    Toxicity of microcystins in the isolated hepatocytes of common carp (Cyprinus carpio L.)

    The toxicity of hepatotoxic microcystins produced mainly by Microcystis aeruginosa in mammals and fishes was well studied in recent years. However, there were scarcely reports in toxic effects of microcystins on isolated hepatocytes of fishes, especially investigation of microcystin-induced apoptosis and/or necrosis in carp hepatocytes. In the present study, the isolated hepatocytes of common carp were exposed to various concentrations of microcystins (0.01, 0.1, 1, 10, 100, 1000 mu g L-1) for 2, 4, 8, 16 and 24 h, respectively, and cytotoxicity of microcystins in the toxin-treated cells was determined. Results of this study showed that cytotoxicity of microcystins on carp hepatocytes was time and dose-dependent, and the approximate LC50 of microcystins in carp hepatocytes was 169.2 mu g L-1. The morphological changes typical of apoptosis, such as blebbing of cell membrane, condensation and fragmentation of cell nucleus were observed in the hepatocytes exposed to microcystins (1, 10 and 100 mu g L-1) using fluorescence and differential interference contrast microscopy. Agarose gel electrophoresis of DNA demonstrated a typical apoptotic "ladder pattern" in microcystin-treated hepatocytes after 16 h of exposure. Results of the present study indicated that the form of cell death in microcystin-treated hepatocytes depend on the exposure dose of toxin. When lower concentration of microcystins (10 and 100 mu g L-1) was used for exposure, carp hepatocytes died in apoptosis while, when higher one used (1000 mu g L-1), they died in the form of necrosis.     Establishment of a Multiplex Real-Time PCR Method for Detection of Four Plant-Derived Components and Its Application to Edible Starches

    A multiplex real-time polymerase chain reaction (PCR) method for simultaneous and rapid detection of components derived from sweet potato, cassava, potato, and corn was established. The g3pdhs gene of sweet potato and cassava, the UGPase gene of potato and the zSSIIb gene of corn were used as target genes to design specific primers and TaqMan probe, and the 18S rRNA gene was used as the internal reference gene. Methodological validation was performed, and the proposed method was applied to simulated samples with different mixing ratios and actual starch samples. The results showed that this method was high-throughput, sensitive and specific. No cross-reaction was found with 15 non-target sources. The detection sensitivity for template DNA was 3 × 10-3 ng/μL, and the proposed method had a good linear relationship and high amplification efficiency. The detection limit for edible starch samples was 0.1%. The results of the PCR method for 50 actual samples were consistent with those of the reference method, indicating that this method could be used for the detection of common adulterants in edible starches

    Case Report: A long-term survival case of diffuse large B-cell lymphoma with left ventricular infiltration and spinal cord compression

    BackgroundDiffuse large B-cell lymphoma (DLBCL) is the most common subtype of non-Hodgkin lymphoma and may occur with lymph node and/or extranodal involvement. However, DLBCL with intracardiac mass is exceedingly rare. In the reported literature, the intracardiac infiltration of DLBCL mostly involves the right ventricle. Lymphoma that invades the heart has an aggressive nature, with symptoms that are easily ignored initially and can lead to multiple complications in severe cases, resulting in a poor prognosis. Early screening and diagnosis may significantly improve the survival rate. Early diagnosis may significantly improve outcomes.Case summaryWe presented a 68-year-old woman with back pain. PET/CT suggested increased FDG metabolism in the left ventricle, right adrenal gland, right erector spinae intramuscularis, multiple bones and multiple lymph nodes. Contrast-enhanced ultrasound showed a left ventricular apical mass with ventricular septum thickening. Cardiac MRI suggested a 1.6*1.1*2.1 cm mass in the apical-central portion of the left ventricle. Biopsy of the right neck mass confirmed the pathologic diagnosis of diffuse large B-cell lymphoma. However, before the pathologic diagnosis was confirmed, the patient was paralyzed due to spinal cord compression caused by the progression of bone metastases. Subsequently, pathology confirmed the diagnosis of diffuse large B-cell lymphoma, and rituximab plus cyclophosphamide, doxorubicin, vincristine, and prednisone (R-CHOP) was treated immediately as first-line therapy. In addition, glucocorticoids and mannitol dehydration were administered to relieve the symptoms of spinal cord compression. After 8 cycles of R-CHOP, the tumor at all sites had almost complete regression. The patient was able to walk normally and had no tumor-related symptoms.ConclusionsWe present a case of DLBCL with a very high tumor load that involved multiple organs, including the left ventricle, but exhibited no cardiac-related symptoms. The combination of various imaging modalities is valuable for the diagnosis of cardiac infiltration. The mass in the left ventricle almost completely regressed after R-CHOP treatment, and no recurrence has occurred in the 5 years of follow-up so far

    Characterization of manufacturing-induced surface scratches and their effect on laser damage resistance performance of diamond fly-cut KDP crystal

    Manufacturing-induced defects have drawn more and more attentions to improve the laser damage resistance performance of KDP crystal applied in high-power laser systems. Here, the morphology of surface scratches on diamond fly-cut KDP crystal is characterized and their effect on the laser damage resistance is theoretically and experimentally investigated. The results indicate that surface scratches could lower laser-induced damage threshold (LIDT) by modulating incident lasers and producing resultant local light intensifications. The induced maximum light intensity enhancement factors (LIEFs) are dependent on scratch shapes and dimensions. The diffraction effects originating from scratch edges are responsible for the strongest light intensification. Even for ultra-precision finished KDP surface with scratches that well satisfy the currently applied scratch/dig specification, the induced LIEFs are quite high, indicating that the actual defect dimension allowance should be amended and specified according to the defect-induced LIEFs. The effect of scratches on laser damage resistance is experimentally verified by the tested LIDT, which is approximately consistent with the simulation one. The morphologies of laser damage sites further confirm the role of scratches in lowering LIDT. This work could offer new perspective and guidance for fully evaluating the performance of ultra-precision manufactured optical materials applied in high-power laser facilities
