    A Comprehensive Evaluation of the DFP Method for Geometric Constraint Solving Algorithm Using PlaneGCS

    The development of open-source geometric constraint solvers is a pressing research topic, as commercially available solvers may not meet the research requirements. In this paper, we examine the use of numerical methods in PlaneGCS, an open-source geometric constraint solver within the FreeCAD CAD software. Our study focuses on PlaneGCS\u27s constraint solving algorithms and the three built-in single-subsystem solving methods: BFGS, LM, and Dogleg. Based on our research results, the DFP method was implemented in PlaneGCS and was successfully verified in FreeCAD. To evaluate the performance of the algorithms, we used the solving state of the constraint system as a test criterion, and analysed their solving time, adaptability, and number of iterations. Our results highlight the performance differences between the algorithms and provide empirical guidance for selection of constraint solving algorithms and research based on open-source geometric constraint solvers

    Towards an Open-Source Industry CAD: A Review of System Development Methods

    Due to the industry knowledge barrier, general computer aided design (CAD) software cannot do everything in digital manufacturing by itself, and industry CAD, therefore, occupies a crucial position in the CAD industry. To develop industry CAD smoothly, open-source is the best choice. We analyzed recent examples of industry CAD development and divided the development methods into four types: development based on the graphics development environment, development based on geometric modelling kernel, secondary development based on general CAD, and hybrid development. We analyzed the characteristics of various methods and believe that the method based on the hybrid development of the geometric modelling kernel and the graphics development environment is the best open-source industry CAD development method. We proposed a system architecture of open-source industry CAD for reference and conducted a preliminary exploration of the reference architecture to verify its feasibility

    Transformational leadership and trust in leadership impacts on employee commitment

    Purpose -This study explores the effects of leadership style and trust in leadership on employees' affective commitment in the context of a global pandemic. Design/methodology/approach -A total of 580 valid questionnaires were collected online, targeting hospitality and tourism employees working from home during a particular period of the COVID-19 Coronavirus crisis. Structural equation modeling was used to analyze the data with AMOS software. Findings -The findings indicated that perceived transformational leadership was a positive predictor of trust in leadership and affective commitment. In addition to the positive contribution to commitment, trust in leadership also mediated the relationship between transformational leadership and organizational commitment. Originality/value -The current study contributes to the literature on leadership and organizational commitment. The results of this study may provide a valuable guide to organizations, leaders, and young employees

    Clinical Application of SARS-CoV-2 IgM and IgG Antibody Detection Using the Colloidal Gold Immunochromatography Assay

    The COVID-19 pandemic, which was caused by SARS-CoV-2, has had a significant effect on global public health, economies, and societies worldwide. Serum antibody testing is a critical method for the diagnosis of COVID-19 and can complement RT-PCR in the diagnosis of COVID-19 patients; however, the performance of rapid antibody assays in the clinical setting has not been established. Rapid antibody assays were evaluated by investigating 296 COVID-19-positive individuals and 542 negative individuals confirmed by clinical diagnosis. The clinical diagnostic results were used as controls to evaluate the sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV), kappa, and 95% confidence interval (CI) of the rapid tests. IgM-positivity had a sensitivity of 86.1% and specificity of 99.1%. IgG-positivity had a sensitivity of 86.5% and specificity of 98.7%. The sensitivity of combined IgM- and IgG-positivity in clinically confirmed patients was 73.1% in the early stage (1-7 days after symptom onset) and reached 99% 15 days after symptom onset. The concordance between rapid antibody-positive tests and clinical diagnosis-positivity had a kappa value of 0.93. In addition, the false-positive rate of IgM and IgG combined nucleic acid detection was 30% in the early stage. The combined use of IgM and IgG could serve as a more suitable alternative detection method for patients with COVID-19. The rapid antibody test can be considered as an excellent supplementary approach for detecting SARS-CoV-2 in clinical application

    The Immungenicity and Cross-Neutralizing Activity of Enterovirus 71 Vaccine Candidate Strains

    This study aimed to evaluate enterovirus 71 (EV-A71) vaccine candidate strains, including their genotypes, immunogenicity and cross-neutralization capacity. From clinical samples, EV-A71 strains were separated by using Vero cells. Six strains were chosen for vaccine candidates, and the sequences were analyzed. To detect the immunogenicity of the strains, we used them to immunize NIH mice at 0 and 14 days. Cytopathic effects (CPE) were examined to determine the EV-A71 neutralizing antibody (NTAb) titer 14 d after the first and second inoculations. To evaluate the cross-neutralizing capacity of the EV-A71 vaccine candidate strains, we tested serum immunized mice with ten EV-A71 genotype strains. Six EV-A71 vaccine candidate strains were identified, all belonging to sub-genotype C4, the prevalent genotype in China. The sequence similarity of the VP1 regions of the six candidate vaccine strains and three approved inactivated vaccines was 97.58%–97.77%, and the VP1 amino acid similarity was 98.65%–99.33%. Experiments were performed to evaluate the immunogenicity and cross-neutralizing activity of the EV-A71 vaccine candidate strains. The strains had good immunogenicity 14 d after two immunizations, inducing an NTAb titer ranging from 1:94 to 1:346. The NTAb seroconversion rates 14 d after one immunization were above 80% (except HB0007), and significantly increased immunogenicity of EV-A71 strains was observed post-inoculation. Furthermore, our candidate vaccine strains had broad cross-neutralizing activity after challenge with ten sub-genotypes of EV-A71. The highest NTAb titer/lowest NTAb titer ratios of sera against EV-A71 sub-genotypes were 8.0 (JS0002), 8.0 (JS0005), 21.3 (HB0005), 21.3 (HB0007), 10.7 (HB0040) and 8.0 (GD0002), respectively. Our EV-A71 strains had good immunogenicity and cross-neutralization activity, and have the potential to serve as vaccine strains for multivalent hand, foot and mouth disease vaccines

