    ACE/VDR gene polymorphisms and bioelectrical impedance analysis in predicting athletic performances of Italian Young soccer players.

    We evaluated the association between 2 genetic polymorphisms known to be involved in fitness and performance, and anthropometric features, body composition, and athletic performances in young male soccer players with the goal of identifying genetic profiles that can be used to achieve maximal results from training. One hundred twenty-five medium-high-level male soccer players were genotyped for angiotensin-converting enzyme (ACE) I/D, and vitamin D receptor (VDR) FokI gene polymorphisms and scored for anthropometric measurements, body composition, and athletic performance. Body mass index, fat mass, fat-free mass, resistance, reactance, impedance, phase angle (PA), and body cell mass were measured. Athletic performance was evaluated by squat jump, countermovement jump (CMJ), 2-kg medicine ball throw, 10- and 20-m sprint time. We observed that the homozygous ff genotype of the VDR gene was significantly more represented in young soccer players than in a matched sedentary population. Values of reactance and PA were differently distributed in ACE and VDR genotypes with high mean values in subjects with DD (ACE) and FF (VDR) genotypes. No correlation was observed between ACE or VDR genotypes and 2-kg medicine ball throw, 10- and 20-m sprint times. The ID genotype of ACE was associated with the best performances in squat jump and CMJ. Our results suggest that determination of ACE and VDR genotypes might help select those young athletes harboring the most favorable genetic potential to succeed in soccer

    Daily consumption of a high-phenol extra-virgin olive oil reduces oxidative DNA damage in postmenopausal women

    Extra-virgin olive oils (EVOO), high in phenolic compounds with antioxidant properties, could be partly responsible for the lower mortality and incidence of cancer and CVD in the Mediterranean region. The present study aims to measure oxidative DNA damage in healthy human subjects consuming olive oils with different concentrations of natural phenols. A randomised cross-over trial of high-phenol EVOO (high-EVOO; 592 mg total phenols/kg) v. low-phenol EVOO (low-EVOO; 147 mg/kg) was conducted in ten postmenopausal women in Florence. Subjects were asked to substitute all types of fat and oils habitually consumed with the study oil (50 g/d) for 8 weeks in each period. Oxidative DNA damage was measured by the comet assay in peripheral blood lymphocytes, collected at each visit during the study period. Urine samples over 24 h were collected to measure the excretion of the olive oil phenols. The average of the four measurements of oxidative DNA damage during treatment with high-EVOO was 30 % lower than the average during the low-EVOO treatment (P=0.02). Urinary excretion of hydroxytyrosol and its metabolite homovanillyl alcohol were significantly increased in subjects consuming high-EVOO. Despite the small sample size, the present study showed a reduction of DNA damage by consumption of an EVOO rich in phenols, particularly hydroxytyrosol

    Association between the BRCA2 N372H variant and male breast cancer risk: a population-based case-control study in Tuscany, Central Italy

    Background: Male breast cancer (MBC) is a rare disease and little is known about its aetiology. Germline mutations of BRCA2 and, at lower frequency, of BRCA1 are implicated in a relatively small proportion of MBC cases. Common polymorphic variants in BRCA1 and BRCA2 genes may represent breast cancer (BC) susceptibility alleles and could be associated with a modestly increased risk of MBC at population level. Considering the relevant role of BRCA2 in MBC, we investigated whether the BRCA2 N372H variant, representing the only common non-synonymous polymorphism in BRCA2, might modulate the risk of BC in male populations.Methods: A case-control study was performed comparing a population-based series of 99 MBC cases, characterized for BRCA1 and BRCA2 mutations, with 261 male population controls, all residing in Tuscany, Central Italy. All MBC cases and controls were genotyped for the BRCA2 N372H allele by TaqMan allelic discrimination assays. To evaluate the genotype specific risk of the BRCA2 N372H variant, MBC carriers of germ-line BRCA1/2 mutations were excluded from the analyses.Results: No association emerged in univariate and age-adjusted analyses. Age-specific analyses suggested an increased risk for the HH homozygous genotype in subjects younger than 60 years. A statistically significant interaction emerged between this genotype and age (p = 0.032). When analyses were restricted to MBC cases enrolled in the first 4 years following diagnosis, a recessive model showed a significantly increased risk of MBC in HH subjects younger than 60 years (OR = 5.63; 95% CI = 1.70; 18.61).Conclusion: Overall, our findings, although based on a relatively small series, suggest that the BRCA2 HH homozygous genotype might be positively associated with an increased risk of MBC in men younger than 60 years

