    The Evolution of the Satratoxin and Atranone Gene Clusters of Stachybotrys chartarum

    Stachybotrys chartarum is frequently isolated from damp building materials or improperly stored animal forage. Human and animal exposure to the secondary metabolites of this mold is linked to severe health effects. The mutually exclusive production of either satratoxins or atranones defines the chemotypes A and S. Based upon the genes (satratoxin cluster, SC1-3, sat or atranone cluster, AC1, atr) that are supposed to be essential for satratoxin and atranone production, S. chartarum can furthermore be divided into three genotypes: the S-type possessing all sat- but no atr-genes, the A-type lacking the sat- but harboring all atr-genes, and the H-type having only certain sat- and all atr-genes. We analyzed the above-mentioned gene clusters and their flanking regions to shed light on the evolutionary relationship. Furthermore, we performed a deep re-sequencing and LC-MS/MS (Liquid chromatography–mass spectrometry) analysis. We propose a first model for the evolution of the S. chartarum genotypes. We assume that genotype H represents the most ancient form. A loss of the AC1 and the concomitant acquisition of the SC2 led to the emergence of the genotype S. According to our model, the genotype H also developed towards genotype A, a process that was accompanied by a loss of SC1 and SC3

    An Optimized Protocol for DNA Extraction from Wheat Seeds and Loop-Mediated Isothermal Amplification (LAMP) to Detect Fusarium graminearum Contamination of Wheat Grain

    A simple, rapid, and efficient method for isolating genomic DNA from germinated seeds of wheat that is free from polysaccharides and polyphenols is reported. DNA was extracted, treated with RNase, measured and tested for completeness using agarose gel electrophoresis. DNA purification from wheat grains yielded abundant, amplifiable DNA with yields typically between 100 and 200 ng DNA/mg. The effectiveness and reliability of the method was tested by assessing quantity and quality of the isolated DNA using three PCR-based markers. Inter-simple sequence repeats (ISSRs) were used to assess the genetic diversity between different wheat varieties. Specific PCR primer pair Tox5-1/Tox5-2 and a loop-mediated isothermal amplification (LAMP) procedure were used to detect genomic DNA of Fusarium graminearum in contaminated wheat seeds. In this method there is no need to use liquid nitrogen for crushing germinated seedlings. The protocol takes approximately one hour to prepare high quality DNA. In combination with the LAMP assay it is a fast and cost-effective alternative to traditional diagnostic methods for the early detection of toxigenic fusaria in cereals

    Epigenetics in radiotherapy: Where are we heading?

    Radiotherapy is an important component of anti-cancer treatment. However, not all cancer patients respond to radiotherapy, and with current knowledge clinicians are unable to predict which patients are at high risk of recurrence after radiotherapy. There is therefore an urgent need for biomarkers to guide clinical decision-making. Although the importance of epigenetic alterations is widely accepted, their application as biomarkers in radiotherapy has not been studied extensively. In addition, it has been suggested that radiotherapy itself introduces epigenetic alterations. As epigenetic alterations can potentially be reversed by drug treatment, they are interesting candidate targets for anticancer therapy or radiotherapy sensitizers. The application of demethylating drugs or histone deacetylase inhibitors to sensitize patients for radiotherapy has been studied in vitro, in vivo as well as in clinical trials with promising results.     Determination of the number of light neutrino species from single photon production at LEP

    A determination of the number of light neutrino families performed by measuring the cross section of single photon production in \ee\ collision near the \Zo\ resonance is reported. From an integrated luminosity of 100 pb1100~\mathrm{pb^{-1}}, collected during the years 1991--94, we have observed 2091 single photon candidates with an energy above 1~\GeV\ in the polar angular region 45<θγ<13545^\circ < \theta_\gamma < 135^\circ. From a maximum likelihood fit to the single photon cross section, the \Zo\ decay width into invisible particles is measured to be \Ginv = 498 \pm 12 \mathrm{(stat)} \pm 12 \mathrm{(sys)~MeV}. Using the Standard Model couplings of neutrinos to the \Zo, the number of light neutrino species is determined to be $N_\nu = 2.98 \pm 0.07 (\mathrm{stat}) \pm 0.07 (\mathrm{sys}).

