    Bile salt deconjugation and cholesterol removal from media by Lactobacillus strains used as probiotics in chickens

    BACKGROUND: Bile salt deconjugation by Lactobacillus strains is often closely linked to bile tolerance and survival of the strains in the gut and lowering of cholesterol in the host. The present study investigated the deconjugation of bile salts and removal of cholesterol by 12 Lactobacillus strains in vitro. The 12 strains were previously isolated from the gastrointestinal tract of chickens. RESULTS: The 12 Lactobacillus strains could deconjugate sodium glycocholate (GCA, 16.87-100%) and sodium taurocholate (TCA, 1.69-57.43%) bile salts to varying degrees, with all strains except L. salivarius I 24 having a higher affinity for GCA. The 12 Lactobacillus strains also showed significant (P < 0.05) differences in their ability to remove cholesterol from the growth medium (26.74-85.41%). Significant (P < 0.05) correlations were observed between cholesterol removal and deconjugation of TCA (r = 0.83) among the L. reuteri strains (C1, C10 and C16) and between cholesterol removal and deconjugation of TCA (r = 0.38) and GCA (r = 0.70) among the L. brevis strains (I 12, I 23, I 25, I 211 and I 218). In contrast, although L. gallinarum I 16 and I 26 and L. panis C 17 showed high deconjugating activity, there was no correlation between cholesterol removal and deconjugation of bile salts in these strains. CONCLUSION: The results showed that the 12 Lactobacillus strains were able to deconjugate bile salts and remove cholesterol in vitro, but not all strains with high deconjugating activity removed cholesterol effectively

    Correlation of the composite equilibrium score of computerized dynamic posturography and clinical balance tests

    AbstractBackgroundThe computerized dynamic posturography has been widely used to access balance control in patients with balance dysfunction. A composite-equilibrium score (CS) can be calculated from the sensory organization test using the computerized dynamic posturography. However, the correlation between the composite equilibrium score and clinical tests and its ability to predict falls has rarely been explored in the past.MethodsA total of 60 patients with chief complaint of dizziness were enrolled in our study, and clinical assessments were done including the sensory organization test (SOT), Timed Up and Go test (TUG), Tinetti Performance-Oriented Mobility Assessment (POMA), and the dynamic gait index (DGI). The age and the subjective feeling of the severity of dizziness quantified by the visual analog scale (VAS) of each patient were also recorded.ResultsStatistical analysis revealed significant correlation between the composite equilibrium score and the TUG, POMA (gait, balance and total scores), and the DGI. However, there is statistically significant correlation between neither the CS and the age nor the VAS of dizziness. When grouping the DGI, POMA (total score), and the TUG cutoff to predict fall risks, the correlations to the CS can still be established except the TUG.ConclusionFrom the results of our study, the validity of the clinical tests was established in assessment of balance function, and clinicians can utilize these tools for preliminary evaluation of patient balance when computerized dynamic posturography is not available.     Fermentation activities of some new species of anaerobic rumen fungi from Malaysia

    The fermentation activities of three new anaerobic rumen fungi, viz, Neocallimastix variabilis, Piromyces spiralis and P. minutus, and a Caecomyces communis isolate grown on straw and filter paper media were studied. N. variabilis, P. spiralis and P. minutus produced carboxymethylcellulase (CMCase), FPase (filter paper activity) and xylanases when grown on straw and filter paper media. C. communis produced only xylanases, The maximum production of all the enzymes was at 72 h of fungal incubation. On both media, N. variabilis showed the highest enzyme activities, followed by P. spiralis and P. minutus. Specific enzyme activities of the three fungal isolates in descending order of production were: Xylanase > CMCase > FPase when grown on straw medium; and CMCase > Xylanase > FPase > cellobiase when grown on filter paper medium. The major end-products of fermentation by the three new fungal isolates were acetate and formate. Ethanol and succinate were produced in low amounts and lactate was not detected. The absence of lactate in the end-products of fermentation observed in this study is the first such report

    Seismic Risk Assessment and Rehabilitation Method of Existing RCC Structures Using Micro Concrete

    Aging reinforced concrete (RC) building structures typically experience more severe damage and are prone to collapse during earthquakes, constituting a primary factor in casualties and direct economic losses. To enhance the seismic performance of these old structures, this paper proposes a seismic risk assessment and a micro-concrete restoration method. It applies the process to an existing three-story reinforced concrete structure. A practical framework for mitigating structural vulnerabilities in seismic-prone regions was proposed. Then an as-built survey was conducted to create as-built architectural and structural drawings. Concrete core tests, ferroscans, and rebar tests were also performed. Based on field surveys and test data, nonlinear static and dynamic analyses have been used to evaluate structural safety. Concrete column jacketing was used to strengthen weak existing columns with micro-concrete. In assessing the structural response of retrofitted buildings, a comparison was made to their initial state. The comparison shows that applying concrete column jacketing with micro concrete can reduce other structural elements' demand capacity ratio (DCR), minimize maximum displacements, and enhance overall stiffness. The results indicate that the proposed method effectively evaluates the seismic risk of aging structures and enhances seismic resilience in existing buildings.     Assessment of Heavy Metal Content and Consumption Risks At Selected Paddy Field in Malaysia: A Review

