Efficacy of an Immune Modulator in Experimental Chlamydia trachomatis Infection of the Female Genital Tract
Objective. The aim of this study was to determine if vaginal application of the immune response modifier imiquimod (Aldara cream, 3M Pharmaceuticals, St Paul, Minn) would alter the course and/or outcome of female genital tract infection with a human isolate of Chlamydia trachomatis in a murine model. Methods. Groups of CF-1 mice were treated with Aldara on three different schedules: (1) ongoing beginning 5 days prior to and continuing through day 5 of infection; (2) a single prophylactic dose 2 hours prior to infection; and (3) therapeutic from day 4 to day 14 of infection. Mice were infected vaginally with a serovar D strain of C trachomatis, and monitored by culture to determine the level of shedding and duration of infection. Results. We observed a significant reduction in both duration of infection and the level of shedding during the acute phase in mice treated on an ongoing basis commencing 5 days prior to infection. There was no effect with respect to the other regimens. Conclusion. These results demonstrate that ongoing Aldara treatment has efficacy and may enhance local innate immunity which reduces the duration of subsequent infection with human isolates of C trachomatis in a murine model of female genital tract infection
Stock Identification of summer Flounder (Paralichthis dentatus) in the Southern Mid-Atlantic Bight
A total of 12, 339 summer flounder were tagged from Virginia waters during 1987-89. A total of 874 were recaptured for an overall return rate of 7 . 1%. Most of the returns ( 48.5%) were from Virginia waters, or areas to the south. A smaller number ( 21.6%) were from areas north and offshore of Virginia. Another 29. 9% were recaptured and returned with inadequate location data. Examining only the returns with adequate location data, yielded a separation of 69.2% and 30.8% between the groups. No differences were noted in the sizes at tagging between these groups. Tagged flounder held at VIMS exhibited no behavioral differences from untagged fish. No differences in growth and mortality were noted in these fish. The sex ratio of males to females was 1:1.16. Male summer flounder reached 50% maturity at approximately 280 mm, while females reached 50% maturity at about 330 mm. A total of 1040 flounder were successfully aged. The population was dominated by young fish ( 0- 2 years old). The compression of age structure i s indicative of a population being heavily overfished

Ensuring access to drinking water and wastewater service is a nationwide policy challenge. Across the United States access is increasingly insecure for many people and places. In this report we comply with scholarship and legal precedent that defines access to include access to residential in-home service, quality service that serves environmental and personal health,and affordable service.Water security is a term in this report used to describe the presence of structural, systemic, and institutional arrangements that ensure everyone has consistent access to drinking water and wastewater services. Water insecurity looks different in the humid east than in the arid west, different in the Midwest from the South, different between urban, suburban, or rural.However different water insecurity problems look at the local level, they are the result of similar institutional, systemic, and structural problems. This is a study of the what persistent water insecurity looks like in the service area of Detroit's drinking and wastewater system (DWSD) and specific places within that system, notably Detroit
Do host genetic traits in the bacterial sensing system play a role in the development of Chlamydia trachomatis-associated tubal pathology in subfertile women?
