    Synthesis of Peptaibolin, an antimicrobial peptide

    To tackle one of the biggest global health problems, the resistance of microorganisms to antibiotics, a collective effort in the search for more effective agents against bacteria was required. Peptides with antimicrobial activity have been raising much attention as a promising alternative for antibiotics. Peptaibols, for instance, are a family of antimicrobial peptides (AMPs) with great biomedical potential, in which the Peptaibolin can be highlighted. This peptide has gained relevance due to its small amino acids content, only four, and its acetyl group and a phenylalaninol residue (Phol) at the N-terminal and C-terminal, respectively. Here, we report the synthesis of Peptaibolin through Solid Phase Peptide Synthesis assisted by Microwave heating (MW-SPPS) in a pre-loaded Phe-Wang resin. Starting from a loading of 0.51 mmol/g, two syntheses were made, using two different combinations of coupling reagents. The best option was DIC/Oxima, achieving a yield of 50.0%. Proton Nuclear Magnetic Resonance (1H-NMR) studies confirmed the peptide structure, while High Performance Liquid Chromatography (HPLC) verified the peptide purity. The peptide solubility was examined against several combinations of solvents. Peptaibolin was not soluble in water, only in organic solvents or in the combination of both. Antimicrobial testing was conducted using Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, and Pseudomonas aeruginosa.     A skin wound if not properly treated can result in a chronic wound susceptible to widespread infections, which can result in the patient's death. Currently, tissue engineering is described as an interdisciplinary field that combines principles of engineering, chemistry and biology to generate solutions that allow to repair, restore and/or improve the functions of injured tissues. In the same sense, the textile area addresses solutions based on polymeric fibers, produced from a wide range of polymers, which allow the generation of structures with a large surface area, porosity and mechanical resistance that can be used as bioactive dressings that promote a healing and efficient antimicrobial activity. This research work focused on the synthesis of Tiger 17, through microwave-assisted solid-phase synthesis methodologies, and Tiger 17 commercially obtained, respective structural characterization and evaluation of the antimicrobial capacity. Simultaneously, nanofibrous polymer matrices were produced using the electrospinning technique with the aim of immobilizing the developed biomolecule and thus creating potential vehicles for a local and sustainable antimicrobial action (controlled release). In order to verify its physical and chemical properties, advanced characterization techniques were used: proton nuclear magnetic resonance, high performance liquid chromatography, optical microscopy, scanning electron microscopy, fourier transform infrared spectroscopy–attenuated total reflectance, thermogravimetry, differential scanning calorimetry, contact angle and surface energy and determination of porosity and hydration.Düzgün tedavi edilmeyen bir cilt yarası, hastanın ölümüyle sonuçlanabilecek yaygın enfeksiyonlara duyarlı kronik bir yaraya neden olabilir. Günümüzde doku mühendisliği, hasarlı dokuların işlevlerini onarmaya, eski haline getirmeye ve/veya iyileştirmeye olanak tanıyan çözümler üretmek için mühendislik, kimya ve biyolojinin ilkelerini birleştiren disiplinler arası bir alan olarak tanımlanmaktadır. Bu kapsamda, tekstil, iyileşmeyi ve etkili antimikrobiyal aktiviteyi teşvik eden biyoaktif pansuman olarak kullanılabilecek, geniş yüzey alanlı, gözenekli ve mekanik dirençli yapıların oluşturulmasına izin veren, geniş bir polimer yelpazesinden üretilen liflere dayalı çözümler sunmaktadır. Bu araştırma çalışması, mikrodalga destekli katı faz sentez metodolojileri aracılığıyla Tiger 17'nin sentezine ve ticari olarak temin edilen Tiger 17 ile birlikte yapısal karakterizasyonunun ve antimikrobiyal kapasitenin değerlendirilmesine odaklanmıştır. Aynı zamanda, geliştirilen biyomolekülü hareketsiz hale getirmek ve böylece lokal ve sürdürülebilir bir antimikrobiyal etki (kontrollü salım) için potansiyel çözümler yaratmak amacıyla elektrik alan lif üretim tekniği kullanılarak nanolifli polimer matrisler üretildi. Yapının fiziksel ve kimyasal özelliklerini doğrulamak için, proton nükleer manyetik rezonans, yüksek performanslı sıvı kromatografisi, optik mikroskopi, taramalı elektron mikroskobu, fourier dönüşümü kızılötesi spektroskopisi-zayıflatılmış toplam yansıma, termogravimetri, diferansiyel taramalı kalorimetri, temas açısı, yüzey enerjisi, gözeneklilik ve su tutma ölçümleri gibi ileri karakterizasyon teknikleri kullanıldı

    Payment for water-ecosystem services monitoring in Brazil.

