    Reliability of spring recession curve analysis as a function of the temporal resolution of the monitoring dataset

    Mountain springs represent one of the largest and most precious sources of potable water in Italy, necessary to meet the water needs of the population. Optimizing the present and future management strategies of mountain groundwater resources has become increasingly necessary. The accuracy and frequency of the flow rate (Q) measurements determine and restrict the processes that can be studied using spring hydrograph and recession curve analysis. Therefore, to properly define mountain aquifers’ hydrogeological properties, it turns out important to highlight the variation of the error in the estimation of the hydrogeological parameters as the time interval of sampling varies. In this paper, recession curve analysis was performed on two different mountain springs (Spring 1 and Spring 2) of north-western Italy, firstly considering available 4-h resolution measuring data and subsequently by resampling data to simulate longer sampling intervals of 1, 3, 7, 15, and 30 days. The resulting distribution of errors introduced by longer acquisition intervals underlined how the percentage error increases with increasing acquisition interval. For obtaining an adequate estimation of mountain aquifer hydrodynamic parameters, in place of continuous hourly data, 1-day and 3-day sampling intervals with associated errors respectively lower than 5% and 10% were found to be valid

    Liquid Foam-Ethyl Vinyl Acetate Adhesive Systems for Lining Process of Paintings: Prospects of a User-Friendly, Harmless Alternative to Conventional Products

    The lining of paintings is a process of conservation science and art restoration used to strengthen, flatten, or consolidate paintings on canvas by attaching by means of adhesives a second canvas to the back of the existing one. To this aim, the prospects of the use of ethyl vinyl acetate (EVA) resins in aqueous dispersion applied as an adhesive in a foam form have been investigated in the present study. The key physical properties of the foam have been investigated, with a deep focus on rheological behavior and the drying rate, comparing the results with those obtained using the liquid products that are commercially available. Dedicated mock-ups have been prepared to test the adhesive for the lining process, inspecting adhesion strength, colorimetric properties, and the influence on the possible chromatic and visual alteration of the surface, also looking at the chemical interaction with painting materials and the deterioration after an artificial ageing process. The results obtained clearly indicated that the proposed technology is very suitable for the targeted application, and an EVA water-based foamed dispersion can be used for paintings' lining, in view of the ease of application, being an appropriate adhesion, no chemical interaction, nor the deterioration of the painting

    Optomechanical two-photon hopping

    The hopping mechanism plays a key role in collective phenomena emerging in many-body physics. The ability to create and control systems that display this feature is important for next generation quantum technologies. Here we study two cavities separated by a vibrating two-sided perfect mirror and show that, within currently available experimental parameters, this system displays photon-pair hopping between the two electromagnetic resonators. In particular, the two-photon hopping is not due to tunneling, but rather to higher-order resonant processes. Starting from the classical problem we quantize the system and show this purely quantum feature. This opens the possibility to investigate a new mechanism of photon-pair propagation in optomechanical lattices

    Wharton’s Jelly Mesenchymal Stromal Cells Support the Expansion of Cord Blood–derived CD34 + Cells Mimicking a Hematopoietic Niche in a Direct Cell–cell Contact Culture System

    Wharton’s jelly mesenchymal stromal cells (WJ-MSCs) have been recently exploited as a feeder layer in coculture systems to expand umbilical cord blood–hematopoietic stem/progenitor cells (UCB-HSPCs). Here, we investigated the role of WJ-MSCs in supporting ex vivo UCB-HSPC expansion either when cultured in direct contact (DC) with WJ-MSCs or separated by a transwell system or in the presence of WJ-MSC–conditioned medium. We found, in short-term culture, a greater degree of expansion of UCB-CD34 + cells in a DC system (15.7 ± 4.1-fold increase) with respect to the other conditions. Moreover, in DC, we evidenced two different CD34 + cell populations (one floating and one adherent to WJ-MSCs) with different phenotypic and functional characteristics. Both multipotent CD34 + /CD38 − and lineage-committed CD34 + /CD38 + hematopoietic progenitors were expanded in a DC system. The former were significantly more represented in the adherent cell fraction than in the floating one (18.7 ± 11.2% vs. 9.7 ± 7.9% over the total CD34 + cells). Short-term colony forming unit (CFU) assays showed that HSPCs adherent to the stromal layer were able to generate a higher frequency of immature colonies (CFU-granulocyte/macrophage and burst-forming unit erythroid/large colonies) with respect to the floating cells. In the attempt to identify molecules that may play a role in supporting the observed ex vivo HSPC growth, we performed secretome analyses. We found a number of proteins involved in the HSPC homing, self-renewal, and differentiation in all tested conditions. It is important to note that a set of sixteen proteins, which are only in part reported to be expressed in any hematopoietic niche, were exclusively found in the DC system secretome. In conclusion, WJ-MSCs allowed a significant ex vivo expansion of multipotent as well as committed HSPCs. This may be relevant for future clinical applications

    Effects of PPARγ agonists on the expression of leptin and vascular endothelial growth factor in breast cancer cells.

    The obesity hormone leptin has been implicated in breast cancer development. Breast cancer cells express the leptin receptor and are able to synthesize leptin in response to obesity-related stimuli. Furthermore, leptin is a positive regulator of vascular endothelial growth factor (VEGF) and high levels of both proteins are associated with worse prognosis in breast cancer patients. Peroxisome proliferator-activated receptor (PPAR) ligands are therapeutic agents used in patient with Type 2 diabetes and obesity which have recently been studied for their potential anti-tumor effect. Here, we studied if these compounds, ciglitazone and GW1929, can affect the expression of leptin and VEGF in breast cancer cells. In MDA-MB-231 and MCF-7 breast cancer cells, treatment with submolar concentrations of ciglitazone and GW1929 elevated the expression of leptin and VEGF mRNA and protein, and increased cell viability and migration. These effects coincided with increased recruitment of PPAR to the proximal leptin promoter and decreased association of a transcriptional factor Sp1 with this DNA region

    Activation of the motor cortex during phasic rapid eye movement sleep

    When dreaming during rapid eye movement (REM) sleep, we can perform complex motor behaviors while remaining motionless. How the motor cortex behaves during this state remains unknown. Here, using intracerebral electrodes sampling the human motor cortex in pharmacoresistant epileptic patients, we report a pattern of electroencephalographic activation during REM sleep similar to that observed during the performance of a voluntary movement during wakefulness. This pattern is present during phasic REM sleep but not during tonic REM sleep, the latter resembling relaxed wakefulness. This finding may help clarify certain phenomenological aspects observed in REM sleep behavior disorder