    As the Malaysian population grows, there is a high demand for rice, the main staple food in this region. This has caused the overuse of agrochemicals that contain heavy metals and the utilization of contaminated groundwater to increase paddy yield, posing a risk to humans. This study reviewed the accumulated heavy metals in paddy fields of Malaysia's Selangor, Kedah and Sabah states and further calculated the consumption risks of rice grains from the selected areas. The study revealed that paddy soil in Ranau Valley (Sabah), Kota Marudu (Sabah) and Tanjung Karang (Selangor) showed presences of Cu and Cd in high concentration, respectively, creating higher potential to be uptake by paddy roots. These findings also revealed that Ranau Valley (Sabah) paddy grains contained high Cu and Cd concentrations, while Sabak Bernam (Selangor) contained high Pb concentrations. Further, a higher Cd concentration was reported from the Ranau Valley (Sabah), while the higher Pb concentrations were reported from the samples collected from Sabak Bernam (Selangor), Tanjung Karang (Selangor) and Kubang Pasu (Kedah). Based on the health risk indices calculation in this study, carcinogenic and non-carcinogenic health risks in all study areas except in Kubang Pasu (Kedah) and Langkawi (Kedah) are likely to occur due to Cu mining activities, ultrabasic soil contamination, utilization of contaminated groundwater and rock phosphate fertilizer and vehicular emission. Regular assessment of heavy metal content and consumption risks of paddy is essential to ensure the paddy field is free from contamination and will help protect the ecosystem and human health

    Effects of polymer molecular weight on relative oral bioavailability of curcumin

This study investigated the influence of polymer molecular weight on the relative oral bioavailability of curcumin, and explored the possible mechanism accounting for the outcome.Methods: Curcumin encapsulated in low (5000&amp;ndash;15,000) and high (40,000&amp;ndash;75,000) molecular weight PLGA (LMw-NPC and HMw-NPC, respectively) were prepared using an emulsification-solvent evaporation method. Curcumin alone and in the nanoformulations was administered orally to freely mobile rats, and blood samples were collected to evaluate the bioavailability of curcumin, LMw-NPC, and HMw-NPC. An ex vivo experimental gut absorption model was used to investigate the effects of different molecular weights of PLGA formulation on absorption of curcumin. High-performance liquid chromatography with diode array detection was used for quantification of curcumin in biosamples.Results: There were no significant differences in particle properties between LMw-NPC and HMw-NPC, but the relative bioavailability of HMw-NPC was 1.67-fold and 40-fold higher than that of LMw-NPC and conventional curcumin, respectively. In addition, the mean peak concentration (Cmax) of conventional curcumin, LMw-NPC, and HMw-NPC was 0.028, 0.042, and 0.057 &amp;micro;g/mL, respectively. The gut absorption study further revealed that the HMw-PLGA formulation markedly increased the absorption rate of curcumin in the duodenum and resulted in excellent bioavailability compared with conventional curcumin and LMw-NPC.Conclusion: Our findings demonstrate that different molecular weights of PLGA have varying bioavailability, contributing to changes in the absorption rate at the duodenum.     Cloning of a novel phytase from an anaerobic rumen bacterium, Mitsuokella jalaludinii, and its expression in Escherichia coli

    The full length phytase gene of Mitsuokella jalaludinii was successfully cloned and was found to be 1047 bp in length, with 348 amino acids, and was designated as PHY7 phytase gene. A comparison of the sequence of PHY7 phytase gene of M. jalaludinii with various microbial phytase gene sequences showed that it was not similar to those from other bacteria except Selenomonas ruminatium, thus suggesting that they may both express a new class of phytase. The PHY7 phytase gene was subsequently subcloned into bacterial expression vector, pET32a, for expression in Escherichia coli strain Rosetta-gami. Expression of the recombinant phytase gene was optimized and characterized. The recombinant phytase was estimated to be approximately 55 kDa by SDS-PAGE analysis. The recombinant phytase exhibited optimum activity at 55°C, pH 4.5 and showed good pH stability from pH 3.5 to 5.5 (>78% relative activity). Metal ions such as Ca2+, Mg2+, and K+ were found to exert significant stimulatory effect on the recombinant phytase activity while Cu2+, Fe3+, and Zn2+ greatly inhibited the enzyme activity. The recombinant phytase showed moderate resistance to trypsin proteolysis, but susceptible to pepsin proteolysis. The results of the study showed that several characteristics of recombinant phytase were slightly different from the native enzyme. Unfavourable characteristics such as reduced pH stability and metal ion effects should be taken into consideration during feed enzyme formulation
