Szilárd bázis felületén végbemenő reakciók tanulmányozása szerves foszforvegyületek körében = Reactions of organophosphorus substrates on the solid bases
Egyszerű mĂłdszert dolgoztunk ki vegyes foszfátok előállĂtására oxo vegyĂĽletekbĹ‘l Ă©s H-foszfonátokbĂłl (dialkil-foszfitokbĂłl) fázistranszfer katalitikus körĂĽlmĂ©nyek között (szilárd kálium-karbonát bázissal), továbbá diasztereo szelektĂv szintĂ©zisben α-hidroxi-foszfinátokat állĂtottunk elĹ‘ aldehidekbĹ‘l Ă©s H-foszfinátokbĂłl. α-Meziloxi-foszfonátok Ă©s dialkil-foszfitok Ăşj reakciĂłját ismertĂĽk fel Ă©s megoldást javasoltunk a foszfono – foszfát szerkezetű termĂ©k keletkezĂ©sĂ©nek mechanizmusára. Megvizsgáltuk savanhidridek keletkezĂ©sĂ©nek mechanizmusát szilárd kálium-karbonát felĂĽletĂ©n. Javaslatunk szerint a kĂĽlönbözĹ‘ savkloridok (foszfor-, szulfon-, karbon-) az un. „interphase”-ben reaktĂv vegyes anhidrid intermediert kĂ©peznek a karbonáttal, ami a megfelelĹ‘ foszforsav, szulfonsav ill. karbonsav anhidriddĂ© alakul tovább. α-Amino-foszfinátok enantio szelektĂv szintĂ©zisĂ©t valĂłsĂtottuk meg aldehidekbĹ‘l, királis aminokbĂłl Ă©s H-foszfinátokbĂłl szilárd bázisok felĂĽlettĂ©n (alumĂnium-oxid, fluoridok) mikrohullámĂş gerjesztĂ©ssel. | A simple method was developed for the synthesis of mixed phosphonates starting from oxo compounds and dialkyl phosphites in phase transfer catalytic circumstances (on solid potassium carbonate), and α-hydroxy phosphonates were obtained in diastereoselective synthesis starting from aldehydes and H phosphinates by the same way. A new reaction of α-meziloxy phosphonates and dialkyl phosphites was recognized, and a mechanism was assumed for the formation of the phosphono phosphate type product. We investigated the mechanism of the formation of acid anhydrides on the surface of solid potassium carbonate. According to our results, various sorts of acid chlorides (phosphoric-, sulfonic-, carboxylic-) form a mixed anhydride type intermediate in the „interphase”, stabilization of which result in the anhydrides of phosphoric-, sulfonic-, carboxylic acids. Az amino-foszfon-, Ă©s foszfinsavak az aminosavak analogonjainak tekinthetĹ‘k, ezĂ©rt mind önmagukban, mind peptidláncba Ă©pĂtve számos Ă©rdekes felhasználási terĂĽlettel rendelkeznek. SzintĂ©zisĂĽkre több eljárás ismeretes, jĂłl használhatĂł sztereoszelektĂv mĂłdszer azonban kevĂ©s van (amino-foszfinsavakra kutatásunk kezdetĂ©ig nem is volt). A lakton-gyűrű nyitásával kapott savamid, ill. savazid-származĂ©kok egyrĂ©szt a cĂ©lvegyĂĽletekkĂ© alakĂthatĂłk, másrĂ©szt enzimes kinetikus reszolválással enantiomerekre bonthatĂłk. | Amino phosphonates and phosphinates as the phosphorus analogues of naturally occurring α-amino acids are finding increasing interests as building blocks of phosphopeptides, and as interesting model compounds in enzyme research. The preparation of the single enantiomers of these compounds still remains partly unsolved and needs new synthetic methods. In this project, a catalytic asymmetric Michael addition was successfully applied in the synthesis of chiral aminophosphonates. As chiral catalysts sugar containing crown ethers and TADDOLs were used with medium and good selectivity, respectively. We were the first to succeed in the enantioselective synthesis of substituted aminophosphinic acids from chiral imines and alkyl/aryl phosphinates. In the case we used enantiopure sulfinimines several unexpected by products were isolated and identified as a result of an interesting side reaction. α-Amino cyclpropane phosphinic acids as conformationally restricted amino acids form a newly synthesized subgroup. As the first step, we developed a diastereoselective cyclopropanation reaction providing cyclopropane lactones, then acid amides and azides were obtained by opening the lacton ring, following by enzyme catalyzed kinetic resolution resulting in useful intermediates as single isomers

