    Nanoparticles of copper stimulate angiogenesis at systemic and molecular level

    Copper is a key element affecting blood vessel growth and muscle development. However, the ions released from Cu salts are toxic. Given their specific physicochemical properties, nanoparticles of Cu (NanoCu) may have different bioactivity and affect the development of blood vessel and muscles in a different manner than Cu salts. The objective of the study was to evaluate the influence of NanoCu on embryo development and angiogenesis at the systemic and molecular level, in experiments using a chick embryo model. Fertilized chicken eggs were divided into a control group, and groups injected with a placebo, CuSO4 or NanoCu. Embryo development at the whole body level and molecular indices using an embryo chorioallantoic membrane model were measured during embryogenesis. The present study indicated for the first time that NanoCu have pro-angiogenic properties at the systemic level, to a greater degree than CuSO4 salt. The properties of NanoCu were confirmed at the molecular level, demonstrating significant effects on mRNA concentration and on mRNA gene expression of all pro-angiogenic and pro-proliferative genes measured herein

    Placenta is Capable of Protecting the Male Fetus from Exposure to Environmental Bisphenol A

    Embryo-fetal exposure to bisphenol A (BPA) could be related to poor male reproductive parameters in rodents, but this concept has not been convincingly confirmed in humans. We investigated the association of environmental BPA exposure of pregnant women with selected endocrine and anthropometric parameters of male newborns. We analyzed plasma BPA from pregnant mothers, umbilical cord, and placental tissues (n = 117/each group) by liquid chromatography and mass spectrometry. LH, FSH, AMH, TGF beta 2, inhibin B, and selected sex steroids were measured in cord plasma. The infant anthropometric parameters included anogenital distance, stretched penile length, head circumference, birthweight, and length. The median BPA concentrations in maternal and umbilical cord plasma, and in placental tissue were 19.0, 8.0, and 22.2 nmol/L, respectively, the levels thus being over twofold lower in the fetal circulation than in the mother or placenta. The BPA concentrations measured were 100-1000-fold lower than those demonstrated in animal experiments to have endocrine disrupting effects. Multivariable regression analysis indicated no significant correlations between the maternal/fetal/placental BPA concentrations and any of the hormone levels or anthropometric parameter measured. Plasma concentrations of BPA confirmed both maternal, placenta, and fetal exposure to environmental BPA, but the concentrations were orders of magnitude lower than those with documented endocrine disrupting activity. Moreover, the maternal/fetal concentration gradient as well as the lack of correlations of BPA levels with any major endocrine or anthropometric parameters measured in the newborns suggest a protective role for the placenta in reducing fetal exposure to the environmental BPA

    Semantic Techniques for the Web: The REWERSE Perspective

    The purpose of this chapter is to report on work that has been done in the REWERSE project concerning hybrid reasoning with rules and ontologies. Two major streams of work have been pursued within REWERSE. They start from the predominant semantics of non-monotonic rules in logic programming. The one stream was an extension of non-monotonic logic programs under answer set semantics, with query interfaces to external knowledge sources. The other stream, in the spirit of the AL-log approach of enhanced deductive databases, was an extension of Datalog (with the well-founded semantics, which is predominant in the database area). The former stream led to so-called non-monotonic dl-programs and hex-programs, and the latter stream to hybrid well-founded semantics. Further variants and derivations of the formalisms (like a well-founded semantics for dl-programs, respecting probabilistic knowledge, priorities, etc.) have been conceived.</p

    Peroneal artery-vein index as a potential factor of thrombosis occurrence in free osteocutaneous fibula flap

    Background: Despite high popularity and great success rates of free osteocutaneous fibula flaps, the flap failure caused by vascular thrombosis is still a challenging problem. The authors present their evaluation of a potential thrombosis risk factor a peroneal artery-vein index. Methods: The authors evaluated the diameters of peroneal vessels and peroneal artery-vein indexes based on the computed tomography angiographies in 10 patients who underwent a mandible reconstruction with a free fibula flap and compared the results with clinical outcome. Results: In one case the flap was lost, because of thrombosis in the donor vein. This patient presented superficial varicose veins of both lower extremities. Peroneal vein diameters in this patient ranged from 5,05 mm to 6,68 mm and were higher than in patients without complications. The peroneal artery-vein index in the patient with thrombosis ranged from 0,37 to 0,50 with median 0,40 and was lower than in patients without complications. Conclusions: High disproportion between peroneal artery and concomitant veins might be a potential risk factor of the occurrence of venal thrombosis. Detailed perioperative examination of peroneal veins in patients with varicosities should be considered. (C) 2016 European Association for Cranio-Maxillo-Facial Surgery. Published by Elsevier Ltd.     Hydrophobin gene deletion and environmental growth conditions impact mechanical properties of mycelium by affecting the density of the material

    Filamentous fungi colonize substrates by forming a mycelium. This network of hyphae can be used as a bio-based material. Here, we assessed the impact of environmental growth conditions and deletion of the hydrophobin gene sc3 on material properties of the mycelium of the mushroom forming fungus Schizophyllum commune. Thermogravimetric analysis showed that Δsc3 mycelium retained more water with increasing temperature when compared to the wild type. The Young's modulus (E) of the mycelium ranged between 438 and 913 MPa when the wild type strain was grown in the dark or in the light at low or high CO2levels. This was accompanied by a maximum tensile strength (σ) of 5.1-9.6 MPa. In contrast, E and σ of the Δsc3 strain were 3-4- fold higher with values of 1237-2727 MPa and 15.6-40.4 MPa, respectively. These values correlated with mycelium density, while no differences in chemical composition of the mycelia were observed as shown by ATR-FTIR. Together, genetic modification and environmental growth conditions impact mechanical properties of the mycelium by affecting the density of the mycelium. As a result, mechanical properties of wild type mycelium were similar to those of natural materials, while those of Δsc3 were more similar to thermoplastics

    Nyctalopin expression in retinal bipolar cells restores visual function in a mouse model of complete X-linked congenital stationary night blindness

    Mutations in the NYX gene that encodes the protein nyctalopin cause congenital stationary night blindness type 1. In no b-wave (nob) mice, a mutation in Nyx results in a functional phenotype that includes the absence of the electroretinogram b-wave and abnormal spontaneous and light-evoked activity in retinal ganglion cells (RGCs). In contrast, there is no morphological abnormality in the retina at either the light or electron microscopic levels. These functional deficits suggest that nyctalopin is required for normal synaptic transmission between retinal photoreceptors and depolarizing bipolar cells (DBCs). However, the synaptic etiology and, specifically, the exact location and function of nyctalopin, remain uncertain. We show that nob DBCs fail to respond to exogenous application of the photoreceptor neurotransmitter, glutamate, thus demonstrating a postsynaptic deficit in photoreceptor to bipolar cell communication. To determine if postsynaptic expression of nyctalopin is necessary and sufficient to rescue the nob phenotype, we constructed transgenic mice that expressed an EYFP-nyctalopin fusion protein on the dendritic tips of the DBCs. Immunohistochemical and ultrastructural studies verified that fusion protein expression was limited to the DBC dendritic tips. Fusion gene expression in nob mice restored normal outer and inner visual function as determined by the electroretinogram and RGC spontaneous and evoked responses.