    Omega-6 and omega-3 polyunsaturated fatty acid levels are reduced in whole blood of Italian patients with a recent myocardial infarction: the AGE-IM study

    OBJECTIVE: The relationship between whole blood fatty acids and myocardial infarction (MI) risk has not been analyzed in detail, especially in Mediterranean countries. The AGE-IM (Acidi Grassi Essenziali e Infarto Miocardico) study was planned to examine the relationships between MI, whole blood fatty acids and the diet in an Italian cohort. METHODS: 119 Patients with a recent MI and 103 control subjects were enrolled in the study. The whole blood fatty acid composition was determined; information on anthropometrics, biochemical parameters and blood pressure values were also obtained. Diet composition was assessed using a validated food frequency questionnaire from 86 cases and 72 controls. RESULTS: Total PUFA, omega-6 and omega-3 PUFA (as percentage of whole blood fatty acids) were significantly lower in MI patients than in matched controls, whereas saturated and monounsaturated fatty acids were higher in cases. MI infarction risk significantly and steadily decreased with increasing levels of total PUFA (OR: 0.14) and of total omega-6 and omega-3 (OR: 0.15 and 0.37, respectively). No correlation was identified between dietary fats and MI risk or between whole blood fatty acid levels and dietary nutrients and fats. CONCLUSION: Percentage levels of total PUFA, total omega-3 PUFA and total omega-6 PUFA are lower in MI patients than in matched control subjects in the AGE-IM cohort. These data support a favorable association not only of whole blood percentage levels of total omega-3, but also of total omega-6, with cardiovascular risk

    BACKGROUND: Sun sensitivity is one of the predictors of melanoma risk, together with other individual characteristics such as skin and eye colour and number of naevi. However, it is unclear how best to measure sun sensitivity in order to quantify the individual risk of melanoma. OBJECTIVES: In this case-control study, the relationship between minimal erythema dose (MED) and skin colour (both instrumentally assessed) was investigated, and their possible role as independent risk factors for melanoma in a Mediterranean population evaluated. METHODS: In total, 143 patients with cutaneous melanoma and 102 controls were enrolled in the study. Skin colour was assessed using a Minolta CR-200 chromameter. For MED calculation, a fluorescent lamp (Philips TL 4W/12) was used as a source of ultraviolet B light. MED was defined as the lowest dose that produced an increase of 2.5 in the redness value, expressed by the parameter a* of the Commission Internationale d'Eclairage (CIE) L*a*b* colour space (Deltaa* = 2.5). RESULTS: A significant excess of risk was associated with increasing L* values of skin colour (P 97.7 mJ/cm(2)), subjects with MED values 2-fold increased risk of melanoma (OR = 2.37, 95% CI 1.05-5.38). The effect of decreasing MED value as a melanoma risk factor persisted after adjustment for skin colour and atypical naevi in a multivariate model. CONCLUSIONS: In conclusion, both instrumentally assessed skin colour and MED are significant risk factors for malignant melanoma in a Mediterranean population. MED seems be an independent variable in establishing the subject's risk profile