    Short-term dietary choline supplementation alters the gut microbiota and liver metabolism of finishing pigs

    Choline is an essential nutrient for pig development and plays a role in the animal's growth performance, carcass characteristics, and reproduction aspects in weaned pigs and sows. However, the effect of choline on finishing pigs and its potential regulatory mechanism remains unclear. Here, we feed finishing pigs with 1% of the hydrochloride salt of choline, such as choline chloride (CHC), under a basic diet condition for a short period of time (14 days). A 14-day supplementation of CHC significantly increased final weight and carcass weight while having no effect on carcass length, average backfat, or eye muscle area compared with control pigs. Mechanically, CHC resulted in a significant alteration of gut microbiota composition in finishing pigs and a remarkably increased relative abundance of bacteria contributing to growth performance and health, including Prevotella, Ruminococcaceae, and Eubacterium. In addition, untargeted metabolomics analysis identified 84 differently abundant metabolites in the liver between CHC pigs and control pigs, of which most metabolites were mainly enriched in signaling pathways related to the improvement of growth, development, and health. Notably, there was no significant difference in the ability of oxidative stress resistance between the two groups, although increased bacteria and metabolites keeping balance in reactive oxygen species showed in finishing pigs after CHC supplementation. Taken together, our results suggest that a short-term supplementation of CHC contributes to increased body weight gain and carcass weight of finishing pigs, which may be involved in the regulation of gut microbiota and alterations of liver metabolism, providing new insights into the potential of choline-mediated gut microbiota/metabolites in improving growth performance, carcass characteristics, and health

    Comparative Analysis of Human Protein-Coding and Noncoding RNAs between Brain and 10 Mixed Cell Lines by RNA-Seq

    In their expression process, different genes can generate diverse functional products, including various protein-coding or noncoding RNAs. Here, we investigated the protein-coding capacities and the expression levels of their isoforms for human known genes, the conservation and disease association of long noncoding RNAs (ncRNAs) with two transcriptome sequencing datasets from human brain tissues and 10 mixed cell lines. Comparative analysis revealed that about two-thirds of the genes expressed between brain and cell lines are the same, but less than one-third of their isoforms are identical. Besides those genes specially expressed in brain and cell lines, about 66% of genes expressed in common encoded different isoforms. Moreover, most genes dominantly expressed one isoform and some genes only generated protein-coding (or noncoding) RNAs in one sample but not in another. We found 282 human genes could encode both protein-coding and noncoding RNAs through alternative splicing in the two samples. We also identified more than 1,000 long ncRNAs, and most of those long ncRNAs contain conserved elements across either 46 vertebrates or 33 placental mammals or 10 primates. Further analysis showed that some long ncRNAs differentially expressed in human breast cancer or lung cancer, several of those differentially expressed long ncRNAs were validated by RT-PCR. In addition, those validated differentially expressed long ncRNAs were found significantly correlated with certain breast cancer or lung cancer related genes, indicating the important biological relevance between long ncRNAs and human cancers. Our findings reveal that the differences of gene expression profile between samples mainly result from the expressed gene isoforms, and highlight the importance of studying genes at the isoform level for completely illustrating the intricate transcriptome

    Notch-RBPJ Pathway for the Differentiation of Bone Marrow Mesenchymal Stem Cells in Femoral Head Necrosis

    Femoral head necrosis (FHN) is a common leg disease in broilers, resulting in economic losses in the poultry industry. The occurrence of FHN is closely related to the decrease in the number of bone marrow mesenchymal stem cells (BMSCs) and the change in differentiation direction. This study aimed to investigate the function of differentiation of BMSCs in the development of FHN. We isolated and cultured BMSCs from spontaneous FHN-affected broilers and normal broilers, assessed the ability of BMSCs into three lineages by multiple staining methods, and found that BMSCs isolated from FHN-affected broilers demonstrated enhanced lipogenic differentiation, activated Notch-RBPJ signaling pathway, and diminished osteogenic and chondrogenic differentiation. The treatment of BMSCs with methylprednisolone (MP) revealed a significant decrease in the expressions of Runx2, BMP2, Col2a1 and Aggrecan, while the expressions of p-Notch1/Notch1, Notch2 and RBPJ were increased significantly. Jagged-1 (JAG-1, Notch activator)/DAPT (γ-secretase inhibitor) could promote/inhibit the osteogenic or chondrogenic ability of MP-treated BMSCs, respectively, whereas the differentiation ability of BMSCs was restored after transfection with si-RBPJ. The above results suggest that the Notch-RBPJ pathway plays important role in FHN progression by modulating the osteogenic and chondrogenic differentiation of BMSCs