A projekt cĂ©lja aminofoszfonátok Ă©s foszfinátok katalitikus sztereoszelektĂv szintĂ©zise volt. KĂĽlönbözĹ‘ ?????Ă©s ?-aminofoszfonát prekurzorokbĂłl kiindulva C-C kötĂ©s kialakĂtásával állĂtottunk elĹ‘ egyik enantiomerben gazdagabb ??C-atomon szubsztituált foszfonátokat. MegállapĂtottuk, hogy a Michael-addiciĂłban alkalmazhatĂł királis katalizátor tĂpusa a prekurzor foszfonát ??helyzetű CH-csoportjának savasságátĂłl fĂĽgg. A legkevĂ©sbĂ© savas vĂ©dett foszfoglicin származĂ©kbĂłl D-glukĂłz-, ill. R-BINOL alapĂş N-tartalmĂş koronaĂ©teekkel erĹ‘s bázis jelenlĂ©tĂ©ben igen magas enantiomer-, Ă©s diasztereomer felesleget sikerĂĽlt elĂ©rni akril-nitrilek Michael-addiciĂłjában. Ciano-metilfoszfonátot, mint viszonylag könnyen deprotonálhatĂł ?-aminofoszfonsav prekurzort organokatalizátorokkal kalkonok Michael-addiciĂłjában tudtuk hasznosĂtani. KatalizátorkĂ©nt cinkona-, Ă©s TADDOL-alapĂş tiokarbamid származĂ©kokat alkalmaztunk. Az ??nitro-etilfoszfonát, mint ??helyzetben kvaterner C-atomot tartalmazĂł aminofoszfonsav származĂ©k prekurzora organokatalizált Michael-addiciĂłban aril-akrilátokkal magas enantioszelektivitással eredmĂ©nyezte a megfelelĹ‘ adduktot, melyet katalitikus hidrogĂ©nezĂ©ssel aminofoszfonáttá alakĂtottunk. Katalitikus P-C kötĂ©s kialakĂtásával dialkil-foszfitokbĂłl Ă©s iminekbĹ‘l is előállĂtottunk közel enantiomertiszta ??aminofoszfonát-származĂ©kokat TADDOL-ok titán-komplexeit alkalmazva Lewis-sav katalizátorkĂ©nt. TöbblĂ©pĂ©ses szintĂ©zisben - az irodalomban eddig nem szereplĹ‘ vegyĂĽletcsalád egyik kĂ©pviselĹ‘jĂ©t - amino-ciklopropán-foszfinsavat állĂtottunk elĹ‘. Az egyik intermedierre enzimatikus kinetikus reszolválási mĂłdszert dolgoztunk ki. | The aim of the project was the catalytic stereoselective synthesis of aminophosphonates and aminophosphinates. Several ?-, and ?-aminophosphonate precursors were used in C-C-bond forming reactions resulting in nonracemic ?-substituted aminophosphonates. It was established that the character of the catalyst suitable in the Michael addition was strongly influenced by the acidity of the ?- CH-group of the precursor. The least acidic protected phosphoglycine derivative could be applied in the Michael addition of acrilonitriles by using strong base and D-glucose/R-BINOL-based crown ethers as catalysts resulting in the adducts in high enantio-, and diastereoselectivity. Cyanomethyl phosphonate as an easily deprotonable ?-aminophosphonate precursor could be utilized in an organocatalytic Michael addition of chalchones. As catalysts chincona-, and TADDOL-based thioureas were applied. Nearly the single isomer of ?-aminophosphonates having quaternary carbon atom next to the phosphorus were obtained from ?-nitro ethylphosphonate by means of aryl acrilate in an organocatalysed Michael addition followed by catalytic hydrogenation. Nearly the single isomer of ?-aminophosphonates were also synthesised from dialkyl phosphites and imines in P-C bond formation reaction using TADDOL-Ti complex as catalyst. A substituted aminocyclopropanephosphinic acid as the first member of a new family of compound was synthesised in a multistep reaction. An enzymatic kinetic resolution was succesfully applied on one of the intermediates
Study of the diastereoisomers formed between (N-alkyl)-pipecolic acid-anilides and 2R,3R-tartaric acid or O,O′-dibenzoyl-2R,3R-tartaric acid.
It was found that during the optical resolution of (N-alkyl)-pipecolic acid-anilides by 2R,3R- tartaric acid and O,O′-dibenzoyl-2R,3R-tartaric acid that the precipitated diastereoisomer was not the salt but a diastereoisomeric complex in 8 cases from 13. The results indicate that tartaric acids may be used as general resolving agents for optical resolution of racemates even having no basic group
