    AbstractEffect of nitric oxide (NO) treatment on storage quality and disease resistance of Glorious oranges was investigated in the experiment. The results showed that NO treatment could effectively reduce disease incidence inoculated with Collletotichum goeosporioides Penz and inhibit the increase of lesion diameter of Glorious oranges during storage. Compared with the control, NO treatment kept higher level of titritable acidity (TA), soluble protein, ascorbic acid (ASA) and reducing sugar, and lower level of weight lose rate and soluble solid concentration (SSC), retarding ripening of fruits

    Prospective assessment of breast lesions AI classification model based on ultrasound dynamic videos and ACR BI-RADS characteristics

    IntroductionAI-assisted ultrasound diagnosis is considered a fast and accurate new method that can reduce the subjective and experience-dependent nature of handheld ultrasound. In order to meet clinical diagnostic needs better, we first proposed a breast lesions AI classification model based on ultrasound dynamic videos and ACR BI-RADS characteristics (hereafter, Auto BI-RADS). In this study, we prospectively verify its performance.MethodsIn this study, the model development was based on retrospective data including 480 ultrasound dynamic videos equivalent to 18122 static images of pathologically proven breast lesions from 420 patients. A total of 292 breast lesions ultrasound dynamic videos from the internal and external hospital were prospectively tested by Auto BI-RADS. The performance of Auto BI-RADS was compared with both experienced and junior radiologists using the DeLong method, Kappa test, and McNemar test.ResultsThe Auto BI-RADS achieved an accuracy, sensitivity, and specificity of 0.87, 0.93, and 0.81, respectively. The consistency of the BI-RADS category between Auto BI-RADS and the experienced group (Kappa:0.82) was higher than that of the juniors (Kappa:0.60). The consistency rates between Auto BI-RADS and the experienced group were higher than those between Auto BI-RADS and the junior group for shape (93% vs. 80%; P = .01), orientation (90% vs. 84%; P = .02), margin (84% vs. 71%; P = .01), echo pattern (69% vs. 56%; P = .001) and posterior features (76% vs. 71%; P = .0046), While the difference of calcification was not significantly different.DiscussionIn this study, we aimed to prospectively verify a novel AI tool based on ultrasound dynamic videos and ACR BI-RADS characteristics. The prospective assessment suggested that the AI tool not only meets the clinical needs better but also reaches the diagnostic efficiency of experienced radiologists

    Predicting Housekeeping Genes Based on Fourier Analysis

    Housekeeping genes (HKGs) generally have fundamental functions in basic biochemical processes in organisms, and usually have relatively steady expression levels across various tissues. They play an important role in the normalization of microarray technology. Using Fourier analysis we transformed gene expression time-series from a Hela cell cycle gene expression dataset into Fourier spectra, and designed an effective computational method for discriminating between HKGs and non-HKGs using the support vector machine (SVM) supervised learning algorithm which can extract significant features of the spectra, providing a basis for identifying specific gene expression patterns. Using our method we identified 510 human HKGs, and then validated them by comparison with two independent sets of tissue expression profiles. Results showed that our predicted HKG set is more reliable than three previously identified sets of HKGs

    Mutations in the C-terminus of the X protein of hepatitis B virus regulate Wnt-5a expression in hepatoma Huh7 cells: cDNA microarray and proteomic analyses

    Background: The hepatitis B virus x gene (HBx) is a promiscuous transactivator implicated in the development of hepatocellular carcinoma (HCC). The present study was designed to investigate the molecular events regulated by HBx. Methods: Genomic and proteomic expression profiling was performed in Huh7 HCC cells transfected with HBx mutants with a C-terminal deletion. The gene and protein expression of wingless-type murine-mammary-tumour virus (MMTV) integration site family, member 5A (Wnt-5a) was validated by analyses of reverse transcription–polymerase chain reaction (RT–PCR), real-time RT–PCR, western blot and immunohistochemistry. Results: Differentially expressed genes and proteins were found in the transfected Huh7 HCC cells; most of them were involved in transcriptional regulation, although others including oncogenes or tumor suppressor genes, and molecules involved in cell junctions, signal transduction pathways, metabolism or the immune response were also observed. The expression of the Wnt-5a gene was elevated >10-fold in Huh7 cells transfected with the HBx3′-30 amino acid deletion mutant. However, the expression was downregulated by the transfection with the HBx3′-40 amino acid deletion mutant. The changes in Wnt-5a expression were also observed in human HCC tissues, compared with corresponding non-cancerous liver tissues. A negative correlation was found between the expression of Wnt-5a and HBx COOH mutations in HCC tissues. Conclusions: HBx mutants may participate in the development and progression of HCC, at least in part through the Wnt-5a pathway

    Primary mesenchymal chondrosarcoma of the kidney with synchronous implant and infiltrating urothelial carcinoma of the ureter

