    Isolation of Nuclei from Physarum flavicomum: Demonstration of Nuclear Cyclic Acid AMP Phosphodiesterase

    Cyclic AMP phosphodiesterase activity in the nucleus of the myxomycete Physarum flavicomum was demonstrated by cytochemical staining utilizing electron microscopy and by enzymatic assays with tritiated cyclic AMP as the substrate. Cytochemical staining showed Physarum\u27s plasmodial phosphodiesterase activity to be located in the nucleus, along the plasma membrane, in vesicles, and free in the cytoplasm. Nuclear phosphodiesterase, which may be cell cycle dependent, was primarily located in the nucleolus. Nuclei from three to five day old microplasmodial cultures were isolated by the method of Henney and Yee. Whole cells were collected through centrifugation and washed. Pellets were homogenized in a medium composed of 0.01 MTris-HC1 (pH 7.2 at 4 °C), 0.25 M sucrose, 0.01% Triton X-100, and 5mM CaC1₂. Nuclei were collected through double filtration and two 1.0 M sucrose density gradient centrifugations. After the nuclei were washed, microscopic examination revealed a purity of over 90%. Radioactive assays of the nuclear preparations demonstrated phosphodiesterase activity consistant with that indicated by cytochemical localization. The specific activity of the nuclear enzyme was 15 nMole of cyclic AMP hydrolyzed /min/mg. of protein

    Impact of the Spruce Budworm (Lepidoptera: Tortricidae) on the Ottawa and Hiawatha National Forests, 1978-1980

    The Michigan Impact Plot System was established during 1978 and 1979 to obtain a data base for quantifying the impact of the spruce budworm in the Ottawa and Hiawatha National Forests. The formulae used to estimate the mean, total, and associated standard errors of the various parameters at the national forest and forest district levels are presented. We present the 1978, 1979, and 1980 impact data for the following parameters; percent mortality, total dead volume. dead volume per ha, live volume per ha, defoliation ranking, frequency and extent of top-kill, and incidence of spruce budworm feeding on saplings and regenera- tion. Statistics from an annual inventory of 108 composite ground sampling units (CGSU) in 1978, and 136 CGSU\u27s in 1979 and 1980 provide a more precise estimate ofthe impact of the spruce budworm in Michigan\u27s Upper Peninsula than ha~ been available to date

    Extracellular Phosphodiesterase from the Growth Medium of the Myxomycete Physarum flavicomum

    The plasmodium of the myxomycete Physarum flavicomum secretes cyclic AMP phosphodiesterase into the medium. The extracellular enzyme had a pH optimum between 7 and 8 and a Km of about 500 μM cyclic AMP and was inhibited by theophylline, caffeine and 3-isobutyl-l-methyxanthine (MIX). A marked decrease of enzyme activity was noted in the presence of EDTA, suggesting the requirement of Mg+ + by the enzyme. Addition of Mg+ + and Ca + + stimulated the enzyme while Zn+ + , Co+ + , Pb+ + , Mn+ + , Fe + + + , Ni+ + , and Cu + + all inhibited phosphodiesterase activity. An interesting feature of this extracellular phosphodiesterase was its ability to retain full catalytic activity after prolonged exposure to elevated temperature

    The effect of caffeine mouth rinse on self-paced cycling performance

    The aim of the study was to determine whether caffeine mouth rinse would improve 30 min self-paced cycling trial. Twelve healthy active males (age 20.5±0.7 years, mass 87.4±18.3 kg) volunteered for the study. They attended the laboratory on 3 separate occasions performing a 30 min self-paced cycling trial. On one occasion water was given as a mouth rinse for 5 s (PLA), on another occasion a 6.4% maltodextrin (CHO) solution was given for 5 s and finally a caffeine solution (containing 32 mg of caffeine dissolved in 125 ml water; CAF) was given for 5 s. Distance cycled, heart rate, ratings of perceived exertion, cadence, speed and power output were recorded throughout all trials. Distance cycled during the CAF mouth rinse trial (16.2±2.8 km) was significantly greater compared to PLA trial (14.9±2.6 km). There was no difference between CHO and CAF trials (P=0.89). Cadence, power and velocity were significantly greater during the CAF trial compared to both PLA and CHO (P0.05). Caffeine mouth rinse improves 30 min cycling performance by allowing the participant to increase cadence, power and velocity without a concurrent increase in perceived exertion and heart rate

    Reflectance anisotropy of Gd5Si2Ge2 and Tb5Si2.2Ge1.8

    Reflectance difference (RD) spectra for the a–b plane of the single crystals of Gd5Si2Ge2and b–c planes of Gd5Si2Ge2 and Tb5Si2.2Ge1.8 were obtained in the photon energy range of 1.5–5.5 eV. Several peaks were observed for these crystals in the measured spectrum range. Similar features were observed in the RD spectra for the b–c planes ofGd5Si2Ge2 and Tb5Si2.2Ge1.8, while different features were observed for the a–b plane and b–c plane of Gd5Si2Ge2. The RD spectra for the crystals arise not only from the surface anisotropy but also from the bulk anisotropy due to the monoclinic structure of the bulk crystal

    Alterations in Mosquito Behaviour by Malaria Parasites: Potential Impact on Force of Infection

    A variety of studies have reported that malaria parasites alter the behaviour of mosquitoes. These behavioural alterations likely increase transmission because they reduce the risk of vector death during parasite development and increase biting after parasites become infectious. A mathematical model is used to investigate the potential impact of these behavioural alterations on the lifetime number of infectious bites delivered. The model is used to explore the importance of assumptions about the magnitude and distribution of mortality as well as the importance of extrinsic incubation period and gonotrophic cycle length. Additionally, the model is applied to four datasets taken from actual transmission settings. The impact of behavioural changes on the relative number of lifetime bites is highly dependent on assumptions about the distribution of mortality over the mosquito-feeding cycle. Even using fairly conservative estimates of these parameters and field collected data, the model outputs suggest that altered feeding could easily cause a doubling in the force of infection.Infection-iduced behavioural alterations have their greatest impact on the lifetime number of infectious bites in environments with high feeding-related adult mortality and many pre-infectious feeding cycles. Interventions that increase feeding-associated mortality are predicted to amplify the relative fitness benefits and hence enhance the strength of selection for behavioural alteration\u

    Physical properties of Heusler-like Fe2VAl

    A comprehensive characterization of the compound Fe2VAl was carried out. Samples grown by arc melting or the Bridgman method have Al and Fe deficiencies of up to 5 at. %. Czochralski-grown samples were Fe rich and Al deficient. X-ray diffraction implies appreciable antisite disorder in all of our samples. Fourier-transform infrared (FTIR) spectroscopy measurements showed that the carrier density and scattering time had little sample-to-sample variation or temperature dependence for near-stoichiometric samples. FTIR and dc resistivity suggest that the transport properties of Fe2VAl are influenced by both localized and delocalized carriers, with the former primarily responsible for the negative temperature coefficient of resistivity. Magnetization measurements reveal that near-stoichiometric samples have superparamagnetic clusters with at least two sizes of moments. We conclude that in Fe2VAl, antisite disorder causes significant modification to the semimetallic band structure proposed theoretically. With antisite disorder considered, we are now able to explain most of the physical properties of Fe2VAl. None of our data suggest heavy-fermion behavior in our samples
