In vitro expanded human CD4+CD25+ regulatory T cells suppress effector T cell proliferation.
Regulatory T cells (Tregs) have been shown to be critical in the balance between autoimmunity and tolerance and have been implicated in several human autoimmune diseases. However, the small number of Tregs in peripheral blood limits their therapeutic potential. Therefore, we developed a protocol that would allow for the expansion of Tregs while retaining their suppressive activity. We isolated CD4+CD25 hi cells from human peripheral blood and expanded them in vitro in the presence of anti-CD3 and anti-CD28 magnetic Xcyte Dynabeads and high concentrations of exogenous Interleukin (IL)-2. Tregs were effectively expanded up to 200-fold while maintaining surface expression of CD25 and other markers of Tregs: CD62L, HLA-DR, CCR6, and FOXP3. The expanded Tregs suppressed proliferation and cytokine secretion of responder PBMCs in co-cultures stimulated with anti-CD3 or alloantigen. Treg expansion is a critical first step before consideration of Tregs as a therapeutic intervention in patients with autoimmune or graft-versus-host disease
Extending Hybrid CSP with Probability and Stochasticity
Probabilistic and stochastic behavior are omnipresent in computer controlled
systems, in particular, so-called safety-critical hybrid systems, because of
fundamental properties of nature, uncertain environments, or simplifications to
overcome complexity. Tightly intertwining discrete, continuous and stochastic
dynamics complicates modelling, analysis and verification of stochastic hybrid
systems (SHSs). In the literature, this issue has been extensively
investigated, but unfortunately it still remains challenging as no promising
general solutions are available yet. In this paper, we give our effort by
proposing a general compositional approach for modelling and verification of
SHSs. First, we extend Hybrid CSP (HCSP), a very expressive and process
algebra-like formal modeling language for hybrid systems, by introducing
probability and stochasticity to model SHSs, which is called stochastic HCSP
(SHCSP). To this end, ordinary differential equations (ODEs) are generalized by
stochastic differential equations (SDEs) and non-deterministic choice is
replaced by probabilistic choice. Then, we extend Hybrid Hoare Logic (HHL) to
specify and reason about SHCSP processes. We demonstrate our approach by an

Long-term average spectral characteristics of Cantonese alaryngeal speech
Objective: In Hong Kong, esophageal (SE), tracheoesophageal (TE), electrolaryngeal (EL), and pneumatic artificial laryngeal (PA) speech are commonly used by laryngectomees as a means to regain verbal communication after total laryngectomy. While SE and TE speech has been studied to some extent, little is known regarding the EL and PA sound quality. The present study examined the sound quality associated with SE, TE, EL, and PA speech, and compared with that associated with laryngeal (NL) speech by using long-term average speech spectra (LTAS). Methods: Continuous speech samples of reading a 136-word passage were obtained from NL, SE, TE, EL, and PA speakers of Cantonese. The alaryngeal speakers were all superior speakers selected from the New Voice Club of Hong Kong, which is a self-help organization for the laryngectomees in Hong Kong. TE speakers were fitted with Provox valve, and EL speakers used Servox-type electrolarynx. Speech samples were digitized at 20 kHz and 16 bits/sample by using Praat, based on which LTAS contours were developed. First spectral peak (FSP), mean spectral energy (MSE), and spectral tilt (ST) derived from the LTAS contours associated with different speaker groups were compared. Results: Data revealed all speakers generally exhibited similar LTA contours. However, PA speakers exhibited the lowest average FSP value and the greatest average MSE value. NL phonation was associated with a significantly greater ST value than alaryngeal speech of Cantonese. Conclusion: The differences in FSP, MSE, and ST values in different speaker groups may be related to the different sound sources being used by the laryngectomees, and the difference in the way the sound source is coupled with the vocal tract system. © 2009 Elsevier Ireland Ltd. An interactive speech training system with virtual reality articulation for Mandarin-speaking hearing impaired children
The present project involved the development of a novel interactive speech training system based on virtual reality articulation and examination of the efficacy of the system for hearing impaired (HI) children. Twenty meaningful Mandarin words were presented to the HI children via a 3-D talking head during articulation training. Electromagnetic Articulography (EMA) and graphic transform technology were used to depict movements of various articulators. In addition, speech corpuses were organized in listening and speaking training modules of the system to help improve language skills of the HI children. Accuracy of virtual reality articulatory movement was evaluated through a series of experiments. Finally, a pilot test was performed to train two HI children using the system. Preliminary results showed improvement in speech production by the HI children, and the system was recognized as acceptable and interesting for children. It can be concluded that the training system is effective and valid in articulation training for HI children. © 2013 IEEE.published_or_final_versio
Nadph oxidase upregulated by AT1 receptor mediates chronic intermittent hypoxia-induced oxidative stress and inflammation in rat adrenal medulla
Poster PresentationOur previous study found that chronic intermittent hypoxia (CIH) associated with recurrent apnea induced
oxidative stress and inflammation in rat adrenal medulla. However, the underline mechanism was not
clear. We hypothesized that, under CIH, the up-regulation of NADPH oxidase mediated by
renin-angiotensin system (RAS) via an activation of angiotensin II receptor 1 (AT1) might take part in the
oxidative stress and local inflammation in the adrenal medulla. Adult male SD rats were exposed to air
(normoxic) control or CIH treatment (8 hours/day) which mimicked a severe recurrent sleep apneic
condition for 14 days. Oral feeding of Telmisartan (10 mg/kg), a specific AT1 receptor blocker, or an
intraperitoneal injection of apocynin (25 mg/kg i.p.), an inhibitor of NADPH oxidase, or vehicle was
performed before the daily hypoxic treatment. The adrenal medulla was harvested for the measurement of
markers for oxidative stress (MDA and NTR), macrophages infiltration (ED1), apoptosis, and
inflammation (pro-inflammatory mediators) using TUNEL assay, real-time PCR, ELISA and Western blot.