    We report on a search for R--parity breaking effects due to supersymmetric tau--sneutrino exchange in the reactions e+e- to e+e- and e+e- to mu+mu- at centre--of--mass energies from 91~{\GeV} to 172~{\GeV}, using the L3 detector at LEP. No evidence for deviations from the Standard Model expectations of the measured cross sections and forward--backward asymmetries for these reactions is found. Upper limits for the couplings λ131\lambda_{131} and λ232\lambda_{232} for sneutrino masses up to m_{\SNT} \leq 190~\GeV are determined from an analysis of the expected effects due to tau sneutrino exchange

    Search for charged Higgs bosons in e+ee^+ e^- collisions at centre-of-mass energies between 130 and 183 GeV

    A search for pair-produced charged Higgs bosons is performed with the L3 detector at LEP using data collected at centre-of-mass energies from \mbox{130 to 183 \GeV{}}, corresponding to an integrated luminosity of 88.3 \pb. The Higgs decays into a charm and a strange quark or into a tau lepton and its associated neutrino are considered. The observed candidates are consistent with the expectations from Standard Model background processes. A lower limit of 57.5 \GeV{} on the charged Higgs mass is derived at 95\% CL, independent of the decay branching ratio \mathrm{Br(H^\pm\ra \tau\nu)}

    Inclusive Charm Production in Two-Photon Collisions at LEP

    The cross section of charm production in γγ\mathrm{\gamma \gamma} collisions σ(e+ee+eccˉX)\mathrm{\sigma (e^+e^- \rightarrow e^+e^-c\bar{c} X)} is measured at LEP with the L3 detector at centre of mass energies from 91 GeV to 183 GeV. Charmed hadrons are identified by electrons and muons from semi-leptonic decays. The direct process γγccˉ\mathrm{\gamma \gamma \rightarrow c \bar{c}} is found to be insufficient to describe the data. The measured cross section values and event distributions require contributions from resolved processes, which are sensitive to the gluon density in the photon

    Heavy Quarkonium Production in Z Decays

    We report measurements of the inclusive production of heavy quarkonium states in Z\mathrm {Z} decays based on the analysis of 3.6 million hadronic events collected by the L3 detector at LEP. The measurement of inclusive J production and an improved 95%95\% confidence level upper limit on Υ\Upsilon production are presented. In addition, two independent measurements of the ratio, fpf_{\mathrm{p}}, of prompt J mesons to those from B decay are made using two different isolation cuts to separate prompt J mesons from J mesons produced in the decays of b hadrons.     Study of Neutral-Current Four-Fermion and ZZ Production in e+ee^+ e^- Collisions at s\sqrt{s}= 183 GeV

    Study of Neutral--Current Four--Fermion and ZZ \\ Production in e+e\rm e^+ e^- Collisions at s=\rm \sqrt{s}= 183 GeV A study of neutral--current four--fermion processes is performed using a data sample corresponding to 55.3~pb1^{-1} of integrated luminosity collected by the L3 detector at LEP at an average centre--of--mass energy of 183~\GeV. The neutral--current four--fermion cross sections for final states with a pair of charged leptons plus jets and with four charged leptons are measured to be consistent with the Standard Model predictions. Events with fermion pair masses close to the Z boson mass are selected in all observable final states and the ZZ production cross section is measured to be %\begin{center} σZZ=0.300.160.03+0.22+0.07pb,\rm \sigma_{ZZ} = 0.30 ^{+0.22\,\,+0.07}_{-0.16\,\,-0.03}\,\mathrm{pb}, %\end{center} in agreement with the Standard Model expectation. No evidence for the existence of anomalous triple gauge boson ZZZ and ZZγ\gamma couplings is found and limits on these couplings are set
