    Essential Oils: The Ultimate Solution to Antimicrobial Resistance in <i>Escherichia coli</i>?

    Antimicrobial resistance (AMR) is on the rise; the only solution for overcoming this is through accelerated drug discovery. At current, bacterial evolutionary rates is still clearly the undisputed winner in this war. To circumvent this, evolution of resistance need to be curbed and this can only be effective via novel approaches, one of which includes the use of a resistance modifying agent. The criterion to qualify as a resistance modifier necessitates the co-administration of the agent with an inhibitor that deactivates the bacterial resistance mechanism, restoring its original effectiveness. Natural products such as plant extracts and essential oils (EOs) have been viewed as a privileged group for investigation of their potential roles to combat antibiotic resistance, due to their compositions of active chemical compounds. The route for multidrug resistance development in Gram‐negative bacteria is primarily mediated by the sophisticated inner and outer membrane barriers, which function to protect the cell against external toxic compounds; hence, bypass of these bacterial membranes would successfully restore or improve efficacy of the antimicrobials. The aim of this chapter is to concisely describe some examples for recent strategies used in the screening of possible resistance modifiers from essential oils specifically against MDR Escherichia coli

    The missing piece: recent approaches investigating the antimicrobial mode of action of essential oils

    Antibiotic resistance is a major global health issue that has seen alarming rates of increase in all parts of the world over the past two decades. The surge in antibiotic resistance has resulted in longer hospital stays, higher medical costs, and elevated mortality rates. Constant attempts have been made to discover newer and more effective antimicrobials to reduce the severity of antibiotic resistance. Plant secondary metabolites, such as essential oils, have been the major focus due to their complexity and bioactive nature. However, the underlying mechanism of their antimicrobial effect remains largely unknown. Understanding the antimicrobial mode of action of essential oils is crucial in developing potential strategies for the use of essential oils in a clinical setting. Recent advances in genomics and proteomics have enhanced our understanding of the antimicrobial mode of action of essential oils. We might well be at the dawn of completing a mystery on how essential oils carry out their antimicrobial activities. Therefore, an overview of essential oils with regard to their antimicrobial activities and mode of action is discussed in this review. Recent approaches used in identifying the antimicrobial mode of action of essential oils, specifically from the perspective of genomics and proteomics, are also synthesized. Based on the information gathered from this review, we offer recommendations for future strategies and prospects for the study of essential oils and their function as antimicrobials

    Co-culture systems for the production of secondary metabolites: current and future prospects

    Microorganisms are the great sources of Natural Products (NPs); these are imperative to their survival apart from conferring competitiveness amongst each other within their environmental niches. Primary and secondary metabolites are the two major classes of NPs that help in cell development, where antimicrobial activity is closely linked with secondary metabolites. To capitalize on the effects of secondary metabolites, co-culture methods have been often used to develop an artificial microbial community that promotes the action of these metabolites. Different analytical techniques will subsequently be employed based on the metabolite specificity and sensitivity to further enhance the metabolite induction. Liquid Chromatography-Mass Spectrometry (LC-MS) and Gas Chromatography (GC)-MS are commonly used for metabolite separation while Nuclear Magnetic Resonance (NMR) and Mass Spectrometry (MS) have been used as tools to elucidate the structure of compounds. This review intends to discuss current systems in use for co-culture in addition to its advantages, with discourse into the investigation of specific techniques in use for the detailed study of secondary metabolites. Further advancements and focus on co-culture technologies are required to fully realize the massive potential in synthetic biological systems

    A review on Tradescantia : phytochemical constituents, biological activities and health-promoting effects

    Tradescantia is a genus of herbaceous and perennial plants belonging to the Commelinaceae family and organized into three infrageneric classifications and 12 sections. More than 80 species within the genus have been used for centuries for medicinal purposes. Phytochemical compounds (from various species of the genus) such as coumarins, alkaloids, saponins, flavonoids, phenolics, tannins, steroids and terpenoids have recently been characterized and described with antioxidant, cytotoxic, anti-inflammatory, anticancer or antimicrobial properties.     Middle Eastern plant extracts: an alternative to modern medicine problems

    Middle Eastern countries are primarily known for their dry sand deserts; however, they have a wider physiographic range which includes upland plateau and mountain ranges. The Middle East is home to various types of plants, such as Phoenix dactylifera (date palm tree), Scrophularia striata (herbaceous plants), and Opuntia ficus-indica (cactus). These plants have been found to have various types of bioactivities, such as antimicrobial activities against both bacteria and fungi, in addition to exhibiting anti-inflammatory effects and anti-cancer characteristics which can be utilized in the clinical setting for treatment. Due to limited reviews focusing on plant extracts from the Middle East, we aim to provide a discourse on plants from this region which have various bioactivities and to provide information on the compounds that can be identified from these plants. This is to enhance our understanding to improve modern medicine problems such as antimicrobial resistance and to find an alternative cure for cancer. It is hoped that the collation of information from this review will enable an assessment of the direct role of Middle Eastern plants in providing therapeutic options to address the predicaments in the medical field

    Lactococcus lactis has progressed a long way since its discovery and initial use in dairy product fermentation, to its present biotechnological applications in genetic engineering for the production of various recombinant proteins and metabolites that transcends the heterologous species barrier. Key desirable features of this gram-positive lactic acid non-colonizing gut bacteria include its generally recognized as safe (GRAS) status, probiotic properties, the absence of inclusion bodies and endotoxins, surface display and extracellular secretion technology, and a diverse selection of cloning and inducible expression vectors. This have made L. lactis a desirable and promising host on par with other well established model bacterial or yeast systems such as Escherichia coli, Salmonella cerevisiae and Bacillus subtilis. In this article, we review recent technological advancements, challenges, future prospects and current diversified examples on the use of L. lactis as a microbial cell factory. Additionally, we will also highlight latest medical-based applications involving whole-cell L. lactis as a live delivery vector for the administration of therapeutics against both communicable and non-communicable diseases

    Membrane disruption properties of essential oils—a double-edged sword?

    The emerging literature has suggested essential oils (EOs) as new possible weapons to fight antimicrobial resistance due to their inherent antimicrobial properties. However, the potential pharmaceutical use of EOs is confronted by several limitations, including being non-specific in terms of drug targeting, possessing a high cytotoxicity as well as posing a high risk for causing skin irritation. Furthermore, some EOs have been demonstrated to adversely affect the cellular lipid profiles and permeability of the cell membrane, which may result in undesirable outcomes for the cells. Nevertheless, owing to their naturally complex compositions, EOs still hold undiscovered potential to mitigate antimicrobial resistance, as an alternative to existing antibiotics. To address the issue of overuse in antibiotics for crops which have led to the growing threat of antimicrobial resistance globally, EOs have also been proposed as potential biopesticides. Since the perceived advantages of antimicrobial attributes in EOs remain largely unexplored, this review aims to provide a discourse into its current practical usefulness in the agricultural setting. Finally, updated bioengineering techniques with emphasis of the biopesticide potential of EOs as a means to alleviate antimicrobial resistance will be included
