    ETHANOL PRODUCTION FROM A MEMBRANE PURIFIED HEMICELLULOSIC HYDROLYSATE DERIVED FROM SUGAR MAPLE BY PICHIA STIPITIS NRRL Y-7124

    In an effort to devise inexpensive and sustainable production of ethanol fuel, experiments were conducted to establish conditions for Pichia stipitis NRRL Y-7124 to ferment a membrane treated wood hydrolysate derived from sugar maple to produce ethanol. The degree of aeration required to effectively utilize xylose, produce ethanol, and minimize xylitol formation as well as the optimal hydrolysate concentration were the conditions examined. P. stipitis produced the highest concentrations of ethanol in shake flasks at 150 rpm (14.3 g/L in 71 h), and 50% hydrolysate maximized ethanol yield (12.4 g/L in 51.5 h). In the 50% hydrolysate cultures, P. stipitis produced ethanol at a rate of 0.24 g/Lh with a yield of 0.41 g ethanol/g wood-derived carbohydrate

    Ecosystem engineers stabilize sand bank systems: <i> Owenia fusiformis</i> aggregations as ecologically important microhabitat

    Ecosystem engineers modify the physical environment and have profound effects on ecosystem functioning and on local biodiversity. Yet, in soft bottom marine environments, they have rarely been included in hydrodynamic studies or in management strategies. The lack of quantified stabilization potential and ecological impact are, respectively, the main reasons for not including ecosystem engineers. The present study evaluates the ecosystem engineering capacity of the tube dwelling polychaete Owenia fusiformis. The ecological implications of aggregations of this species are investigated using a long term dataset (1994- 2006). Results show that its presence has significant implications for species richness and species density. These aggregations are further investigated with different remote sensing tools. Owenia fusiformis is able to stabilize sand dunes that normally migrate 12 m a year. Specific biological characteristics explain the stabilizing effects in a highly dynamic environment. Our results confirm the need to pay attention to biota in sediment transport modelling. We conclude that O. fusiformis creates an important marine system which can be investigated with sensing techniques. These techniques can therefore be used to visualize hotspots of biodiversity, to quantify their biogeomorphological impacts and to underpin ecosystems based management in the marine environment

The symmetry-point approach which maximizes simultaneously both types of correct classification is one way to determine an optimal cut-off point. In this article, we study methods for constructing confidence intervals independently for the symmetry point and its corresponding sensitivity, as well as respective joint nonparametric confidence regions. We illustrate using data on the generation of antibodies elicited two weeks post-injection after the second dose of the Pfizer/BioNTech vaccine in adult healthcare workers

    Inclusion of multiple high‐risk histopathological criteria improves the prediction of adjuvant chemotherapy efficacy in lung adenocarcinoma

    AIMS: The decision to consider adjuvant chemotherapy (AC) for non-small cell lung cancer is currently governed by clinical stage. This study aims to assess other routinely collected pathological variables related to metastasis and survival for their ability to predict the efficacy of AC in lung adenocarcinoma. METHODS AND RESULTS: A retrospective single-centre series of 620 resected lung non-mucinous adenocarcinoma cases from 2005-2015 was used. Digital images of all slides were subjected to central review, and data on tumour histopathology, AC treatment and patient survival were compiled. A statistical case matching approach was used to counter selection bias. Several high-risk pathological criteria predict both pathological nodal involvement and early death: positive vascular invasion status (VI+) (HR=2.10 P<0.001), positive visceral pleural invasion status (VPI+) (HR=2.16 P<0.001), and solid/micropapillary-predominant WHO tumour type (SPA/MPPA) (HR=3.29 P<0.001). Crucially, these criteria also identify patient groups benefiting from AC (VI+ HR=0.69 P=0.167, VPI+ HR=0.44 P=0.004, SPA/MPPA HR=0.36 P=0.006). Cases showing VI+/VPI+/SPA/MPPA histology in the absence of AC stage criteria were common (170 of 620 total), and 8 had actually received AC. This group showed much better outcomes than equivalent untreated cases in matched analysis (3-year OS 100.0% vs 31.3%). Inclusion of patients with VI+/VPI+/SPA/MPPA histology would increase AC-eligible patients from 51.0% to 84.0% of non-mucinous tumours in our cohort. CONCLUSIONS: Our data provide preliminary evidence that the consideration of AC in patients with additional high-risk pathological indicators may significantly improve outcomes in operable lung adenocarcinoma, and that AC may be currently underused

