    Learning statistical correlation for fast prostate registration in image-guided radiotherapy: Learning statistical correlation for radiotherapy

    Purpose: In adaptive radiation therapy of prostate cancer, fast and accurate registration between the planning image and treatment images of the patient is of essential importance. With the authors’ recently developed deformable surface model, prostate boundaries in each treatment image can be rapidly segmented and their correspondences (or relative deformations) to the prostate boundaries in the planning image are also established automatically. However, the dense correspondences on the nonboundary regions, which are important especially for transforming the treatment plan designed in the planning image space to each treatment image space, are remained unresolved. This paper presents a novel approach to learn the statistical correlation between deformations of prostate boundary and nonboundary regions, for rapidly estimating deformations of the nonboundary regions when given the deformations of the prostate boundary at a new treatment image

    Immunoproteomic analysis of outer membrane proteins and extracellular proteins of Actinobacillus pleuropneumoniae JL03 serotype 3

    <p>Abstract</p> <p>Background</p> <p><it>Actinobacillus pleuropneumoniae </it>is the causative agent of porcine contagious pleuropneumonia, a highly contagious respiratory infection in pigs, and all the 15 serotypes are able to cause disease. Current vaccines including subunit vaccines could not provide satisfactory protection against <it>A. pleuropneumoniae</it>. In this study, the immunoproteomic approach was applied to the analysis of extracellular and outer membrane proteins of <it>A. pleuropneumoniae </it>JL03 serotype 3 for the identification of novel immunogenic proteins for <it>A. pleuropneumoniae</it>.</p> <p>Results</p> <p>A total of 30 immunogenic proteins were identified from outer membrane and extracellular proteins of JL03 serotype 3, of which 6 were known antigens and 24 were novel immunogenic proteins for <it>A. pleuropneumoniae</it>.</p> <p>Conclusion</p> <p>These data provide information about novel immunogenic proteins for <it>A. pleuropneumoniae </it>serotype 3, and are expected to aid in development of novel vaccines against <it>A. pleuropneumoniae</it>.</p

    Memory-Enhancing Effects of the Crude Extract of Polygala tenuifolia on Aged Mice.

    Learning and memory disorders arise from distinct age-associated processes, and aging animals are often used as a model of memory impairment. The root of Polygala tenuifolia has been commonly used in some Asian countries as memory enhancer and its memory improvement has been reported in various animal models. However, there is less research to verify its effect on memory functions in aged animals. Herein, the memory-enhancing effects of the crude extract of Polygala tenuifolia (EPT) on normal aged mice were assessed by Morris water maze (MWM) and step-down passive avoidance tests. In MWM tests, the impaired spatial memory of the aged mice was partly reversed by EPT (100 and 200 mg/kg; P < 0.05) as compared with the aged control mice. In step-down tests, the nonspatial memory of the aged mice was improved by EPT (100 and 200 mg/kg; P < 0.05). Additionally, EPT could increase superoxide dismutase (SOD) and catalase (CAT) activities, inhibit monoamine oxidase (MAO) and acetyl cholinesterase (AChE) activities, and decrease the levels of malondialdehyde (MDA) in the brain tissue of the aged mice. The results showed that EPT improved memory functions of the aged mice probably via its antioxidant properties and via decreasing the activities of MAO and AChE

    The Prostate Basal Cell (BC) Heterogeneity and the p63-Positive BC Differentiation Spectrum in Mice

    The prostate epithelium is composed of basal (BC), luminal (LEC), and neuroendocrine (NEC) cells. It is unclear how many subtypes of BCs in the prostate and which subtype of BCs contains the main stem cell niche in the adult prostate. Here we report seven BC subpopulations according to their p63, cytokeratin 14 (K14) and K5 expression patterns, including p63-positive/K14-negative/K5-negative (p63+/K14-/K5-), p63-/K14+/K5-, p63-/K14-/K5+, p63+/K14+/K5-, p63+/K14-/K5+, p63-/K14+/K5+, and p63+/K14+/K5+ BCs. We generated a p63-CreERT2 knock-in mouse line that expresses tamoxifen-inducible Cre activity in the p63-expressing cells, including the prostate BCs. We then crossbred this line with ROSA26R mice, and generated p63-CreERT2×ROSA26R bi-genic mice harboring the Cre-activated β-galactosidase reporter gene. We treated these bi-genic mice with tamoxifen to mark the p63+ BCs at different ages or under different hormonal conditions, and then traced the lineage differentiation of these genetically labeled BCs. We discovered that these p63+ BCs contain self-renewable stem cells in culture and efficiently differentiated into LECs, NECs and BCs in the postnatal, adult and re-generating mouse prostates. Therefore, BC population contains heterogeneous BCs that express different combinations of the p63, K14 and K5 differentiation markers. Because K14+ and K5+ BCs were previously shown to be extremely inefficient to produce LECs in adulthood, we propose that the p63+/K5-/K14- subpopulation of BCs contains most stem-like cells, especially in adult animals

    Quantifying the impact of synoptic circulation patterns on ozone variability in northern China from April to October 2013-2017