    The increasing understanding of the genetic influences in sport has prompted an association study between the athletic performances and the polymorphisms of the angiotensin-converting enzyme (ACE), the α-actinin-3 (ACTN3), and the vitamin D receptor genes. The details of these gene polymorphisms can provide useful information to improve and plan new modern training programs for elite athletes. Eighty Italian male high level gymnasts were trained and tested for gymnastic-specific exercises and tested in all the men's artistic gymnastic apparatus (floor, pommel horse, rings, vault, parallel bars, and horizontal bar), and then genotyped. The training parameters of volume, intensity, and density of each gymnast were periodically measured during the season in each apparatus from the tests performed, and the seasonal average values were calculated. Gene polymorphisms were determined by polymerase chain reaction restriction fragment length polymorphism assay and studied in association with the performance results. The performances of ACE II gymnasts were significantly lower than that of the ACE ID/DD gymnasts in the apparatus expressing power features, confirming the predisposition of these athletes toward power-oriented sport. Gymnasts with ACTN3 RR/RX genotypes did not show a predisposition to the power-oriented apparatus, having worse performances compared with that of the ACTN3 XX gymnasts. Similarly, gymnasts with ACE II + ACTN3 RR/RX combined genotypes showed lower performances in comparison with that of the other gymnasts. Vitamin D receptor polymorphisms showed no significant association with the athletic performances. Because ACE insertion/deletion (I/D) and ACTN3 R577X polymorphisms heavily affect the physical performance of elite male gymnasts, the Italian Gymnastic Federation trainers have started to customize the current high-level training programs

    Integrated Approach for a Successful Geothermal Wells Location in the Mt. Amiata Area (Southern Tuscany)

    Sorgenia Geothermal has been recently awarded some exploration licenses in Southern Tuscany, near the geothermal fields of Bagnore and Piancastagnaio, around the Mt. Amiata volcano complex. In order to successfully locate the first deep wells, an integrated exploration program has been developed, firstly based on a complete collection and analysis of existing critical data, which include mining data, previously drilled geothermal wells data and some geophysical data (gradient wells data, gravimetric data, geoelectrical data, CROP 18B and SIF 12–15/87 seismic reflection raw data). The available seismic data have been reprocessed in order to better focus on the geothermal target and at the same time a new planned 2D seismic reflection survey (approx. 50 km) will be carried out in 2014 to achieve the best coverage in the areas under exploration. An MT survey has been carefully designed and performed (250 stations) considering the severe increase of EM noise in the areas. Geological and hydrogeological surveys, as well as the soil CO2 degassing measurements, a useful tool in confirming the existence of a deep-seated active geothermal system, have been also carried out. The whole dataset has been uploaded and analyzed in a dedicated geodatabase and 2D gravimetric modeling has been performed. Subsequently, the 3D structural model of the area, mainly based on the 3D resistivity and gravity models properly calibrated with wells data, has been shaped and utilized as the basis of the 3D thermofluid-dynamic model of the existing reservoir. Hydrothermal circulation has been modelled using the THOUGH2 code (simulation of multi-dimensional, multiphase and multi component flow and heat transport in porous and fractured media), a useful tool for the assessment of the geothermal potential and for the prediction of the evolution of the system. This integrated approach contributes to substantially reduce the mining risk in deep geothermal exploration, leading to the building of a better-defined conceptual model of the geothermal reservoir towards a more precise resource detection and assessment for the most appropriate and sustainable exploitation of the field

    Salivary testosterone and cortisol levels to assess conditioning training program in rugby union players

    Aim. To prevent overtraining and to ensure that the athletic training program will result in performance improvements, regular performance tests as well as heart rate evaluation and questionnaires are necessary. However none of them are routinely performed on regular basis mainly for their interference with the session training thus leading to the risk of an overreaching which in turn may result in an overtraining. In this study we evaluate the possibility to monitor the development of the training program of high level rugby players by a simple, non invasive and fast salivary testosterone and cortisol measure. Methods. Testosterone and cortisol samplings were performed in several moments of the pre-season training program. Measures were correlated with anthropometric evaluations performed by bioelectrical impedance analysis before and after the training program. To assess a potential role of genetic, athletes were also genotyped for ACTN3 and ACE gene polymorphisms. Results. Data from this study show that hormone salivary measures correlate with changes in anthropometric values during the training program. No correlations were observed between genotypes and variations in hormone levels; however, ACE genotype correlates with the different team position of rugby players. Conclusion. Hormone salivary measures represent a valid, fast , non-invasive and stress-free system of monitoring the athletic preparation
