    Improved bioavailability of timolol maleate via transdermal transfersomal gel: Statistical optimization, characterization, and pharmacokinetic assessment

    AbstractTimolol maleate (TiM), a nonselective β-adrenergic blocker, is a potent highly effective agent for management of hypertension. The drug suffers from extensive first pass effect, resulting in a reduction of oral bioavailability (F%) to 50% and a short elimination half-life of 4h; parameters necessitating its frequent administration. The current study was therefore, designed to formulate and optimize the transfersomal TiM gel for transdermal delivery. TiM loaded transfersomal gel was optimized using two 23 full factorial designs; where the effects of egg phosphatidyl choline (PC): surfactant (SAA) molar ratio, solvent volumetric ratio, and the drug amount were evaluated. The formulation variables; including particle size, drug entrapment efficiency (%EE), and release rate were characterized. The optimized transfersomal gel was prepared with 4.65:1 PC:SAA molar ratio, 3:1 solvent volumetric ratio, and 13mg drug amount with particle size of 2.722μm, %EE of 39.96%, and a release rate of 134.49μg/cm2/h. The permeation rate of the optimized formulation through the rat skin was excellent (151.53μg/cm2/h) and showed four times increase in relative bioavailability with prolonged plasma profile up to 72h compared with oral aqueous solution.     Poultry and Beef Meat as Potential Seedbeds for Antimicrobial Resistant Enterotoxigenic Bacillus Species: A Materializing Epidemiological and Potential Severe Health Hazard

    Although Bacillus cereus is of particular concern in food safety and public health, the role of other Bacillus species was overlooked. Therefore, we investigated the presence of eight enterotoxigenic genes, a hemolytic gene and phenotypic antibiotic resistance profiles of Bacillusspecies in retail meat samples. From 255 samples, 124 Bacillus isolates were recovered, 27 belonged to B. cereusand 97 were non-B. cereus species. Interestingly, the non-B. cereus isolates carried the virulence genes and exhibited phenotypic virulence characteristics as the B. cereus. However, correlation matrix analysis revealed the B. cereus group positively correlates with the presence of the genes hblA, hblC, and plc, and the detection of hemolysis (p \u3c 0.05), while the other Bacillus sp. groups are negatively correlated. Tests for antimicrobial resistance against ten antibiotics revealed extensive drug and multi-drug resistant isolates. Statistical analyses didn’t support a correlation of antibiotic resistance to tested virulence factors suggesting independence of these phenotypic markers and virulence genes. Of special interest was the isolation of Paenibacillus alvei and Geobacillus stearothermophilus from the imported meat samples being the first recorded. The isolation of non-B. cereus species carrying enterotoxigenic genes in meat within Egypt, suggests their impact on food safety and public health and should therefore not be minimised, posing an area that requires further research

    Alternate utility responses for possible risks of lower frequencyelectromagnetic fields

    This paper outlines a series of alternate responses that electric utilities might take in the face of the possible risks posed by human exposure to low frequency electric and magnetic fields. There are basically three arguments that can be used to justify limiting people's exposures to low frequency fields: safety, equity and prudence. A safety based standard would limit field exposure to those exposure circumstances which do not pose a risk to health. Scientifically based safety standard is not possible today due to conflicting results of field effects. Standards based on considerations of equity do not provide any assurance of safety. Regulations based on prudence are designed to keep people out of fields with modest investments of time and resources. This paper explores thoroughly different options, some of which could be combined, that electric utilities may adopt as strategy and approach in the face of field effects issue. These options include: denial, passive and active information supply, research and development support, limited response for new facilities or major response, elimination of selected man-power line field exposures, and limited and major retrofits of old facilities.
