    Anti-tumor effect of polysaccharides from rhizome of Curculigo orchioides Gaertn on cervical cancer

    Purpose: To investigate the anti-tumor effects of polysaccharides from Curculigo orchioides (PDC) on cervical cancer and the possible mechanisms involved.Methods: A Box–Behnken design (BBD) was employed to optimize extraction conditions for PDC. The anti-tumor effect of PDC on cervical cancer was investigated in vivo in mice injected with Hela cells. The parameters measured were tumor volume and weight. In vitro anti-tumor effects of PDC were assessed by measuring expressions of caspase-3, caspase-9 and P53 proteins in Hela cells via ELISA assay. Thymus and spleen indices were calculated for assessment of PDC effect on immune function.Results: The optimum extraction conditions predicted by the response surface methodology (RSM) were: extraction time = 1.58 h, ratio-of-water-to-sample = 30.05 mL/g and extraction number = 1.95. PDC showed significant anti-tumor effect on cervical cancer in mice. It significantly increased thymus and spleen indices in mice; and significantly up-regulated expressions of caspase-3, caspase-9 and P53 proteins in HeLa cells.Conclusion: PDC has significant anti-tumor effect on cervical cancer in vivo and in vitro, most probably through mechanisms involving enhancement on immune function and induction of apoptosis.Keyword: Curculigo orchioides, Polysaccharides, Cervical cancer, HeLa cells, Apoptosi

    Cloning and Expression Analysis of a PISTILLATA Homologous Gene from Pineapple (Ananas comosus L. Merr)

    PISTILLATA (PI)-like genes are crucial regulators of flowering in angiosperms. A homologue of PI, designated as AcPI (Genbank accession number HQ717796), was isolated from pineapple cultivar Comte de Paris by reverse transcriptase polymerase chain reaction (RT-PCR) and rapid amplification of cDNA ends (RACE). The cDNA sequence of AcPI is 907 bp in length and contains an open reading frame of 594 bp, which encodes a protein of 197 amino acids. The molecular weight was 2.29 kDa and the isoelectric point was 9.28. The alignment showed that AcPI had a high identity with CsPIC2 (78.6%), AoPI (77.4%), OrcPI (75.7%) and HPI2 (72.4%). Quantitative real-time polymerase chain reaction (qRT-PCR) analyses in different tissues showed that the expression pattern of AcPI was different from the B-class genes in eudicots. AcPI was expressed in all the tissues investigated. The expression level was very low in fruit stems, bracts, leaves and sepals, high in petals and carpels, and moderate in apical meristems, flesh and stamens. The qRT-PCR analyses in different stages indicated that the expression of AcPI reached the highest level at 40 days after flower inducement, when the multiple fruit and floral organs were forming. It proved the important role of AcPI in floral organs and fruit development. The 35S::AcPI transgenic Arabidopsis plants flowered earlier and had more inflorescences or branches than wild type plants

    Antifungal effects of sisal leaf juice on Lasiodiplodia theobromae, the causal agent of mulberry root rot

    This study was carried out to evaluate the antifungal activities of leaf juices (fresh juice, fermented juice, boiled juice and sterile juice) of nine sisal varieties on Lasiodiplodia theobromae, the causal agent of mulberry root rot. Results show that all the leaf juices could inhibit the mycelial growth in different degrees (the inhibitory rates ranged from 63.3 to 100%), due to different varieties and treatments. Among the nine varieties, the inhibition effects of hybrid 76416 and Agave americana were the best with absolute inhibition of all the leaf juice treatments against the mycelial growth, followed by Agave Amaniensis, Agave virdis, Agave angustifolia and Hybrid 11648. The inhibitory effect of some fresh juices would be cut down after being fermented, boiled and sterilized. The treated mycelia of L.theobromae were malformed, enlarged, broken and plasma leaked when observed under the microscope. Most of the leaf juices could inhibit the conidial germination absolutely, except A.amaniensis, H.11648 and A. angustifolia. The average germination rate of A. amaniensis, H.11648 and A. angustifolia was 72.4, 16.6 and 13%, respectively. The control efficiency of the fresh juice of H. 11648 against mulberry root rot in the field reached 73.1%.Key words: Sisal, leaf juice, anti-fungi, anti-fungal activities, mulberry root rot, Lasiodiplodia theobromae

