    <p>Abstract</p> <p>Background</p> <p>Intestinal cell kinase (ICK; GeneID 22858) is a conserved MAPK and CDK-like kinase that is widely expressed in human tissues. Data from the Cancer Genome Anatomy Project indicated ICK mRNA is increased in cancer, and that its expression correlated with expression of mRNA for an uncharacterized F-box protein, FBX9 (GeneID: 26268). <it>ICK </it>and <it>FBX9 </it>genes are arranged head-to-head on opposite strands, with start sites for transcription separated by ~3.3 kb. We hypothesized ICK and FBX9 are potentially important genes in cancer controlled by a bidirectional promoter.</p> <p>Results</p> <p>We assessed promoter activity of the intergenic region in both orientations in cancer cell lines derived from breast (AU565, SKBR3), colon (HCT-15, KM12), and stomach (AGS) cancers, as well as in embryonic human kidney (HEK293T) cells. The intergenic segment was active in both orientations in all of these lines, and ICK promoter activity was greater than FBX9 promoter activity. Results from deletions and truncations defined a minimal promoter for ICK, and revealed that repressors and enhancers differentially regulate ICK versus FBX9 promoter activity. The ICK promoter contains consensus motifs for several FOX-family transcription factors that align when mouse and human are compared using EMBOSS. FOXA1 and FOXA2 increase luciferase activity of a minimal promoter 10-20 fold in HEK293T cells. Consensus sites for TCF7L2 (TCF4) (Gene Id: 6934) are also present in both mouse and human. The expression of β-catenin increased activity of the minimal promoter ~10 fold. ICK reference mRNAs (NM_014920.3, NM_016513) are expressed in low copy number and increased in some breast cancers, using a ten base tag 5'-TCAACCTTAT-3' specific for both ICK transcripts.</p> <p>Conclusion</p> <p><it>ICK </it>and <it>FBX9 </it>are divergently transcribed from a bidirectional promoter that is GC-rich and contains a CpG island. A minimal promoter for <it>ICK </it>contains functional sites for β-cateinin/TCF7L2 and FOXA. These data are consistent with functions that have been proposed for ICK in development and in proliferation or survival of some breast and colon cancers.</p

    The Role of Ligand Steric Bulk in New Monovalent Aluminum Compounds

    The article of record as published may be located at https://doi.org/10.1021/acs.jpca.7b02075The tetrameric Al(I) cyclopentadienyl compound Al4Cp*4 (Cp* = C5Me5) is a prototypical low-valence Al compound, with delocalized bonding between four Al(I) atoms and η5 ligands bound to the cluster exterior. The synthesis of new [AlR]4 (R = C5Me4Pr, C5Me4iPr) tetramers is presented. Though these systems failed to crystallize, comparison of variable-temperature 27Al NMR data with density functional theory (DFT) calculations indicate that these are Al4R4 tetramers analogous to Al4Cp*4 but with increased ligand steric bulk. NMR, DFT, and Atoms in Molecules analyses show that these clusters are enthalpically more stable as tetramers than the Cp* variant, due in part to noncovalent interactions across the bulkier ligand groups. Thermochemistry calculations for the low-valence metal interactions were found to be extremely sensitive to the DFT methodology used; the M06-2X functional with a cc-pVTZ basis set is shown to provide very accurate values for the enthalpy of tetramerization and 27Al NMR shifts. This computational method is then used to predict geometrical structures, noncovalent ligand interactions, and monomer/tetramer equilibrium in solution for a series of Al(I) cyclopentadienyl compounds of varying steric bulk

    Two Small Planets Transiting HD 3167

    We report the discovery of two super-Earth-sized planets transiting the bright (V = 8.94, K = 7.07) nearby late G-dwarf HD 3167, using data collected by the K2 mission. The inner planet, HD 3167 b, has a radius of 1.6 R_e and an ultra-short orbital period of only 0.96 days. The outer planet, HD 3167 c, has a radius of 2.9 R_e and orbits its host star every 29.85 days. At a distance of just 45.8 +/- 2.2 pc, HD 3167 is one of the closest and brightest stars hosting multiple transiting planets, making HD 3167 b and c well suited for follow-up observations. The star is chromospherically inactive with low rotational line-broadening, ideal for radial velocity observations to measure the planets' masses. The outer planet is large enough that it likely has a thick gaseous envelope which could be studied via transmission spectroscopy. Planets transiting bright, nearby stars like HD 3167 are valuable objects to study leading up to the launch of the James Webb Space Telescope.Comment: Accepted by ApJL. 6 pages, 1 figure, 2 table

    Late-time evolution of the Yang-Mills field in the spherically symmetric gravitational collapse

    We investigate the late-time evolution of the Yang-Mills field in the self-gravitating backgrounds: Schwarzschild and Reissner-Nordstr\"om spacetimes. The late-time power-law tails develop in the three asymptotic regions: the future timelike infinity, the future null infinity and the black hole horizon. In these two backgrounds, however, the late-time evolution has quantitative and qualitative differences. In the Schwarzschild black hole background, the late-time tails of the Yang-Mills field are the same as those of the neutral massless scalar field with multipole moment l=1. The late-time evolution is dominated by the spacetime curvature. When the background is the Reissner-Nordstr\"om black hole, the late-time tails have not only a smaller power-law exponent, but also an oscillatory factor. The late-time evolution is dominated by the self-interacting term of the Yang-Mills field. The cause responsible for the differences is revealed.Comment: Revtex, 14 pages, no figure

    Black hole polarization and new entropy bounds

    Zaslavskii has suggested how to tighten Bekenstein's bound on entropy when the object is electrically charged. Recently Hod has provided a second tighter version of the bound applicable when the object is rotating. Here we derive Zaslavskii's optimized bound by considering the accretion of an ordinary charged object by a black hole. The force originating from the polarization of the black hole by a nearby charge is central to the derivation of the bound from the generalized second law. We also conjecture an entropy bound for charged rotating objects, a synthesis of Zaslavskii's and Hod's. On the basis of the no hair principle for black holes, we show that this last bound cannot be tightened further in a generic way by knowledge of ``global'' conserved charges, e.g., baryon number, which may be borne by the object.Comment: 21 pages, RevTex, Regularization of potential made clearer.     Education and articulation: Laclau and Mouffe’s radical democracy in school

    This paper outlines a theory of radical democratic education by addressing a key concept in Laclau and Mouffe’s Hegemony and Socialist Strategy: articulation. Through their concept of articulation, Laclau and Mouffe attempt to liberate Gramsci’s theory of hegemony from Marxist economism, and adapt it to a political sphere inhabited by a plurality of struggles and agents none of which is predominant. However, while for Gramsci the political process of hegemony formation has an explicit educational dimension, Laclau and Mouffe ignore this dimension altogether. My discussion starts with elaborating the concept of articulation and analysing it in terms of three dimensions: performance, connection and transformation. I then address the role of education in Gramsci’s politics, in which the figure of the intellectual is central, and argue that radical democratic education requires renouncing that figure. In the final section, I offer a theory of such education, in which both teacher and students articulate their political differences and identities
