    How Generalizable are Deepfake Detectors? An Empirical Study

    Deepfake videos and images are becoming increasingly credible, posing a significant threat given their potential to facilitate fraud or bypass access control systems. This has motivated the development of deepfake detection methods, in which deep learning models are trained to distinguish between real and synthesized footage. Unfortunately, existing detection models struggle to generalize to deepfakes from datasets they were not trained on, but little work has been done to examine why or how this limitation can be addressed. In this paper, we present the first empirical study on the generalizability of deepfake detectors, an essential goal for detectors to stay one step ahead of attackers. Our study utilizes six deepfake datasets, five deepfake detection methods, and two model augmentation approaches, confirming that detectors do not generalize in zero-shot settings. Additionally, we find that detectors are learning unwanted properties specific to synthesis methods and struggling to extract discriminative features, limiting their ability to generalize. Finally, we find that there are neurons universally contributing to detection across seen and unseen datasets, illuminating a possible path forward to zero-shot generalizability.Comment: This work has been submitted to the IEEE for possible publication.     Enzyme-linked immunosorbent assay of changes in serum levels of growth hormone (cGH) in common carps (Cyprinus carpio)

    The aim of the present study was to purify the common native carp growth hormone (ncGH), produce monoclonal antibodies (mAbs) to common native carp growth hormone (ncGH), and further enhance the sensitivity of enzyme-linked immunosorbent assays (ELISA) for ncGH. Additionally, we investigated changes in serum ncGH levels in carps raised in different environmental conditions. The recombinant grass carp (Ctenopharyngodon idella) growth hormone was purified and used as antigen to immunize the rabbit. The natural ncGH was isolated from the pituitaries of common carp. SDS-PAGE and Western blot utilizing the polyclonal anti-rgcGH antibody confirmed the purification of ncGH from pituitaries. Purified ncGH was then used as an immunogen in the B lymphocyte hybridoma technique. A total of 14 hybridoma cell lines (FMU-cGH 1-14) were established that were able to stably secrete mAbs against ncGH. Among them, eight clones (FMU-cGH1-6, 12 and 13) were successfully used for Western blot while nine clones (FMU-cGH 1-7, 9 and 10) were used in fluorescent staining and immunohistochemistry. Epitope mapping by competitive ELISA demonstrated that these mAbs recognized five different epitopes. A sensitive sandwich ELISA for detection of ncGH was developed using FMU-cGH12 as the coating mAb and FMU-cGH6 as the enzyme labeled mAb. This detection system was found to be highly stable and sensitive, with detection levels of 70 pg/mL.     The K2-HERMES Survey: Age and Metallicity of the Thick Disc

    Asteroseismology is a promising tool to study Galactic structure and evolution because it can probe the ages of stars. Earlier attempts comparing seismic data from the {\it Kepler} satellite with predictions from Galaxy models found that the models predicted more low-mass stars compared to the observed distribution of masses. It was unclear if the mismatch was due to inaccuracies in the Galactic models, or the unknown aspects of the selection function of the stars. Using new data from the K2 mission, which has a well-defined selection function, we find that an old metal-poor thick disc, as used in previous Galactic models, is incompatible with the asteroseismic information. We show that spectroscopic measurements of [Fe/H] and [α\alpha/Fe] elemental abundances from the GALAH survey indicate a mean metallicity of log(Z/Z)=0.16\log (Z/Z_{\odot})=-0.16 for the thick disc. Here ZZ is the effective solar-scaled metallicity, which is a function of [Fe/H] and [α\alpha/Fe]. With the revised disc metallicities, for the first time, the theoretically predicted distribution of seismic masses show excellent agreement with the observed distribution of masses. This provides an indirect verification of the asteroseismic mass scaling relation is good to within five percent. Using an importance-sampling framework that takes the selection function into account, we fit a population synthesis model of the Galaxy to the observed seismic and spectroscopic data. Assuming the asteroseismic scaling relations are correct, we estimate the mean age of the thick disc to be about 10 Gyr, in agreement with the traditional idea of an old α\alpha-enhanced thick disc.Comment: 21 pages, submitted to MNRA

    Distribution of HLA-A, -B and -DRB1 Genes and Haplotypes in the Tujia Population Living in the Wufeng Region of Hubei Province, China

    BACKGROUND: The distribution of HLA alleles and haplotypes varies widely between different ethnic populations and geographic areas. Before any genetic marker can be used in a disease-associated study it is therefore essential to investigate allelic frequencies and establish a genetic database. METHODOLOGY/PRINCIPAL FINDINGS: This is the first report of HLA typing in the Tujia group using the Luminex HLA-SSO method HLA-A, -B and -DRB1 allelic distributions were determined in 124 unrelated healthy Tujia individuals, and haplotypic frequencies and linkage disequilibrium parameters were estimated using the maximum-likelihood method. In total 10 alleles were detected at the HLA-A locus, 21 alleles at the HLA-B locus and 14 alleles at the HLA-DRB1 locus. The most frequently observed alleles in the HLA-I group were HLA-A*02 (35.48%), A*11 (28.23%), A*24 (15.73%); HLA-B*40 (25.00%), B*46 (16.13%), and B*15 (15.73%). Among HLA-DRB1 alleles, high frequencies of HLA-DRB1*09 (25.81%) were observed, followed by HLA-DRB1*15 (12.9%), and DRB1*12 (10.89%). The two-locus haplotypes at the highest frequency were A*02-B*46A (8.47%), followed by A*11-B*40 (7.66%), A*02-B*40 (8.87%), A*11-B*15 (6.45%), A*02-B*15 (6.05%), B*40-DRB1*09 (9.27%) and B*46-DRB1*09 (6.45%). The most common three-locus haplotypes found in the Tujia population were A*02-B*46-DRB1*09 (4.84%) and A*02-B*40-DRB1*09 (4.03%). Fourteen two-loci haplotypes had significant linkage disequilibrium. Construction of a neighbor-joining phylogenetic tree and principal component analysis using the allelic frequencies at HLA-A was performed to compare the Tujia group and twelve other previously reported populations. The Tujia population in the Wufeng of Hubei Province had the closest genetic relationship with the central Han population, and then to the Shui, the Miao, the southern Han and the northern Han ethnic groups. CONCLUSIONS/SIGNIFICANCE: These results will become a valuable source of data for tracing population migration, planning clinical organ transplantation, carrying out HLA-linked disease-associated studies and forensic identification