BACKGROUND: In women, Chlamydia (C.) trachomatis upper genital tract infection can cause distal tubal damage and occlusion, increasing the risk of tubal factor subfertility and ectopic pregnancy. Variations, like single nucleotide polymorphisms (SNPs), in immunologically important host genes are assumed to play a role in the course and outcome of a C. trachomatis infection. We studied whether genetic traits (carrying multiple SNPs in different genes) in the bacterial sensing system are associated with an aberrant immune response and subsequently with tubal pathology following a C. trachomatis infection. The genes studied all encode for pattern recognition receptors (PRRs) involved in sensing bacterial components. METHODS: Of 227 subfertile women, serum was available for C. trachomatis IgG antibody testing and genotyping (common versus rare allele) of the PRR genes TLR9, TLR4, CD14 and CARD15/NOD2. In all women, a laparoscopy was performed to assess the grade of tubal pathology. Tubal pathology was defined as extensive peri-adnexal adhesions and/or distal occlusion of at least one tube. RESULTS: Following a C. trachomatis infection (i.e. C. trachomatis IgG positive), subfertile women carrying two or more SNPs in C. trachomatis PRR genes were at increased risk of tubal pathology compared to women carrying less than two SNPs (73% vs 33% risk). The differences were not statistically significant (P = 0.15), but a trend was observed. CONCLUSION: Carrying multiple SNPs in C. trachomatis PRR genes tends to result in an aberrant immune response and a higher risk of tubal pathology following a C. trachomatis infection. Larger studies are needed to confirm our preliminary findings
Isotopic Signature of Massive, Buried Ice in Eastern Taylor Valley, Antarctica: Implications for Its Origin
The coastal regions of the McMurdo Dry Valleys, Antarctica, contain deposits of the Ross Sea Drift, sedimentary material left from the Ross Sea ice sheet from the advance of the West Antarctic ice sheet during the Last Glacial Maximum. Much of this deposit is ice-cored, but data on the stable isotopic composition of water from this ice, which may contain a valuable climate archive, are sparse or incomplete. Widespread thermokarstic ground subsidence in this “coastal thaw zone” of the McMurdo Dry Valleys suggests that these potential records are rapidly being lost due to the melting of ground ice and permafrost. We collected samples of massive buried ice from the Ross Sea Drift in eastern Taylor Valley for δ18O-H2O and δ2H-H2O and measured a broad range of values (δ18O = −27.7 to −37.3 ‰; δ2H = −210 to −295 ‰). These buried ice deposits do not show evidence of alteration through sublimation or evaporation, plot along the local meteoric water line, and have values that indicate ice deposition under a colder climate than present conditions. We propose that this ice was sourced from the Ross Sea ice sheet during the Last Glacial Maximum and contains a valuable and accessible climate record

Hypersaline “wet patches” in Taylor Valley, Antarctica
Spatially isolated patches of soil located in Taylor Valley, McMurdo Dry Valleys, Antarctica, are sites of elevated salt content and soil moisture. During Antarctic spring, in the absence of snow melt, visibly wet (reduced albedo) patches of soil are present at the surface. The soil pore fluids are hypersaline and have average water activity of 0.74 (the water activity of a solution determines the equilibrium vapor pressure of that solution), and are an order of magnitude more saline than average soils in the Dry Valleys. These salty soils are 3–5 times more water rich than average soils. Geochemical and meteorological analyses show that these wet patches are sites of direct vapor emplacement into soil pore fluids that may ultimately be sourced by the deliquescence of soil salts. These wet patches represent a non-precipitation, non-groundwater source for water into Antarctic permafrost
Weight loss for overweight and obese patients with prostate cancer: A study protocol of a randomised trial comparing clinic-based versus telehealth delivered exercise and nutrition intervention (the TelEX trial)
Introduction Obese men with prostate cancer have an increased risk of biochemical recurrence, metastatic disease and mortality. For those undergoing androgen deprivation therapy (ADT), substantial increases in fat mass are observed in the first year of treatment. Recently, we showed that a targeted supervised clinic-based exercise and nutrition intervention can result in a substantial reduction in fat mass with muscle mass preserved in ADT-treated patients. However, the intervention needs to be accessible to all patients and not just those who can access a supervised clinic-based programme. The purpose of this study was to evaluate the efficacy of telehealth delivered compared with supervised clinic-based delivered exercise and nutrition intervention in overweight/obese patients with prostate cancer. Methods and analysis A single-blinded, two-arm parallel group, non-inferiority randomised trial will be undertaken with 104 overweight/obese men with prostate cancer (body fat percentage ≥ 25%) randomly allocated in a ratio of 1:1 to a telehealth-delivered, virtually supervised exercise and nutrition programme or a clinic-based, face-to-face supervised exercise and nutrition programme. Exercise will consist of supervised resistance and aerobic exercise performed three times a week plus additional self-directed aerobic exercise performed 4 days/week for the first 6 months. Thereafter, for months 7-12, the programmes will be self-managed. The primary endpoint will be fat mass. Secondary endpoints include lean mass and abdominal aortic calcification, anthropometric measures and blood pressure assessment, objective measures of physical function and physical activity levels, patient-reported outcomes and blood markers. Measurements will be undertaken at baseline, 6 months (post intervention), and at 12 months of follow-up. Data will be analysed using intention-to-treat and per protocol approaches. Ethics and dissemination Ethics approval has been obtained from the Edith Cowan University Human Research Ethics Committee (ID: 2021-02157-GALVAO). Outcomes from the study will be published in academic journals and presented in scientific and consumer meetings. Trial registration number: ACTRN12621001312831