    Cameriere’s European formula for age estimation: A study on the children in Bosnia and Herzegovina

    Introduction: A method for age estimation, based on measurements of projections of open apices and heights of developing permanentteeth on orthopantomograms (OPTs), was presented by Cameriere in 2006 and adopted European formula was presented in 2007. Aim: This cross-sectional study tested the accuracy of Cameriere’s European formula on a sample from the City of Sarajevo, Bosniaand Herzegovina. Materials and methods: A final sample of 560 OPTs of 305 girls and 255 boys aged 8 to 14 years was obtained. The sample was collected at the Department of Orthodontics, School of Dental Medicine at the University of Sarajevo (SFUNSA). Dental age was compared to chronological age and mean absolute error (MAE) was calculated. Intra-rater and inter-rater agreement of the evaluated variables were calculated. Results: The dental age was underestimated when compared to chronological age, precisely, mean underestimation was -0.14 years ingirls and -0.17 years in boys. The values of MAE were 0.62 years in girls and 0.56 years in boys. The greatest error was found for the 14-year old group; DA was -1.04 years and -0.70 years in girls and boys respectively. Conclusion: Our results showed that Cameriere’s European formula might be a useful tool for age estimation in children from Bosnia and Herzegovina under the age of 14 years

    Metabolomic, hormonal and physiological responses to hypoglycemia versus euglycemia during exercise in adults with type 1 diabetes

    Introduction This study sought to compare the metabolomic, hormonal and physiological responses to hypoglycemia versus euglycemia during exercise in adults with type 1 diabetes (T1D).Research design and methods Thirteen individuals with T1D (hemoglobin; 7.0%±1.3% (52.6±13.9 mmol/mol), age; 36±15 years, duration diabetes; 15±12 years) performed a maximum of 45 min submaximal exercise (60%±6% V̇O2max). Retrospectively identified exercise sessions that ended in hypoglycemia ((HypoEx) blood glucose (BG)≀3.9 mmol/L) were compared against a participant-matched euglycemic condition ((EuEx) BG≄4.0, BG≀10.0 mmol/L). Samples were compared for detailed physiological and hormonal parameters as well as metabolically profiled via large scale targeted ultra-high-performance liquid chromatography coupled to tandem mass spectrometry. Data were assessed using univariate and multivariate analysis techniques with false discovery rate adjustment. Significant results were considered at p≀0.05.Results Cardiorespiratory and counterregulatory hormone responses, whole-body fuel use and perception of fatigue during exercise were similar under conditions of hypoglycemia and euglycemia (BG 3.5±0.3 vs 5.8±1.1 mmol/L, respectively p<0.001). HypoEx was associated with greater adenosine salvage pathway activity (5’-methylthioadenosine, p=0.023 and higher cysteine and methionine metabolism), increased utilization of glucogenic amino acids (glutamine, p=0.021, alanine, aspartate and glutamate metabolism and homoserine/threonine, p=0.045) and evidence of enhanced ÎČ-oxidation (lower carnitine p<0.001, higher long-chain acylcarnitines).Conclusions Exposure to acute hypoglycemia during exercise potentiates alterations in subclinical indices of metabolic stress at the level of the metabolome. However, the physiological responses induced by dynamic physical exercise may mask the symptomatic recognition of mild hypoglycemia during exercise in people with T1D, a potential clinical safety concern that reinforces the need for diligent glucose management