    MICA/B expression is inhibited by unfolded protein response and associated with poor prognosis in human hepatocellular carcinoma

    BackgroundMICA/B are major ligands for NK cell activating receptor NKG2D and previous studies showed that the serum level of soluble MICA (sMICA) is an independent prognostic factor for advanced human hepatocellular carcinoma. However, the correlation between cellular MICA/B expression pattern and human hepatocellular carcinoma progression has not been well explored. The unfolded protein response is one of the main causes of resistance to chemotherapy and radiotherapy in tumor cells. However, whether the UPR in HCC could regulate the expression levels of MICA/B and affect the sensitivity of HCC cells to NK cell cytolysis has not been established yet.MethodsMICA/B expression pattern was evaluated by immunohistochemistry and Kaplan-Meier survival analysis was done to explore the relationship between MICA/B expression level and patient survival. The protein and mRNA expression levels of MICA/B in SMMC7721 and HepG2 cells treated by tunicamycin were evaluated by flow cytometry, Western Blot and RT-PCR. The cytotoxicity analysis was performed with the CytoTox 96 Non-Radioactive LDH Cytotoxicity Assay.ResultsMICA/B was highly expressed in human hepatocellular carcinoma and the expression level was significantly and negatively associated with tumor-node metastasis (TNM) stages. Patients with low level of MICA/B expression showed a trend of shorter survival time. The unfolded protein response (UPR) downregulated the expression of MICA/B. This decreased protein expression occurred via post-transcriptional regulation and was associated with proteasomal degradation. Moreover, decreased expression level of MICA/B led to the attenuated sensitivity of human HCC to NK cell cytotoxicity.ConclusionThese new findings of the connection of MICA/B, UPR and NK cells may represent a new concrete theory of NK cell regulation in HCC, and suggest that targeting this novel NK cell-associated immune evasion pathway may be meaningful in treating patients with HCC.Electronic supplementary materialThe online version of this article (doi:10.1186/s13046-014-0076-7) contains supplementary material, which is available to authorized users

    Effect of Colloidal Silica on the Hydration Behavior of Calcium Aluminate Cement

    The effect of colloidal silica (CS) on the hydrate phases and microstructure evolution of calcium aluminate cement (CAC) was investigated. Samples hydrated with CS were obtained and characterized by X-ray diffraction (XRD), Field Emission Scanning Electron Microscopy (FESEM), Fourier Transform Infrared spectroscopy (FT-IR), hydration heat measurement and Nuclear Magnetic Resonance (NMR). The results revealed that SiO2 nanoparticles may affect the hydrates crystallization process. There was a compact structure in the CAC paste with CS, while petal-shaped hydrates with a porous structure were formed in the pure CAC paste. The maximum value of electrical conductivity for CAC paste with CS suggested that the early stage of hydration for CAC was accelerated. However, the hydration heat curves revealed that the late stage of the CAC hydration process was inhibited, and the hydration degree was reduced, this result was in accordance with Thermogravimetry-Differential scanning calorimetry(TG-DSC) curves. The fitting results of hydration heat curves further showed that the hydration degree at NG (nucleation and crystal growth) process stage was promoted, while it was limited at the phase boundaries stage, and the diffusion stage in the hydration reaction was brought forward due to the addition of CS. According to these results and analyses, the differences in the hydration process for CAC with and without CS can be attributed to the distribution and nucleation effect of SiO2 nanoparticles

    Research Status and Prospect on Vanadium-Based Catalysts for NH3-SCR Denitration

    Selective catalytic reduction of NOx with NH3 is one of the most widely used technologies in denitration. Vanadium-based catalysts have been extensively studied for the deNOx process. V2O5/WO3(MoO3)TiO2 as a commercial catalyst has excellent catalytic activity in the medium temperature range. However, it has usually faced several problems in practical industrial applications, including narrow windows of operation temperatures, and the deactivation of catalysts. The modification of vanadium-based catalysts will be the focus in future research. In this paper, the chemical composition of vanadium-based catalysts, catalytic mechanism, the broadening of the temperature range, and the improvement of erosion resistance are reviewed. Furthermore, the effects of four major systems of copper, iron, cerium and manganese on the modification of vanadium-based catalysts are introduced and analyzed. It is worth noting that the addition of modified elements as promoters has greatly improved the catalytic performance. They can enhance the surface acidity, which leads to the increasing adsorption capacity of NH3. Surface defects and oxygen vacancies have also been increased, resulting in more active sites. Finally, the future development of vanadium-based catalysts for denitration is prospected. It is indicated that the main purpose for the research of vanadium-based modification will help to obtain safe, environmentally friendly, efficient, and economical catalysts
