    Transcriptome analysis of stem development in the tumourous stem mustard Brassica juncea var. tumida Tsen et Lee by RNA sequencing

    <p>Abstract</p> <p>Background</p> <p>Tumourous stem mustard (<it>Brassica juncea </it>var. <it>tumida </it>Tsen et Lee) is an economically and nutritionally important vegetable crop of the <it>Cruciferae </it>family that also provides the raw material for <it>Fuling </it>mustard. The genetics breeding, physiology, biochemistry and classification of mustards have been extensively studied, but little information is available on tumourous stem mustard at the molecular level. To gain greater insight into the molecular mechanisms underlying stem swelling in this vegetable and to provide additional information for molecular research and breeding, we sequenced the transcriptome of tumourous stem mustard at various stem developmental stages and compared it with that of a mutant variety lacking swollen stems.</p> <p>Results</p> <p>Using Illumina short-read technology with a tag-based digital gene expression (DGE) system, we performed <it>de novo </it>transcriptome assembly and gene expression analysis. In our analysis, we assembled genetic information for tumourous stem mustard at various stem developmental stages. In addition, we constructed five DGE libraries, which covered the strains <it>Yong'an </it>and <it>Dayejie </it>at various development stages. Illumina sequencing identified 146,265 unigenes, including 11,245 clusters and 135,020 singletons. The unigenes were subjected to a BLAST search and annotated using the GO and KO databases. We also compared the gene expression profiles of three swollen stem samples with those of two non-swollen stem samples. A total of 1,042 genes with significantly different expression levels occurring simultaneously in the six comparison groups were screened out. Finally, the altered expression levels of a number of randomly selected genes were confirmed by quantitative real-time PCR.</p> <p>Conclusions</p> <p>Our data provide comprehensive gene expression information at the transcriptional level and the first insight into the understanding of the molecular mechanisms and regulatory pathways of stem swelling and development in this plant, and will help define new mechanisms of stem development in non-model plant organisms.</p

    Due to the continuous changes of political environment, consumption habits, technological progress and other factors, the external environment of enterprises is full of uncertainty. The turbulence of external environment is not conducive to the long-term operation and development of enterprises, but also brings great challenges to the selection of suppliers. This makes the competition of enterprises focus on how to choose long-term cooperation suppliers in the uncertain external environment. In addition, due to the deterioration of the global environment, governments pay more and more attention to environmental pollution, and consumers are more and more inclined to green consumption, which makes many companies pay more and more attention to environmental indicators when selecting suppliers. In the case of external environment turbulence and serious environmental pollution, the evaluation and selection of green suppliers in uncertain environment is particularly important for the long-term development of enterprises. What&rsquo;s more, when the supplier&rsquo;s capability gap is small, the decision-maker often hesitates among several suppliers. In this paper, the hesitant fuzzy is used to describe the hesitant psychology of decision-makers in selecting suppliers, the variance fluctuation is used to describe the characteristics of hesitant fuzzy numbers, and the probability is used to measure the uncertainty of the environment. A green supplier evaluation model under the uncertainty environment is proposed, which comprehensively evaluates the green suppliers under the uncertain environment. Furthermore, it is compared with other methods that do not consider the uncertainty and the adaptability of evaluation method and right confirmation method, so as to reflect the influence of uncertainty to green supplier evaluation and the importance of adaptability of evaluation method and right confirmation method

    Coupling Coordinated Evolution and Forecast of Tourism-Urbanization-Ecological Environment: The Case Study of Chongqing, China

    As an important pioneering city in western China, Chongqing’s urbanization and tourism development have brought increasing pressure on the eco-environmental system. This paper presents a comprehensive index system for assessment of the level of tourism based on four aspects: market size, tourism economy, tourism resources, and tourism services. In this paper, we use five indices—economic urbanization, demographic urbanization, social urbanization, spatial urbanization, and cultural urbanization—to establishe a comprehensive index system for urbanization. The developed index system also characterizes the eco-environment based on three factors: eco-environmental pressure, eco-environmental response, and eco-environmental state. A coupling coordination degree model (CCDM) focusing on the degree of coupling coordination between tourism, urbanization, and the eco-environment was established using panel data collected from 2000 to 2017 for Chongqing, China. In addition, we used the gray prediction model to predict the degree of coupling coordination of the three systems in Chongqing from 2018 to 2023. The results showed that (1) overall, despite the twists and turns in the middle of the time period, the degree of coupling coordination of the three systems has become increasingly coordinated over time; (2) the three parameters (α-tourism, ß-urbanization, and d-eco-environmental) that have been widely used in previous studies had little effect on the coupling coordinated system; and (3) according to the prediction results, the degree of coupling coordination of the three systems will continue to rise slowly, and the cooperation between the three systems will be close to perfect by 2023, which means that the development of one system will greatly drive the other two systems

    Melittin (MEL) is a 26-amino acid polypeptide with a variety of pharmacological and toxicological effects, which include strong surface activity on cell lipid membranes, hemolytic activity, and potential anti-tumor properties. However, the clinical application of melittin is restricted due to its severe hemolytic activity. Different nanocarrier systems have been developed to achieve stable loading, side effects shielding, and tumor-targeted delivery, such as liposomes, cationic polymers, lipodisks, etc. In addition, MEL can be modified on nano drugs as a non-selective cytolytic peptide to enhance cellular uptake and endosomal/lysosomal escape. In this review, we discuss recent advances in MEL&rsquo;s nano-delivery systems and MEL-modified nano drug carriers for cancer therapy

    ObjectiveThis study aimed to assess the static balance ability of the older adults with mild cognitive impairment (MCI) while standing on soft and hard support surfaces.MethodsForty older adults participated in this study (21 in the MCI group and 19 in the control group). Participants were required to perform balance tests under four conditions of standing: standing on a hard support surface with eyes open, standing on a soft support surface with eyes open, standing on a hard support surface with eyes closed, and standing on a soft support surface with eyes closed. Each test was measured in three trials and each trial lasted 30 seconds. Participants were asked to take off their shoes and place their feet in a parallel position with a 20-centimeter distance for bipedal support. The trajectories of the center of pressure (COP) were measured using a Kistler force platform with a frequency of 1000 Hz to assess balance while standing in both groups, with larger COP trajectories indicating poorer static balance in older adults.ResultsWith eyes open, the displacement of COP in the anterior-posterior direction(D-ap) (hard support surface: P = 0.003) and the 95% confidence ellipse area(95%AREA-CE) (soft support surface: P = 0.001, hard support surface: P ml), and 95%AREA-CE of the COP were no significant between-group differences when standing on hard support surfaces. However, the RDIST (P = 0.014), RDISTml (P = 0.014), and 95%AREA-CE (P = 0.001) of the COP in the MCI group on the soft support surfaces were significantly larger than the control group.