    <p>Abstract</p> <p>Primary mesenchymal chondrosarcoma of the kidney is rare, and it shows distinct undifferentiated tumor cells and well differentiated cartilagenous components. Also assident infiltrating urothelial carcinoma of the ureter is an extremely rare cancer. We report a case of primary mesenchymal chondrosarcoma occurring in the left kidney with an ipsilateral and distinct distal ureteric implant, and a coexisting infiltrating urothelial carcinoma of the ureter in a 64-year-old man. Histopathological examination and immunohistochemical studuies showed the classic features of mesenchymal chondrosarcoma in kidney, as well as a few infiltrating urothelial in ureter. Multitarget fluorescence in situ hybridization (FISH) suggested that the development of the urothelial carcinoma in the ureter may be triggered or induced by the chondrosarcoma component. The patient died 2 month after left nephro-ureterectomy.     Porous highly fluorinated polyimide/polydopamine nanocomposite films with simultaneously enhanced toughness, UV-shielding and photostability for aerospace applications

    Polydopamine (PDA), also known as artificial melanin, was adopted as ultraviolet absorbent to prepare a series of porous highly fluorinated polyimide/polydopamine (FPI/PDA) films by cyclodehydration via chemical and thermal imidization. The hydrogen bond and porous structure endowed the reinforced thermal, mechanical, UV-shielding and photostability. The initial decomposition and glass transition temperatures of FPI/PDA (1 wt %) film were as high as 461.5 °C and 341.5 °C, respectively beyond pure FPI by 54.1 °C and 26.0 °C. The stress-strain curves indicated that tensile strength and fracture elongation first increased and then decreased with the increase of PDA content, and reached the maximum at 0.5 wt % PDA content, which were 1.41 and 29.47 times of FPI, respectively. UV–vis transmittance spectra, curcumin photolysis and recyclability measurement confirmed the splendid UV-shielding properties with simultaneous excellent visible light transmittance and durability of porous FPI/PDA film. FTIR and AFM manifested its superior photostability with mild variation in carbonyl absorption peaks and surface roughness after 720 h UV irradiation. The above enhancement mechanisms included synergistic UV absorption of porous FPI and PDA, photothermal conversion in a stable FPI, and PDA's free radical trapping, promoting porous FPI/PDA film as an aerospace UV shielding material

    Triple critical feature capture network: A triple critical feature capture network for weakly supervised object detection

    Weakly supervised object detection (WSOD) is becoming increasingly important for computer vision tasks, as it alleviates the burden of manual annotation. Most WSOD techniques rely on multiple instance learning (MIL), which tends to localise the discriminative parts of salient objects instead of the whole object. In addition, network training is often supervised using simple image-level annotations, without including object quantities or location information. However, this can lead to ambiguous differentiation of object instances, both in terms of location and semantics. To address these issues, propose an end-to-end triple critical feature capture network (TCFCNet) for WSOD is proposed. Specifically, a multi-task branch, which can perform fully supervised classification and regression task, was integrated with a PCL in an end-to-end network for refining object locations in an online method. A cyclic parametric dropblock module (CPDM) was then designed to help the detector focus on the contextual information by using cyclic masking techniques to maximise the removal of the discriminative components of an object instance to alleviate the part domination problem. Finally, a feature decoupling module (FDM) is proposed to further reduce the ambiguous distinction of object instances by adaptively constructing robust critical features that adapt to multi-task branch for classification and regression tasks, which contains a feature enhancement module and task-specific polarisation functions. Comprehensive experiments are carried out on the challenging Pascal VOC 2007 and VOC 2012 datasets. The proposed method achieves a 54.6% mAP and a 44.3% mAP on the Pascal VOC 2007 and VOC 2012 datasets respectively, showed that our method outperformed existing mainstream techniques by a considerable margin

    Untargeted Metabolomics Reveals the Protective Effect of a Traditional Chinese Herbal Decoction on Cisplatin-Induced Acute Kidney Injury

    Our previous studies have demonstrated that Jian-Pi-Yi-Shen formula (JPYSF), a traditional Chinese herbal decoction, has a renoprotective effect in 5/6 nephrectomy-induced chronic kidney injury. However, the role and potential mechanisms of JPYSF in the treatment of acute kidney injury (AKI) remain unknown. This study was designed to test the beneficial effect of JPYSF in an AKI mouse model and to investigate the underlying mechanism by using metabolomics analysis. The AKI mouse model was induced by a single intraperitoneal injection of cisplatin at a dose of 20 mg/kg. The mice in the treatment group were pretreated orally with JPYSF (18.35 g/kg/d) for 5 days before cisplatin injection. Seventy-two hours after cisplatin injection, serum and kidney samples were collected for biochemical and histological examination. Ultra-high-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UHPLC-QTOF/MS) was applied to analyze metabolic profiling variations in the kidney. The results showed that pretreatment with JPYSF obviously reduced the levels of serum creatinine and blood urea nitrogen and alleviated renal pathological injury in AKI mice. Orthogonal partial least-squares discriminant analysis (OPLS-DA) score plot revealed a clear separation between the AKI and AKI + JPYSF group. A total of 68 and 87 significantly differentially expressed metabolites were identified in the kidney of AKI mice responding to JPYSF treatment in negative and positive ion mode, respectively. The pivotal pathways affected by JPYSF included vitamin B6 metabolism, alanine, aspartate and glutamate metabolism, lysine biosynthesis, and butanoate metabolism. In conclusion, JPYSF can protect the kidney from cisplatin-induced AKI, which may be associated with regulating renal metabolic disorders
