    With the rise of smartphones and tablets, travelers now have remarkable connectivity that allows them to make travel arrangements and share information before, during, and after their trips. A 2014 survey by the Pew Research Center found that 58 percent of adults in the United States owned a smartphone.1 With that rapid expansion of mobile devices, one concern is that the hospitality industry might not be ready for the level of mobile accessibility and functionality that customers expect.     PhDProperties of Escherichia coli considered to be of importance in overcoming host defence mechanisms against urinary tract infection (OTI) were investigated. These were: 1) 0 and H serotype 2) K antigen content 3) Sensitivity to the bactericidal activity of human serum 4) Haemolysin production 5) Fimbriae production 6) Fermentation of sucrose, salicin and dulcitol 7) Sensitivity to serine, spermine and urea 8) Growth requirements 9) Mucinase production Ecoli strains isolated from normal subjects and patients attending the Nephrourological Clinic at St. Bartholomew's Hospital because of known or suspected UTI, were studied. Strains isolated from urines more frequently belonged to 0 serogroups 2,4,6,8,18ab and 75, had high K antigen titres, were haemolytic and fimbriate, and fermented salicin than periurethral strains from normal subjects. These findings support the concept of "special pathogenicity", that certain strains are more invasive for the urinary tract than others. Strains rich in these "pathogenic properties" were rarely isolated from normal subjects but were significantly more frequently isolated from periurethral swabs of patients. Periurethral strains from symptomatic, abacteriuric (urethral syndrome) patients were similar to those from bacteriuric patients when they were between infections. Previous work has not implicated bacteria in the aetiology of most cases of this disease and this finding remains unexplained. Strains isolated from the upper tracts of patients undergoing localisation tests more frequently exhibited pathogenic properties than those isolated from only the lower tract, and this was considered to reflect the superior ability of these strains to reach the upper tract or better combat host defence mechanisms

    Polarized P-glycoprotein expression by the immortalised human brain endothelial cell line, hCMEC/D3, restricts apical-to-basolateral permeability to rhodamine 123

    P-glycoprotein (P-gp) expression at the blood-brain barrier prevents unwanted blood-borne toxins and signalling molecules from entering the brain. Primary and immortalised human brain endothelial cells (BECs) represent two suitable options for studying P-gp function in vitro. The limited supply of primary human BECs and their instability over passage number makes this choice unattractive for medium/high throughput studies. The aim of this study was to further characterise the expression of P-gp by an immortalised human BEC line, hCMEC/D3, in order to evaluate their use as an in vitro human blood-brain barrier model. P-gp expression was stable over a high passage number (up to passage 38) and was polarised on the apical plasma membrane, consistent with human BECs in vivo. In addition, hCMEC/D3 cell P-gp expression was comparable, albeit slightly lower to that observed in primary isolated human BECs although P-gp function was similar in both cell lines. The P-gp inhibitors tariquidar and vinblastine prevented the efflux of rhodamine 123 (rh123) from hCMEC/D3 cells, indicative of functional P-gp expression. hCMEC/D3 cells also displayed polarised P-gp transport, since both tariquidar and vinblasine selectively increased the apical-to-basolateral permeability of hCMEC/D3 cells to rh123. The results presented here demonstrate that hCMEC/D3 cells are a suitable model to investigate substrate specificity of P-gp in BECs of human origin

    The Feasibility and Impact of Delivering a Mind-Body Intervention in a Virtual World

    Introduction: Mind-body medical approaches may ameliorate chronic disease. Stress reduction is particularly helpful, but face-to-face delivery systems cannot reach all those who might benefit. An online, 3-dimensional virtual world may be able to support the rich interpersonal interactions required of this approach. In this pilot study, we explore the feasibility of translating a face-to-face stress reduction program into an online virtual setting and estimate the effect size of the intervention. Methods and Findings: Domain experts in virtual world technology joined with mind body practitioners to translate an existing 8 week relaxation response-based resiliency program into an 8-week virtual world-based program in Second Lifeâ„¢ (SL). Twenty-four healthy volunteers with at least one month's experience in SL completed the program. Each subject filled out the Perceived Stress Scale (PSS) and the Symptom Checklist 90- Revised (SCL-90-R) before and after taking part. Participants took part in one of 3 groups of about 10 subjects. The participants found the program to be helpful and enjoyable. Many reported that the virtual environment was an excellent substitute for the preferred face-to-face approach. On quantitative measures, there was a general trend toward decreased perceived stress, (15.7 to 15.0), symptoms of depression, (57.6 to 57.0) and anxiety (56.8 to 54.8). There was a significant decrease of 2.8 points on the SCL-90-R Global Severity Index (p<0.05). Conclusions: This pilot project showed that it is feasible to deliver a typical mind-body medical intervention through a virtual environment and that it is well received. Moreover, the small reduction in psychological distress suggests further research is warranted. Based on the data collected for this project, a randomized trial with less than 50 subjects would be appropriately powered if perceived stress is the primary outcome

    This thesis is about my poetry and how I use poetry as an emotional release. I will debate the notion that self-published poetry is not as prestigious as works published by university or commercial presses. To secure this debate I will compare my poetry to that of famous poets, such as Shakespeare, Dickinson, and Rilke and show the different ways poetry can be approached and judged. Then I will use samples of my poetry and relate them to psychological theories. Theories related to grief and self-actualization will show the gravity of my poetry. This, in tum, will show how poetry can be used for therapeutic purposes, as well as strengthen the argument pertaining to the emotional appeal my poetry has

    Estimating critical habitat based on year-round movements of the endangered Jefferson salamander (Ambystoma jeffersonianum) and their unisexual dependents

    Habitat protection is a key component of endangered species conservation but critical habitat designations are often based on limited data or habitat use during only a portion of a species’ life cycle. Protected habitat around breeding pools for the endangered Jefferson salamander (Ambystoma jeffersonianum (Green, 1827)) and their unisexual dependants (Ambystoma laterale - (2) jeffersonianum) is based upon limited movement data from primarily spring and summer seasons. Furthermore, despite their treatment as distinct species under Canada’s Species at Risk Act, differences in habitat use have not been directly compared in areas where they co-occur. We used radio telemetry to track A. jeffersonianum and A. laterale – jeffersonianum (LJJ and LJJJ) during fall migrations to overwintering sites. We used these data and summarized available movement data from past studies that tracked movements in other periods of the annual cycle to estimate year-round critical habitat. A. jeffersonianum travelled significantly longer distances to overwintering locations than unisexuals. Individuals were more likely to overwinter next to a similar genomotype individual than not. Critical habitat encompassing the entire annual cycle of A. jeffersonianum extends up to 400-450 m from breeding ponds indicating existing regulatory habitat protections in Canada do not currently protect sufficient habitat.The accepted manuscript in pdf format is listed with the files at the bottom of this page.