    The reduction of tropical forests has generated a loss of ecosystem services across the globe. In Brazil, essential biomes related to water provision (such as the Atlantic Forest and Savanna) have been degraded, compromising water-ecosystem services. Payment for water-ecosystem services (water PES) has been implemented as a tool to stimulate changes in the use and management of these areas. Many water PES projects have emerged in Brazil using forest restoration, aiming to improve water ecosystem services. In this context, this study identified the types of monitoring carried out in Brazilian water PES projects, to include their main characteristics and gaps. Five Brazilian projects were selected for analysis as case studies. Interviews were then conducted with stakeholders to get current data on their monitoring practices. The data from the literature review, case study approach, and interviews were analyzed from the perspective of monitoring guides recommendations. Different aspects were analyzed, such as objectives, institutional arrangements, type of monitoring, indicators, and frequency of monitoring. The study indicates that there is a lack of standardized methods, making it difficult to specify the results of the implemented actions. The central gap is related to benefit monitoring. It is necessary to establish a holistic monitoring system, dealing with the ecosystem as a complex socio-ecological system. Some perspectives to solve the problems were proposed. The results of this work may help not only improve the current and future PES schemes in Brazil but also in other countries, especially developing ones, where vulnerable populations depend upon them

    Ao avaliar os projetos de pagamentos por serviços ambientais (PSA) no Brasil, observam-se dificuldades recorrentes relacionadas à falta de consolidação das metodologias para a seleção de áreas prioritárias e, sobretudo, para o monitoramento das áreas de prestação dos serviços. A estratégia de pagamentos por serviços ambientais é cada vez mais presente nas políticas ambientais, devido ao reconhecimento da necessidade de novos mecanismos de estímulo à conservação e à manutenção de serviços essenciais à vida, como a provisão de água. A partir da análise das experiências vivenciadas em projetos de Pagamento por Serviços Ambientais (PSA) relacionados à água de alguns estados brasileiros, o trabalho apresenta um panorama destas experiências, de maneira a identificar as tendências, os obstáculos enfrentados e, sobretudo, os aspectos relevantes das metodologias de monitoramento aplicadas

    Analysis of the dynamics of a spacecraft in the vicinity of an asteroid binary system with equal masses

    In this work, we performed a dynamical analysis of a spacecraft around a nearly equal-mass binary near-Earth asteroid with application to the asteroid 2017 YE5, which is also a possible dormant Jupiter-family comet. Thus, we investigated the motion of a particle around this binary system using the circular restricted three-body problem. We calculated the locations of the Lagrangian points of the system and their Jacobi constant. Through numerical simulations, using the Poincar\'e Surface of Sections, it was possible to find several prograde and retrograde periodic orbits around each binary system's primary, some exhibiting significantly-sized higher-order behavior. We also calculated the stability of these orbits. After finding the periodic orbits, we investigated the influence of solar radiation pressure on these orbits. For this analysis, we considered that the area-to-mass ratio equals 0.01 and 0.1. We also performed a spacecraft lifetime analysis considering the physical and orbital characteristics of the 2017YE5 system and investigated the behavior of a spacecraft in the vicinity of this system. We analyzed direct and retrograde orbits for different values of Jacobi's constant. This study investigated orbits that survive for at least six months, not colliding or escaping the system during that time. We also analyze the initial conditions that cause the spacecraft to collide with M1M_1 or M2M_2, or escape from the system. In this work, we take into account the gravitational forces of the binary asteroid system and the solar radiation pressure (SRP). Finally, we calculated optimal bi-impulsive orbital maneuvers between the collinear Lagrangian points.     Padrões nutricionais para lavouras arrozeiras irrigadas por inundação pelos métodos da CND e Chance Matemática.

    Para o arroz irrigado, poucos trabalhos utilizam métodos de diagnose foliar desenvolvidos para as condições locais de clima, solo ou cultivares. O objetivo deste trabalho foi avaliar os métodos da Diagnose da Composição Nutricional e da Chance Matemática na definição dos padrões nutricionais de lavouras arrozeiras do Estado do Rio Grande do Sul. Resultados de produtividade de grãos e teores foliares de N, P, K, Ca, Mg, S, B, Cu, Fe, Mn, Zn e Mo de 356 lavouras arrozeiras cultivadas sob sistema de irrigação por inundação foram utilizados para a determinação das faixas de suficiência calculadas pelo método da Chance Matemática. As faixas de suficiência foram comparadas com valores críticos propostos pela literatura e com o intervalo de confiança do teor médio dos nutrientes em lavouras consideradas nutricionalmente equilibradas, identificadas pelo método Diagnose da Composição Nutricional. Observou-se pouca concordância entre os valores das faixas de suficiência indicados pelos métodos da Chance Matemática e da Diagnose da Composição Nutricional e os respectivos valores indicados na literatura. A faixa de teores foliares adequados, consistentes com maior produtividade média das lavouras arrozeiras, foi indicada ser de 23 a 28 g kg-1 para N; 11 a 14 g kg-1 para K; 1,4 a 2,0 g kg-1 para S; 6 a 12 mg kg-1 para B; e 70 a 200 mg kg-1 para Fe. Para os teores foliares de P, Ca, Mg, B, Cu, Mn e Zn e Mo nenhuma das faixas adequadas testadas indicou capacidade para distinguir as lavouras arrozeiras quanto à produtividade média
