cost, and planning whilst making moves increased with lower access cost. These novel results are discussed in the context of the soft constraints hypothesis

    Opening dialogue and fostering collaboration: different ways of knowing in fisheries research

    We set out to explore some of the impediments which hinder effective communication among fishers, fisheries researchers and managers using detailed ethnographic research amongst commercial handline fishers from two sites- one on the southern Cape coast and the other on the west coast of South Africa. Rather than assuming that the knowledge of fishers and scientists is inherently divergent and incompatible, we discuss an emerging relational approach to working with multiple ways of knowing and suggest that this approach might benefit future collaborative endeavours. Three major themes arising from the ethnographic fieldwork findings are explored: different classifications of species and things; bringing enumerative approaches into dialogue with relational approaches; and the challenge of articulating embodied ways of relating to fish and the sea. Although disconcertments arise when apparently incommensurable approaches are brought into dialogue, we suggest that working with multiple ways of knowing is both productive and indeed necessary in the current South African fisheries research and management contexts. The research findings and discussion on opening dialogue offered in this work suggest a need to rethink contemporary approaches to fisheries research in order to mobilise otherwise stagnant conversations, bringing different ways of knowing into productive conversation

    Sex-dependent influence of endogenous estrogen in pulmonary hypertension

    Rationale: The incidence of pulmonary arterial hypertension (PAH) is greater in women suggesting estrogens may play a role in the disease pathogenesis. Experimentally, in males exogenously administered estrogen can protect against PH; however in models that display female susceptibility estrogens may play a causative role. Objectives: To clarify the influence of endogenous estrogen and gender in PH and assess the therapeutic potential of a clinically available aromatase inhibitor. Methods: We interrogated the effect of reduced endogenous estrogen in males and females using the aromatase inhibitor, anastrozole, in two models of PH; the hypoxic mouse and Sugen 5416/hypoxic rat. We also determined the effects of gender on pulmonary expression of aromatase in these models and in lungs from PAH patients. Results: Anastrozole attenuated PH in both models studied, but only in females. To verify this effect was due to reduced estrogenic activity we confirmed that in hypoxic mice inhibition of estrogen receptor alpha also has a therapeutic effect specifically in females. Female rodent lung displays increased aromatase and decreased BMPR2 and Id1 expression compared to male. Anastrozole treatment reversed the impaired BMPR2 pathway in females. Increased aromatase expression was also detected in female human pulmonary artery smooth muscle cells compared to male. Conclusions: The unique phenotype of female pulmonary arteries facilitates the therapeutic effects of anastrozole in experimental PH confirming a role for endogenous estrogen in the disease pathogenesis in females and suggests aromatase inhibitors may have therapeutic potential

    Assay of matrix metalloproteinases types 1, 2, 3 and 9 in breast cancer.

    Matrix metalloproteinases (MMPs) are zinc dependent endopeptidases implicated in cancer invasion and metastasis. Gelatin zymography was performed on 84 human breast carcinomas and seven normal breast tissues. The precursor form of MMP-2 (72 kDa) was found in 11 (12%) samples, while its two activated forms, i.e. 62 kDa and 59 kDa, were found in three (6%) and 34 (40%) samples respectively. In contrast to MMP-2, most of the samples (52%) contained MMP-9 in its precursor form. Using ELISA, MMP-1 levels were found in 12% of the samples while MMP-3 levels were found in only 2% of the samples. Levels of MMP-2, -3 and -9 correlated inversely with numbers of nodal metastases. Neither MMP-2 nor -9 levels were significantly related to patient outcome. However, patients with high levels of a 50-kDa gelatinase band after zymography had a significantly better survival than patients with low levels. This species was never observed in normal breast tissue
