    Contribution of cyclodextrins in the developement of different pharmaceutical formulations of a nex matrix metalloproteinase inhibitor

    Ro 28-2653 is a new synthetic inhibitor of matrix metalloproteinases. The ability of these enzymes to degrade various components of the extracellular matrix seems to play a major role in tumors progression and is potentially effective against bronchial remodeling in asthma and BPCO. Ro 28-2653 is very poorly soluble in water. This low solubility estimated at about 0.56 lg/ml in water at 25 C gives rise to difficulties in pharmaceutical formulation of oral, injectable or nebulizable solutions. The purpose of our study is to prepare and to characterize inclusion complexes between Ro 28-2653 and cyclodextrins and to investigate the biopharmaceutical repercussion of the inclusion of the active substance.The complex formation was investigated by phase solubility studies. 1H-NMR spectroscopy and molecular modeling studies were carried out to elucidate the structure of the inclusion complex between Ro 28-2653 and cyclodextrin. Oral, intravenous and nebulizable solutions of Ro 28-2653 were developed with cyclodextrin. The in vivo studies were performed on healthy sheep for the pharmacokinetic evaluation of the oral and intravenous formulations while the nebulization of the complex solution was studied by using an asthma model in mouse

    Automated method for the determination of a new matrix metalloproteinase inhibitor in ovine plasma and serum by coupling of restricted access material for on-line sample clean-up to liquid chromatography

    A fully automated liquid chromatographic method was developed for the determination of Ro 28-2653, a new synthetic inhibitor of matrix metalloproteinases (MMPs), in ovine serum and plasma. The method was based on the coupling of a pre-column packed with restricted access material, namely LiChrospher RP-8 ADS (alkyl diol silica), for sample clean-up to an analytical column containing octyl silica stationary phase. One hundred mul of biological sample, to which 2-propanol was automatically added, were injected onto the ADS pre-column, which was then washed with a washing liquid consisting of a mixture of 25 mM phosphate buffer (pH 7.0) and acetonitrile (90: 10; v/v) for 10 min. By rotation of the switching valve, the analyte was then eluted in the back-flush mode with the LC mobile phase composed of a mixture of acetonitrile and 25 mM phosphate buffer (pH 7.0) (57:43; v/v). The UV detection was performed at 395 nm. The main parameters likely to influence the sample preparation technique were investigated. The method was then validated over a concentration range from 17.5 to 1950 ng/mI, the first concentration level corresponding to the lower limit of quantitation. At this concentration level, the mean bias and the R.S.D. value for intermediate precision were -2.4% and 4.2%, respectively. (C) 2004 Elsevier B.V. All rights reserved

    Archeohandi: protocol for a national disabilities database in archaeology in France

    The archaeology of disability is a relatively recent and little-known approach in France. While the study of palaeopathology now goes hand in hand with funerary archaeology and osteoarchaeology, the French study of disabilities and disabling pathologies remains marginal and unevenly treated, depending on location, chronology and researcher’s interest. This paper focuses on highlighting the compatibility between this new research area, the obligations of osteoarchaeology, and the benefits of developing a national, diachronic, and interdisciplinary study. A database is designed within an interpretive, consensual framework, that can be adapted to overcome limitations and promote open-minded research on the care of the disabled in their own communities. A preliminary category selection of disabling pathologies has been made. These are trepanation, completely edentulous and/or compensating denture, neuronal impairment, severe scoliosis, Paget's disease, Diffuse Idiopathic Skeletal Hyperostosis (DISH), rickets, dwarfism, infectious diseases, unreduced fracture, amputation, severe degenerative disease and others. This list has been critically reviewed by experts in the field; it will evolve in a somewhat Darwinian fashion. Our database is hosted on the Huma-Num platform, with a management interface and quick access based on multiple tabs. The data includes information about archaeological operations, subjects, and pathologies; it is complemented by pictorial data stored on the Nakala platform. The development involved creating a prototype using HTML, CSS, JavaScript, SQL, and PHP, with features to display, add, modify, and delete operations and subjects. Enhancements have been made, including search optimization, charts, and the ability to export data in CSV format. The database, whose administrative interface can be accessed at archeohandi.huma-num.fr, contains so far 211 existing operations with a total of 1232 registered subjects spread throughout metropolitan France. These initial data reveal numerous research perspectives in osteoarchaeology that can be combined with other research topics, such as virtual reality

    peer reviewedL'industrialisation des productions animales a mis en évidence la nécessité d'éviter des administrations réitérées chez des animaux dont la manipulation est difficile, et de développer des formes à libération contrôlée ou du moins fortement prolongée. L'anatomie particulière du tube digestive des ruminants offre en outre des possibilités inédites pour le développement de tels systèmes thérapeutiques, notamment grâce à la rétention dans le réticulo-rumen. De telles formes ont essentiellement été développées pour l'administration de médicaments antiparasitaires et antibactériens, de suppléments nutritionnels et de promoteurs croissance. Cette revue décrit les applications potentielles de la technologie des formes à libération contrôlée et prolongée en médecine vétérinaire et les différents dispositifs à rétention dans le réticulo-rumen décrits dans la littérature, brevetés ou commercialisés. More intensive methods ofanimal husbandry have revealed the necessity of avoiding repeated administration ofmedication to animals which are difficult to handle and of developing controlled and sustained release dosage forms. The physiological characteristics of the digestive tract of ruminants provide unique opportunity for controlled release technology, especially due to the animal's reticulo-rumen retention ability. The majorareas of application of these controlled and sustained release dosage forms include the delivery of antibiotic and antiparasitic agents, trace nutrients and growth promoting agents. This review describes the potential applications of controlled and sustained release technology in veterinary medicine and the various reticulo-rumen retention devices which are patented, marketed or described in the literature

    Le SIDA est un fléau mondial, en 2007, 2 millions de personnes en sont décédées. En France, les jeunes sont privilégiés, de nombreuses associations interviennent afin de les prévenir des risques de contamination. C est le cas de l Association STOP SIDA, qui met en place des interventions dans les établissements scolaires de la région Nord Pas-de-Calais assurées par des étudiants en cinquième année de pharmacie.