Levels of MDA and NTR were significantly increased in the hypoxic (CIH) group when compared with
the normoxic control, but were normalized in the hypoxic groups treated with apocynin (AIH) or
telmisartan (TIH). The expression levels of macrophage marker ED1-immunoreactivity and the
pro-inflammatory mediators (TNFa, IL6) were also elevated in the CIH group, but were significantly
ameliorated by the apocynin or telmisartan treatment. In addition, the amount of apoptotic cells in the CIH
group was significantly higher than that of the AIH and TIH groups. Moreover, the mRNA levels of
NADPH oxidase subunits (Nox2, Nox4) were increased significantly in the CIH group when compared
with that of the AIH and TIH groups. Also, the protein expression of RAS components (AGT, AT1) was
also increased in the CIH group. In conclusion, we showed that an up-regulation of NADPH oxidase via
AT1 receptor activation mediates CIH-induced oxidative stress and inflammation in rat adrenal medulla.published_or_final_versio
Effects of stitching on delamination of satin weave carbon-epoxy laminates under mode I, mode II and mixed-mode I/II loadings
The objective of the present study is to characterize the effect of modified chain stitching on the delamination growth under mixed-mode I/II loading conditions. Delamination toughness under mode I is experimentally determined, for unstitched and stitched laminates, by using untabbed and tabbed double cantilever beam (TDCB) tests. The effect of the reinforcing tabs on mode I toughness is investigated. Stitching improves the energy release rate (ERR) up to 4 times in mode I. Mode II delamination toughness is evaluated in end-notched flexure (ENF) tests. Different geometries of stitched specimens are tested. Crack propagation occurs without any failure of stitching yarns. The final crack length attains the mid-span or it stops before and the specimen breaks in bending. The ERR is initially low and gradually increases with crack length to very high values. The mixedmode delamination behaviour is investigated using a mixed-mode bending (MMB) test. For unstitched specimens, a simple mixed-mode criterion is identified. For stitched specimens, stitching yarns do not break during 25% of mode I ratio tests and the ERR increase is relatively small compared to unstitched values. For 70% and 50% of mode I ratios, failures of yarns are observed during crack propagation and tests are able to capture correctly the effect of the stitching: it clearly improves the ERR for these two mixed modes, as much as threefold
Drawing-Based Automatic Dementia Screening Using Gaussian Process Markov Chains
Screening tests play an important role for early detection of dementia. Among those widely used screening tests, drawing tests have gained much attention in clinical psychology. Traditional evaluation of drawing tests totally relies on the appearance of drawn picture, but does not consider any time-dependent behaviour. We demonstrated that the processing speed and direction can reflect the decline of cognitive function, and thus may be useful for disease screening. We proposed a model of Gaussian process Markov chains (GPMC) to study the complex associations within the drawing data. Specifically, we modeled the process of drawing in a state-space form, where a drawing state is composed of drawing direction and velocity with consideration of the processing time. For temporal modeling, our scope focused more on discrete-time Markov chains on continuous state space. Because of the short processing time of picture drawing, we applied higher-order of Markov chains to model long-term temporal correlation across drawing states. Gaussian process regression was used for universal function approximation to flexibly infer the state transition function. With Gaussian process prior to the distribution of function space, we could encode high-level function properties such as noisiness, smoothness and periodicity. We also derived an efficient training mechanism for complex Gaussian process regression on bivariate Markov chains. With GPMC, we present an optimal decision rule based on Bayesian decision theory. We applied our proposed method to a drawing test for dementia screening, i.e. interlocking pentagon-drawing test. We tested our models with 256 subjects who are aged from 65 to 95. Finally, comparing to the traditional methods, our models showed remarkable improvement in drawing test for dementia screening