    Adrenal, thyroid and gonadal axes are affected at high altitude

    Humans cannot live at very high altitude for reasons, which are not completely understood. Since these reasons are not restricted to cardiorespiratory changes alone, changes in the endocrine system might also be involved. Therefore, hormonal changes during prolonged hypobaric hypoxia were comprehensively assessed to determine effects of altitude and hypoxia on stress, thyroid and gonadal hypothalamus–pituitary hormone axes. Twenty-one male and 19 female participants were examined repetitively during a high-altitude expedition. Cortisol, prolactin, thyroid-stimulating hormone (TSH), fT4 and fT3 and in males follicle-stimulating hormone (FSH), luteinizing hormone (LH) and total testosterone were analysed as well as parameters of hypoxemia, such as SaO2 and paO2 at 550 m (baseline) (n = 40), during ascent at 4844 m (n = 38), 6022 m (n = 31) and 7050 m (n = 13), at 4844 m (n = 29) after acclimatization and after the expedition (n = 38). Correlation analysis of hormone concentrations with oxygen parameters and with altitude revealed statistical association in most cases only with altitude. Adrenal, thyroid and gonadal axes were affected by increasing altitude. Adrenal axis and prolactin were first supressed at 4844 m and then activated with increasing altitude; thyroid and gonadal axes were directly activated or suppressed respectively with increasing altitude. Acclimatisation at 4844 m led to normalization of adrenal and gonadal but not of thyroid axes. In conclusion, acclimatization partly leads to a normalization of the adrenal, thyroid and gonadal axes at around 5000 m. However, at higher altitude, endocrine dysregulation is pronounced and might contribute to the physical degradation found at high altitude

    Bayesian Bootstrap Inference for the ROC Surface

    Accurate diagnosis of disease is of great importance in clinical practice and medical research. The receiver operating characteristic (ROC) surface is a popular tool for evaluating the discriminatory ability of continuous diagnostic test outcomes when there exist three ordered disease classes (e.g., no disease, mild disease, advanced disease). We propose the Bayesian bootstrap, a fully nonparametric method, for conducting inference about the ROC surface and its functionals, such as the volume under the surface. The proposed method is based on a simple, yet interesting, representation of the ROC surface in terms of placement variables. Results from a simulation study demonstrate the ability of our method to successfully recover the true ROC surface and to produce valid inferences in a variety of complex scenarios. An application to data from the Trail Making Test to assess cognitive impairment in Parkinson's disease patients is provided

    Computer-aided assessment of diagnostic images for epidemiological research

    <p>Abstract</p> <p>Background</p> <p>Diagnostic images are often assessed for clinical outcomes using subjective methods, which are limited by the skill of the reviewer. Computer-aided diagnosis (CAD) algorithms that assist reviewers in their decisions concerning outcomes have been developed to increase sensitivity and specificity in the clinical setting. However, these systems have not been well utilized in research settings to improve the measurement of clinical endpoints. Reductions in bias through their use could have important implications for etiologic research.</p> <p>Methods</p> <p>Using the example of cortical cataract detection, we developed an algorithm for assisting a reviewer in evaluating digital images for the presence and severity of lesions. Available image processing and statistical methods that were easily implementable were used as the basis for the CAD algorithm. The performance of the system was compared to the subjective assessment of five reviewers using 60 simulated images. Cortical cataract severity scores from 0 to 16 were assigned to the images by the reviewers and the CAD system, with each image assessed twice to obtain a measure of variability. Image characteristics that affected reviewer bias were also assessed by systematically varying the appearance of the simulated images.</p> <p>Results</p> <p>The algorithm yielded severity scores with smaller bias on images where cataract severity was mild to moderate (approximately ≀ 6/16<sup><it>ths</it></sup>). On high severity images, the bias of the CAD system exceeded that of the reviewers. The variability of the CAD system was zero on repeated images but ranged from 0.48 to 1.22 for the reviewers. The direction and magnitude of the bias exhibited by the reviewers was a function of the number of cataract opacities, the shape and the contrast of the lesions in the simulated images.</p> <p>Conclusion</p> <p>CAD systems are feasible to implement with available software and can be valuable when medical images contain exposure or outcome information for etiologic research. Our results indicate that such systems have the potential to decrease bias and discriminate very small changes in disease severity. Simulated images are a tool that can be used to assess performance of a CAD system when a gold standard is not available.</p