    Effect of rosiglitazone on rabbit model of myocardial ischemia-reperfusion injury

    AbstractObjectiveTo explore mechanism and protective effect of rosiglitazone on myocardial ischemia reperfusion (I/R) injury.MethodsA total of 48 male Japanese white big-ear rabbits were randomly divided into control group (A), I/R group (B), low dose of rosiglitazone group (C), high dose of rosiglitazone group (D). Plasma concentration of and also reduced the concentration of plasma serum creatine kinase (CK), CK-MB, high-sensitivity C-reactive protein (hsCRP), ultra-superoxide dismutase (SOD), malondialdehyde (MDA), lactic acid glutathione skin peroxidase (GSH-PX), nitric oxide (NO) and endothelin (ET) were measured 1 h later after I/R. Twenty-four hours after I/R the hearts were harvested for pathological and ultrastructural analysis. Area of myocardial infarction were tested.ResultsPlasma concentration of CK, CK-MB, hsCRP, NO, MDA and ET were decreased in C, D group compared with group B. Plasma concentration of T-SOD and GSH-Px were increased significantly in C, D group compared with group B. Compared with group B, pathological and ultrastructural changes in C and D group were slightly. There was significant difference in myocardial infarction area between group C, D and group B (P<0.05). Myocardial infarction area and arrhythmia rate were lower in group C, D compare with group B.ConclusionsRosiglitazone may protect myocardium from I/R injury by enhancing T-SOD and GSH-Px concentration, inhibit inflammatory reaction, and improve endothelial function

    Safety and efficacy of etomidate and propofol anesthesia in elderly patients undergoing gastroscopy: A double-blind randomized clinical study

    The aim of the present study is to compare the safety, efficacy and cost effectiveness of anesthetic regimens by compound, using etomidate and propofol in elderly patients undergoing gastroscopy. A total of 200 volunteers (65–79 years of age) scheduled for gastroscopy under anesthesia were randomly divided into the following groups: P, propofol (1.5–2.0 mg/kg); E, etomidate (0.15-0.2 mg/kg); P+E, propofol (0.75–1 mg/kg) followed by etomidate (0.075-0.1 mg/kg); and E+P, etomidate (0.075-0.01 mg/kg) followed by propofol (0.75–1 mg/kg). Vital signs and bispectral index were monitored at different time points. Complications, induction and examination time, anesthesia duration, and recovery and discharge time were recorded. At the end of the procedure, the satisfaction of patients, endoscopists and the anesthetist were evaluated. The recovery (6.1±1.2 h) and discharge times (24.8±2.8 h) in group E were significantly longer compared with groups P, P+E and E+P (P<0.05). The occurrence of injection pain in group P+E was significantly higher compared with the other three groups (P<0.05). In addition, the incidence of myoclonus and post-operative nausea and vomiting were significantly higher in group P+E compared with the other three groups (P<0.05). There was no statistical difference among the four groups with regards to the patients' immediate, post-procedure satisfaction (P>0.05). Furthermore, there was no difference in the satisfaction of anesthesia, as evaluated by the anesthetist and endoscopist, among the four groups (P>0.05). The present study demonstrates that anesthesia for gastroscopy in elderly patients can be safely and effectively accomplished using a drug regimen that combines propofol with etomidate. The combined use of propofol and etomidate has unique characteristics which improve hemodynamic stability, cause minimal respiratory depression and less side effects, provide rapid return to full activity and result in high levels of satisfaction

    Increased serum myeloid-related protein 8/14 level is associated with atherosclerosis in type 2 diabetic patients

