    Genetically Encoded Whole Cell Biosensor for Drug Discovery of HIF-1 Interaction Inhibitors

    The heterodimeric transcription factor, hypoxia inducible factor-1 (HIF-1), is an important anticancer target as it supports the adaptation and response of tumors to hypoxia. Here, we optimized the repressed transactivator yeast two-hybrid system to further develop it as part of a versatile yeast-based drug discovery platform and validated it using HIF-1. We demonstrate both fluorescence-based and auxotrophy-based selections that could detect HIF-1α/HIF-1β dimerization inhibition. The engineered genetic selection is tunable and able to differentiate between strong and weak interactions, shows a large dynamic range, and is stable over different growth phases. Furthermore, we engineered mechanisms to control for cellular activity and off-target drug effects. We thoroughly characterized all parts of the biosensor system and argue this tool will be generally applicable to a wide array of protein-protein interaction targets. We anticipate this biosensor will be useful as part of a drug discovery platform, particularly when screening DNA-encoded new modality drugs

    Bone Lining Cells: Structure and Function

    Bone lining cells (BLC\u27s) cover inactive (nonremodeling) bone surfaces, particularly evident in the adult skeleton. BLC\u27s are thinly extended over bone surfaces, have flat or slightly ovoid nuclei, connect to other BLC\u27s via gap junctions, and send cell processes into surface canaliculi. BLC\u27s can be induced to proliferate and differentiate into osteogenic cells and may represent a source of determined osteogenic precursors. BLC\u27s and other cells of the endosteal tissues may be an integral part of the marrow stromal system and have important functions in hematopoiesis, perhaps by controlling the inductive microenvironment. Because activation of bone remodeling occurs on inactive bone surfaces, BLC\u27s may be involved in the propagation of the activation signal that initiates bone resorption and bone remodeling. Evidence also suggests that BLC\u27s are important in the maintenance of the bone fluids and the fluxes of ions between the bone fluid and interstitial fluid compartments for mineral homeostasis

    VectorBase: improvements to a bioinformatics resource for invertebrate vector genomics.

    VectorBase (http://www.vectorbase.org) is a NIAID-supported bioinformatics resource for invertebrate vectors of human pathogens. It hosts data for nine genomes: mosquitoes (three Anopheles gambiae genomes, Aedes aegypti and Culex quinquefasciatus), tick (Ixodes scapularis), body louse (Pediculus humanus), kissing bug (Rhodnius prolixus) and tsetse fly (Glossina morsitans). Hosted data range from genomic features and expression data to population genetics and ontologies. We describe improvements and integration of new data that expand our taxonomic coverage. Releases are bi-monthly and include the delivery of preliminary data for emerging genomes. Frequent updates of the genome browser provide VectorBase users with increasing options for visualizing their own high-throughput data. One major development is a new population biology resource for storing genomic variations, insecticide resistance data and their associated metadata. It takes advantage of improved ontologies and controlled vocabularies. Combined, these new features ensure timely release of multiple types of data in the public domain while helping overcome the bottlenecks of bioinformatics and annotation by engaging with our user community
