    KAT5 Inhibitor NU9056 Suppresses Anaplastic Thyroid Carcinoma Progression through c-Myc/miR-202 Pathway

    Background. Anaplastic thyroid carcinoma (ATC) is considered to be one of the most aggressive cancers. Our previous study proved that highly expressed lysine acetyltransferase 5 (KAT5) in ATC is associated with a poorer prognosis. Here, this study examined the effects of a KAT5 inhibitor (NU9056) in human ATC cells. Methods. First, the Cancer Genome Atlas (TCGA) dataset was used to detect the relationship between KAT5 expression and outcomes of thyroid carcinoma patients. Then, both in vitro and in vivo experiments were conducted to investigate the effects of NU9056 on normal and ATC human thyroid cells. Finally, microRNA sequencing, qPCR, and dual-luciferase reporter assay were performed to explore potential mechanisms by identifying downstream microRNA related to NU9056. Results. KAT5 dysregulation correlated with more advanced-stage and poorer outcomes of thyroid carcinoma patients. Endogenous KAT5 protein and mRNA levels were much higher in ATC cells than in normal thyroid cells. Suppression of KAT5 by NU9056 inhibited survival, growth, migration, invasion, and tube formation, and increased radiosensitivity and chemosensitivity in ATC cells but showed no impact on normal thyroid cells. Mechanistically, microRNA-202-5p (miR-202) was identified as the most significantly decreased miRNA after NU9056 treatment. Knockdown of miR-202 suppressed ATC cell progression, while forced expression of miR-202 partially blocked the inhibitory effect of NU9056 on ATC cells. Furthermore, c-Myc was validated as the transcription factor of miR-202, and NU9056 decreased the c-Myc protein level by shortening its half-life. Finally, we proved that NU9056 inhibited ATC proliferation in vivo. Conclusions. Our results indicated that NU9056 targets KAT5, shortens c-Myc half-life, subsequently downregulates miR-202 expression, and results in the suppression of ATC cells. Overall, KAT5 could be a potential target for clinical treatment for ATC

    Source Analysis and Contamination Assessment of Potentially Toxic Element in Soil of Small Watershed in Mountainous Area of Southern Henan, China

    In this study, the concentrations of potentially toxic elements in 283 topsoil samples were determined. Håkanson toxicity response coefficient modified matter element extension model was introduced to evaluate the soil elements contamination, and the results were compared with the pollution index method. The sources and spatial distribution of soil elements were analyzed by the combination of the PMF model and IDW interpolation. The results are as follows, 1: The concentration distribution of potentially toxic elements is different in space. Higher concentrations were found in the vicinity of the mining area and farmland. 2: The weight of all elements has changed significantly. The evaluation result of the matter-element extension model shows that 68.55% of the topsoil in the study area is clean soil, and Hg is the main contamination element. The evaluation result is roughly the same as that of the pollution index method, indicating that the evaluation result of the matter-element extension model with modified is accurate and reasonable. 3: Potentially toxic elements mainly come from the mixed sources of atmospheric sedimentation and agricultural activities (22.59%), the mixed sources of agricultural activities and mining (20.26%), the mixed sources of traffic activities, nature and mining (36.30%), the mixed sources of pesticide use and soil parent material (20.85%)

    Investigation of Dynamic Mechanical Behavior and Damage Characteristics in TC32 Alloy

    The dynamic shearing experiment was been done by using split Hopkinson pressure bar(SHPB) technique in TC32 alloy with lamellar, bimodal and basket microstructures. The damage Characteristics of TC32 alloy was investigated by using optical microscope(OP) and scanning electron microscopy(SEM). The results show that the critical fracture velocity is 2400 s-1, 2700 s-1, and 2600 s-1 for lamellar, bimodal, and basket microstructures respectively. The bimodal microstructure exhibit the best Dynamic mechanical behavior compared with the other two microstructures. Adiabatic shear bands(ASBs) and microvoids initiation, growth, and coalescence to damage in adiabatic shear bands(ASBs) were observed in all of three microstructures. Also, microvoids initiation and growth are prior to the interface between ASBs and matrix. Investigation indicated that plastic flow characteristic is not obvious at the interface between ASBs and matrix, which observed long crack in lamellar microstructure. In bimodal microstructure, fibrous a adiabatic shear bands(ASBs) and surrounding region are shown. Because of strong shear deformation, the plastic flow characteristic appears clearly, and primary α phase was elongated. Microvoids initiation is also prior to the α/β phase boundaries. The damage characteristics of basket microstructure are similar to bimodal microstructure. But unlike lamellar and basket microstructures, the microvoids are initiatied when the acicular primary α phase arranged in order is perpendicular to the adiabatic shear bands(ASBs) in lamellar microstructure. ASBs is mostly consisted of equiaxed grains, and the deformation mechanism still wasn't defined

    Information disclosure is an important way for investors to obtain information, the annual report text carries a lot of information, lazy information disclosure is an important form of information disclosure of the annual report text. This paper takes China’s A-share listed companies from 2011 to 2022 as the research sample, takes the annual report text information disclosure form as the entry point, uses the computer text analysis technology to measure the text similarity of the annual report to measure the lazy information disclosure, and explores its impact on stock price crash risk. The results show that there is a positive correlation between the similarity of annual report text and the risk of stock price crash, that is, when the information of annual report text is presented in the form of lazy information disclosure, the risk of stock price crash increases. For companies audited by key auditing institutions, the positive correlation between the similarity of their annual reports and the risk of stock price crash is not significant, indicating that key auditing institutions will weaken the positive correlation between lazy information disclosure and the risk of stock price crash. Further, through external attention and analysis of the time delay of annual report disclosure, it is concluded that the management lacks the opportunity and time to hide the bad news, so it is clear that the lazy information disclosure comes from the business situation "the fact is so". The research conclusion of this paper provides evidence support for the influence of lazy information disclosure on stock price crash risk, and also provides useful reference for regulators to improve information disclosure policies and effectively prevents and resolves stock price crash risk.</div