    <p>Abstract</p> <p>Background</p> <p>Myeloid-related protein 8/14 (MRP8/14) is a stable heterodimer formed by two different calcium-binding proteins (MRP8 and MRP14). Studies have identified that MRP8/14 regulates vascular inflammation and serves as a novel marker of acute coronary syndrome. In this study, we evaluated the correlation between serum levels of MRP8/14, hsCRP, endogenous secretory receptor for advanced glycation end-products (esRAGE) and the occurrence of coronary artery disease (CAD), or carotid intima-media thickness (IMT) when CAD was not yet developed in diabetic patients.</p> <p>Methods</p> <p>Serum levels of MRP8/14, esRAGE and hsCRP were measured in 375 diabetic patients. Then the results of those who had CAD were compared against who had not. Also, we investigated the associations between above-mentioned indicators and IMT of subjects without CAD in both diabetic group and non-diabetic one.</p> <p>Results</p> <p>Serum MRP8/14 was significantly higher in CAD than in non-CAD group (9.7 ± 3.6 ug/ml vs. 8.2 ± 3.0 ug/ml, <it>P </it>< 0.001). It was associated with severity of CAD (<it>r </it>= 0.16, <it>P </it>= 0.026). In non-CAD group, MRP8/14 was associated with IMT in patients with (<it>r </it>= 0.30, <it>P </it>< 0.001) or without diabetes (<it>r </it>= 0.26, <it>P </it>= 0.015). The areas under the curves of receiver operating characteristic for CAD were 0.63 (95% CI 0.57-0.68) for MRP8/14, 0.76 (95% CI 0.71-0.81) for hsCRP and 0.62 (95% CI 0.56 -0.67) for esRAGE.</p> <p>Conclusion</p> <p>In summary, we report that diabetic patients with CAD had elevated plasma MRP8/14 levels which were also positively correlated with the severity of CAD and carotid IMT in patients without clinically overt CAD.</p

    The interaction between different types of activated RAW 264.7 cells and macrophage inflammatory protein-1 alpha

    <p>Abstract</p> <p>Background</p> <p>Two major ways of macrophage (MΦ) activation can occur in radiation-induced pulmonary injury (RPI): classical and alternative MΦ activation, which play important roles in the pathogenesis of RPI. MΦ can produce chemokine MΦ inflammatory protein-1α (MIP-1α), while MIP-1α can recruit MΦ. The difference in the chemotactic ability of MIP-1α toward distinct activated MΦ is unclear. We speculated that there has been important interaction of MIP-1α with different activated MΦ, which might contribute to the pathogenesis of RPI.</p> <p>Methods</p> <p>Classically and alternatively activated MΦ were produced by stimulating murine MΦ cell line RAW 264.7 cells with three different stimuli (LPS, IL-4 and IL-13); Then we used recombinant MIP-1α to attract two types of activated MΦ. In addition, we measured the ability of two types of activated MΦ to produce MIP-1α at the protein or mRNA level.</p> <p>Results</p> <p>Chemotactic ability of recombinant MIP-1α toward IL-13-treated MΦ was the strongest, was moderate for IL-4-treated MΦ, and was weakest for LPS-stimulated MΦ (p < 0.01). The ability of LPS-stimulated MΦ to secrete MIP-1α was significantly stronger than that of IL-4-treated or IL-13-treated MΦ (p < 0.01). The ability of LPS-stimulated MΦ to express MIP-1α mRNA also was stronger than that of IL-4- or IL-13-stimulated MΦ (p < 0.01).</p> <p>Conclusions</p> <p>The chemotactic ability of MIP-1α toward alternatively activated MΦ (M2) was significantly greater than that for classically activated MΦ (M1). Meanwhile, both at the mRNA and protein level, the capacity of M1 to produce MIP-1α is better than that of M2. Thus, chemokine MIP-1α may play an important role in modulating the transition from radiation pneumonitis to pulmonary fibrosis <it>in vivo</it>, through the different chemotactic affinity for M1 and M2.</p

    Co-expression of the Arabidopsis SOS genes enhances salt tolerance in transgenic tall fescue (Festuca arundinacea Schreb.)

    Crop productivity is greatly affected by soil salinity; therefore, improvement in salinity tolerance of crops is a major goal in salt-tolerant breeding. The Salt Overly Sensitive (SOS) signal-transduction pathway plays a key role in ion homeostasis and salt tolerance in plants. Here, we report that overexpression of Arabidopsis thaliana SOS1+SOS2+SOS3 genes enhanced salt tolerance in tall fescue. The transgenic plants displayed superior growth and accumulated less Na(+) and more K(+) in roots after 350 mM NaCl treatment. Moreover, Na(+) enflux, K(+) influx, and Ca(2+) influx were higher in the transgenic plants than in the wild-type plants. The activities of the enzyme superoxide dismutase, peroxidase, catalase, and proline content in the transgenic plants were significantly increased; however, the malondialdehyde content decreased in transgenic plants compared to the controls. These results suggested that co-expression of A. thaliana SOS1+SOS2+SOS3 genes enhanced the salt tolerance in transgenic tall fescue.
