    A Distance-Heuristic Tree Building Approach in Application Layer Multicast

    In the application layer multicast (ALM), clustering nearby nodes can effectively improve the multicast performance. However, it is difficult for the ALM solution to quickly and accurately position the newcomer, because group members have no direct knowledge of underlying network topology. Additionally, ALM delivery trees with different performances are built when group members join the group in different join sequences. To alleviate the above problems, this paper proposes a distance-heuristic tree building protocol (called DHTB). DHTB uses our proposed distance-constrained cluster model and close-member-first-receive (CF) rule. In the model, most nearby nodes are grouped into some distance-constrained clusters, with little cluster organization and maintenance overhead. The CF rule arranges or rearranges the locations of group members according to related distances, and effectively positions the newcomer with the help of on-demand landmarks. Both the distance-constrained cluster model and CF rule are distance-heuristic. Therefore DHTB can alleviate the join sequence problem, and build the ALM tree with desirable performance

    Proteomics-based analysis of differentially expressed proteins in the CXCR1-knockdown gastric carcinoma MKN45 cell line and its parental cell

    C-X-C chemokine receptor types 1 (CXCR1), a cell-surface G-protein-coupled receptor has been found to be associated with tumorigenesis, development, and progression of some tumors. Previously, we have found that CXCR1 overexpression is associated with late-stage gastric adenocarcinoma. We also have demonstrated that knockdown of CXCR1 could inhibit cell proliferation in vitro and in vivo. In this study, we compared the changes of protein expression profile between gastric carcinoma MKN45 cell line and CXCR1-knockdown MKN45 cell line by 2D electrophoresis. Among the 101 quantified proteins, 29 spots were significantly different, among which 13 were downregulated and 16 were up-regulated after CXCR1 knockdown. These proteins were further identified by mass spectrometry analysis. Among them, several up-regulated proteins such as hCG2020155, Keratin8, heterogeneous nuclear ribonucleoprotein C (C1/C2), and several downregulated proteins such as Sorcin, heat shock protein 27, serpin B6 isoform b, and heterogeneous nuclear ribonucleoprotein K were confirmed. These proteins are related to cell cycle, the transcription regulation, cell adherence, cellular metabolism, drug resistance, and so on. These results provide an additional support to the hypothesis that CXCR1 might play an important role in proliferation, invasion, metastasis, and prognosis, and drug resistance of gastric carcinoma

    A Clustering Scheme in Application Layer Multicast

    In application layer multicast (ALM), member hosts lack direct knowledge of underlying network topology, which brings some performance penalty. This paper investigates an effective way to rapidly obtain some related topology knowledge, i.e. getting topology hints from existing IP registered resources -- WHOIS database. We further propose a clustering scheme, which can be integrated into the existing ALM solutions. Our proposed scheme can cluster some nearby member hosts no matter when these hosts join the group. Therefore the scheme also solves the join sequences problem in some degree. We also present an application framework of the clustering scheme, and give an application example named HMTP-E that integrates the scheme into HMTP protocol. The experiment results show that the clustering scheme plays a positive role on improving the performance of existing ALM solutions

    Effect of Salt Tracer Dosages on the Mixing Process in the Water Model of a Single Snorkel Refining Furnace

    The improvement in mixing conditions in a vacuum refining unit plays an important role in enhancing the purity and decarburization of molten steel. Mixing time is an important index to evaluate the operation efficiency of a metallurgical reactor. However, in water models, the effect of salt tracer dosages on the measured mixing time in a vacuum reactor is not clear. In this study, a water model of a Single Snorkel Refining Furnace (SSRF) was established to study the effect of salt solution tracer dosages on the mixing time of monitor points. The experimental results show that, in some areas at the top of the ladle, the mixing time decreases first and then increases when increasing the tracer dosage. Numerical simulation results show that, when the tracer dosage increases, the tracer flows downwards at a higher pace from the vacuum chamber to the bottom of the ladle. This may compensate for the injection time interval of large dosage cases. However, the mass fraction of the KCl tracer at the right side of the bottom is the highest, which indicates that there may be a dead zone. For the dimensionless concentration time curves and a 99% mixing time, at the top of the vacuum chamber, the curve shifts to the right side and the mixing time decreases gradually with the increase in tracer dosage. At the bottom of the ladle, with the increase in tracer dosage, the peak value of the dimensionless concentration time curve is increased slightly. The mixing time of the bottom of the ladle decreases significantly with the increase in tracer dosage. However, in the dead zone, the mixing time will increase when the tracer dosage is large. At the top of the ladle, the effect of the tracer dosage is not obvious. The mixing time of the top of the ladle decreases first and then increases when increasing the tracer dosage. In addition, the mixing time of the top of the ladle is the shortest, which means that sampling at the top of the ladle in industrial production cannot represent the entire mixing state in the ladle

    Effect of drying methods on the texture properties and active ingredients in Longan flesh

    In order to determine the best drying method of longan flesh, the effects of different drying method (microwave drying, blast drying,vacuum freeze-drying) on the texture characteristics and active ingredients were compared. The results show that different drying methods will increase the hardness, cohesion and chewiness of longan flesh, and decrease the viscosity and elasticity to varying degrees. For active substances, the content of flavonoids and polyphenols is the largest under vacuum freeze-drying, followed by microwave drying and blast drying, but the loss of vitamin C is the opposite. On the whole, vacuum freeze-drying does not cause shrinkage of the flesh, and maintains good hardness, cohesion, chewiness and active ingredients. Comprehensive texture characteristics and changes in active ingredients, vacuum freeze drying is more suitable for drying and processing longan flesh

    The potential response of immune checkpoint blockade (ICB) in thymic neuroendocrine neoplasms (T-NEN) is largely unknown and full of great expectations. The expression of immune checkpoint molecules and immune infiltrates greatly determine the response to ICB. However, studies regarding the immune landscape in T-NEN are scarce. This work was aimed to characterize the immune landscape and its association with clinical characteristics in T-NEN. The expression of programmed cell death protein 1 (PD-1) and its ligand, programmed death ligand-1 (PD-L1), and the density of tumor-infiltrating lymphocytes (TILs), monocytes, and granulocytes were determined by immunohistochemical (IHC) staining on tumor tissues from T-NEN. Immune landscapes were delineated and correlated with clinicopathological factors. We found that T-NEN with increased immune cell infiltration and enhanced expression of PD-1/PD-L1 tended to have restricted tumor size and less metastases. A higher density of CD8+ TILs was associated with a significantly lower rate of bone metastasis. In addition, we presented three cases of T-NEN who progressed after multiple lines of therapies and received ICB for alternative treatment. ICB elicited durable partial responses with satisfactory safety in two patients with atypical carcinoid, but showed resistance in 1 patient with large cell neuroendocrine carcinoma. This innovative study delineated for the first time the heterogeneous immune landscape in T-NEN and identified CD8+ TILs as a potential marker to predict bone metastasis. An “immune-inflamed” landscape with the presence of TILs predominated in T-NEN, making T-NEN a potentially favorable target for ICB treatment. Further judicious designs of “tailor-made” clinical trials of ICB in T-NEN are urgently needed.</p
