    Seleção de clones de braquiária para resistência à lagarta‑do‑cartucho

    The objective of this work was to evaluate and select signal grass (Urochloa ruziziensis) clones resistant to fall armyworm, Spodoptera frugiperda (Lepidoptera: Noctuidae), for use in a crop‑livestock‑forest integration system. One hundred and forty-seven clones of U. ruziziensis were evaluated in a completely randomized design with three replicates. An artificial diet, adequate for insect development, was used as a control and susceptibility standard. Leaves of different U. ruziziensis clones were collected and offered as food to the insects, kept individually in rearing units. Larval survival, duration, and biomass of larvae and pupa were evaluated. Significant differences were observed between clones for all evaluated traits, showing the marked effect of U. ruziziensis clones on the development of S. frugiperda. Twenty clones were identified in which there was no development of larvae and 55 clones in which there was an average larval survival of 33%. For the other clones, which were better hosts, insect development was affected by the longer larval stage and by the reduction in pupa biomass. The obtained results show the potential of using Urochloa ruziziensis clones for the reduction of Spodoptera frugiperda populations in a crop‑livestock‑forest integration system.O objetivo deste trabalho foi avaliar e selecionar clones de braquiária (Urochloa ruziziensis) resistentes à lagarta‑do‑cartucho, Spodoptera frugiperda (Lepidoptera: Noctuidae), para uso em sistema de integração lavoura‑pecuária‑floresta. Foram avaliados 147 clones de U. ruziziensis em delineamento inteiramente casualizado, com três repetições. Como testemunha e padrão de suscetibilidade, foi utilizada uma dieta artificial adequada ao desenvolvimento do inseto. Folhas dos diferentes clones de U. ruziziensis foram coletadas e oferecidas como alimento aos insetos, mantidos individualizados em unidades de criação. Foram avaliadas a sobrevivência, a duração e a biomassa de lagartas e pupas. Observaram-se diferenças significativas entre os clones para todas as características avaliadas, o que evidencia a influência acentuada dos clones de U. ruziziensis no desenvolvimento de S. frugiperda. Foram identificados 20 clones nos quais não houve desenvolvimento da lagarta e 55 clones nos quais houve sobrevivência média das lagartas de 33%. Para os demais clones, que se mostraram melhores hospedeiros, o desenvolvimento do inseto foi afetado pela maior duração do estágio larval e pela redução da biomassa da pupa. Os resultados obtidos são indicativos do potencial de utilização de clones de Urochloa ruziziensis para redução de populações de Spodoptera frugiperda em sistema de integração lavoura‑pecuária‑floresta

São Cristóvão, SE, Brasil.This study evaluated three different methods for the formation of an inclusion complex between alpha- and beta-cyclodextrin (α- and β-CD) and limonene (LIM) with the goal of improving the physicochemical properties of limonene. The study samples were prepared through physical mixing (PM), paste complexation (PC), and slurry complexation (SC) methods in the molar ratio of 1:1 (cyclodextrin:limonene). The complexes prepared were evaluated with thermogravimetry/derivate thermogravimetry, infrared spectroscopy, X-ray diffraction, complexation efficiency through gas chromatography/mass spectrometry analyses, molecular modeling, and nuclear magnetic resonance. The results showed that the physical mixing procedure did not produce complexation, but the paste and slurry methods produced inclusion complexes, which demonstrated interactions outside of the cavity of the CDs. However, the paste obtained with β-cyclodextrin did not demonstrate complexation in the gas chromatographic technique because, after extraction, most of the limonene was either surface-adsorbed by β-cyclodextrin or volatilized during the procedure. We conclude that paste complexation and slurry complexation are effective and economic methods to improve the physicochemical character of limonene and could have important applications in pharmacological activities in terms of an increase in solubility

São Cristóvão, SE, BrasilIsopulegol (ISOP) is a monoterpenoid alcohol presented in the essential oils of several plants that possesses therapeutic properties The aim of this work was prepare samples with ISOP and α- and β-cyclodextrins (α- and β-CD) through three different methods: physical mixture, paste method (PC) and slurry complexation (SC). In order to evaluate the formation of inclusion complexes, the techniques of differential scanning calorimetry, thermogravimetry/derivative thermogravimetry, fourier transform infrared spectroscopy, X-ray diffractometry (XRD), gas chromatography–mass spectrometry analyses (GC/MS), docking, nuclear magnetic resonance and scanning electron microscopy were considered. The analyses of the α-CD or β-CD/ISOP revealed the formation of a complex mainly through the PC and SC methods for α-CD and β-CD, respectively. XRD diffraction characteristics presented formation of a trend to new solid phase, which suggested the formation of inclusion complexes. The GC/MS demonstrated that the PC method was the best one to form complexation with α-CD (48.8 %). Concerning β-CD, the SC method exhibited the strongest complexation (68.3 %). Furthermore, the molecular theoretical docking study demonstrated that α-CD/ISOP inclusion complex formed a more stable complex than did the β-CD/ISOP inclusion complex

    Effect of α-Bisabolol and Its β-Cyclodextrin Complex as TetK and NorA Efflux Pump Inhibitors in Staphylococcus aureus Strains

    Efflux pumps are proteins present in the plasma membrane of bacteria, which transport antibiotics and other compounds into the extracellular medium, conferring resistance. The discovery of natural efflux pump inhibitors is a promising alternative. α-Bisabolol is a sesquiterpene isolated from several plants such as Matricaria chamomilla L. and has important properties such as antibacterial and anti-inflammatory activity. Currently, the formation of inclusion complexes with β-Cyclodextrin has been used for improving the physicochemical characteristics of the host molecule. This study evaluated the effect of α-Bisabolol, in isolation and in complexation with β-Cyclodextrin, as TetK and NorA efflux pump inhibitors in Staphylococcus aureus strains. The minimum inhibitory concentration (MIC) was determined. Subsequently, inhibitory activity over the pumps was observed by an MIC reduction for the antibiotics, by using subinhibitory concentrations (MIC/8) in combination with tetracycline and norfloxacin. The MIC of the compounds was ≥1024 μg/mL. α-Bisabolol potentiated the action of tetracycline and reduced the MIC of norfloxacin to a clinically relevant concentration. The complexed substance showed synergism however, the effect of the isolated α-Bisabolol was superior to the complex. These results indicate α-Bisabolol is a potential substance to be used as an efflux pump inhibitor

This oil is important therapeutically, but the low solubility and bioavailability compromises your use. Therein, the aim of this study was to obtain and to characterize physico-chemically the samples obtained by physical mixture (PM), paste complexation (PC) and slurry complexation (SC) of the essential oil Hyptis martiusii (EOHM) in β-CD, and to compare the antibacterial and modulatory-antibiotic activity of products obtained and oil free. The physicochemical characterization was performed by differential scanning calorimetry (DSC), thermogravimetry/derivative thermogravimetry (TG/DTG), scanning electron microscopy (SEM), X-ray diffraction (XRD) and Karl Fischer titration. Additionally, the antibacterial tests were performed by microdilution technique. Thus, it was observed that the PM method showed low complexing capacity, unlike PC and SC in which it was observed the formation of inclusion complexes. In addition, the second stage of the TG/DTG curves showed that SC was the best method inclusion with mass loss of 6.9% over the PC that was 6.0%. The XRD results corroborate with the results above suggesting the formation of new solid phase and the SEM photomicrographs showed the porous surface of the samples PC and SC. The essential oil alone demonstrated an antibacterial and modulatory effect against the S. aureus and the Gram negative strain, respectively. However, the β-CD and the inclusion complex did not demonstrate any biological activity in the performed antibacterial assays