    The characteristics of ozone variations and the impacts of synoptic and local meteorological factors in northern China were quantitatively analyzed during the warm season from 2013 to 2017 based on multi-city in situ ozone and meteorological data as well as meteorological reanalysis. The domain-averaged maximum daily 8 h running average O-3 (MDA8 O-3) concentration was 122 +/- 11 mu g m(-3), with an increase rate of 7.88 lug mu g m(-3) yr(-1), and the three most polluted months were closely related to the variations in the synoptic circulation patterns, which occurred in June (149 mu m(-3)), May (138 mu m(-3)) and July (132 mu g m(-3)). A total of 26 weather types (merged into five weather categories) were objectively identified using the Lamb-Jenkinson method. The highly polluted weather categories included the S-W-N directions (geostrophic wind direction diverts from south to north), low-pressure-related weather types (LP) and cyclone type, which the study area controlled by a low-pressure center (C), and the corresponding domain-averaged MDA8 03 concentrations were 122, 126 and 128 mu g m(-3), respectively. Based on the frequency and intensity changes of the synoptic circulation patterns, 39.2 % of the interannual increase in the domain-averaged O-3 from 2013 to 2017 was attributed to synoptic changes, and the intensity of the synoptic circulation patterns was the dominant factor. Using synoptic classification and local meteorological factors, the segmented synoptic-regression approach was established to evaluate and forecast daily ozone variability on an urban scale. The results showed that this method is practical in most cities, and the dominant factors are the maximum temperature, southerly winds, relative humidity on the previous day and on the same day, and total cloud cover. Overall, 41 %-63 % of the day-today variability in the MDA8 O-3 concentrations was due to local meteorological variations in most cities over northern China, except for two cities: QHD (Qinhuangdao) at 34 % and ZZ (Zhengzhou) at 20 %. Our quantitative exploration of the influence of both synoptic and local meteorological factors on interannual and day-to-day ozone variability will provide a scientific basis for evaluating emission reduction measures that have been implemented by the national and local governments to mitigate air pollution in northern China.Peer reviewe

    The Steroid Receptor Coactivator-3 Is Required for Developing Neuroendocrine Tumor in the Mouse Prostate

    Neuroendocrine tumor cells (NETCs) are commonly observed in prostate cancer. Their presence is associated with castration resistance, metastasis and poor prognosis. Cellular and molecular mechanisms for NETC initiation and growth are unknown. TRAMP mice develop heterogeneous adenocarcinomas induced by expression of the SV40-T/t oncogene in prostate epithelial cells. Here, we demonstrate prostate tumors in TRAMP mice with a mixed genetic background are characterized mostly by atypical hyperplasia (AH) containing steroid receptor coactiator-3-positive, androgen receptor-positive and synaptophysin-negative (SRC-3+/AR+/Syp-) cells. Few SRC-3+/AR-/Syp+ NETCs are present in their prostates. We generated TRAMP mice in which SRC-3 was specifically ablated in AR+/Syp- prostatic epithelial cells (termed PE3KOT mice). In these animals, we observed a substantial reduction in SRC-3-/AR+/Syp- AH tumor growth. There was a corresponding increase in SRC-3-/AR+/Syp- phyllodes lesions, suggesting SRC-3 knockout can convert aggressive AH tumors with mostly epithelial tumor cells into less aggressive phyllodes lesions with mostly stromal tissue. Surprisingly, PE3KOT mice developed many more SRC-3+/AR-/Syp+ NETCs versus control TRAMP mice, indicating SRC-3 expression was retained in NETCs. In contrast, TRAMP mice with global SRC-3 knockout did not develop any NETC, indicating SRC-3 is required for developing NETC. Analysis of cell-differentiating markers revealed that these NETCs might not be derived from the mature AR-/Syp+ neuroendocrine cells or the AR+/Syp- luminal epithelial tumor cells. Instead, these NETCs might originate from the SV40-T/t-transformed intermediate/progenitor epithelial cells. In summary, SRC-3 is required for both AR+/Syp- AH tumor growth and AR-/Syp+ NETC development, suggesting SRC-3 is a target for inhibiting aggressive prostate cancer containing NETCs

    Real-time rapid prediction of variations of Earth's rotational rate

    Real-time rapid prediction of variations of the Earth's rotational rate is of great scientific and practical importance. However, due to the complicated time-variable characteristics of variations of the Earth's rotational rate (i.e., length of day, LOD), it is usually difficult to obtain satisfactory predictions by conventional linear time series analysis methods. This study employs the nonlinear artificial neural networks (ANN) to predict the LOD variations. The topology of the ANN model is determined by minimizing the root mean square errors (RMSE) of the predictions. Considering the close relationships between the LOD variations and the atmospheric circulation movement, the operational prediction series of axial atmospheric angular momentum (AAM) is incorporated into the ANN model as an additional input in the real-time rapid prediction of LOD variations with 1-5 days ahead. The results show that the LOD prediction is significantly improved after introducing the operational prediction series of AAM into the ANN model
