    Querying Schemas With Access Restrictions

We study verification of systems whose transitions consist of accesses to a Web-based data-source. An access is a lookup on a relation within a relational database, fixing values for a set of positions in the relation. For example, a transition can represent access to a Web form, where the user is restricted to filling in values for a particular set of fields. We look at verifying properties of a schema describing the possible accesses of such a system. We present a language where one can describe the properties of an access path, and also specify additional restrictions on accesses that are enforced by the schema. Our main property language, AccLTL, is based on a first-order extension of linear-time temporal logic, interpreting access paths as sequences of relational structures. We also present a lower-level automaton model, Aautomata, which AccLTL specifications can compile into. We show that AccLTL and A-automata can express static analysis problems related to "querying with limited access patterns" that have been studied in the database literature in the past, such as whether an access is relevant to answering a query, and whether two queries are equivalent in the accessible data they can return. We prove decidability and complexity results for several restrictions and variants of AccLTL, and explain which properties of paths can be expressed in each restriction.Comment: VLDB201

Leucine Supplementation in Cuprizone-Induced Oligodendrocyte Toxicity

Cuprizone is a copper chelator that induces demyelination in the central nervous system when fed to mice. This compound is thought to target Complex IV and disrupt mitochondrial metabolism leading to loss of myelin-producing oligodendrocytes. In this proposal, we further examine the chemical and toxicological properties of cuprizone. Immunofluorescence imaging and microarray data on MO3.13 cells treated with cuprizone revealed the possibility of alterations in lysosomal function, as well as mitochondrial disruption via mTOR and Electron Transport Chain (ETC) related pathways. Cell viability assays conducted suggest that addition of Branched-Chain Amino Acids, most commonly used from literature, leucine, can increase cell vitality after being treated with cuprizone. Additional studies examined the solubility of cuprizone and interaction with zinc. Taken altogether, this experiment reveals that cuprizone affects lysosomal and mitochondrial function, and provides insight into leucine supplementation at an attempt to rescue cells from neurotoxicity

Uniqueness Results for Second Order Bellman-Isaacs Equations under Quadratic Growth Assumptions and Applications

In this paper, we prove a comparison result between semicontinuous viscosity sub and supersolutions growing at most quadratically of second-order degenerate parabolic Hamilton-Jacobi-Bellman and Isaacs equations. As an application, we characterize the value function of a finite horizon stochastic control problem with unbounded controls as the unique viscosity solution of the corresponding dynamic programming equation

CXCR2- and E-Selectin–induced Neutrophil Arrest during Inflammation In Vivo

The signaling events leading to the activation of integrins and firm arrest of rolling neutrophils in inflamed venules have yet to be elucidated. In vitro assays suggest that both E-selectin and chemokines can trigger arrest of rolling neutrophils, but E-selectin−/− mice have normal levels of adherent neutrophils in inflamed venules. To test whether chemokine-induced neutrophil arrest in vivo can be unmasked by blocking E-selectin, we investigated neutrophil adhesion in inflamed cremaster muscle venules in tumor necrosis factor (TNF)-α–treated CXCR2−/− or wild-type (WT) mice injected with E-selectin blocking monoclonal antibody (mAb) 9A9. To block chemokine receptor signaling, we investigated E-selectin−/− or WT mice treated with pertussis toxin (PTx) intravenously. Neutrophil adhesion was unchanged in CXCR2−/−, E-selectin−/−, PTx-treated WT, or mAb 9A9–treated WT mice. However, TNF-α–induced neutrophil adhesion was almost completely abrogated in E-selectin−/− mice treated with PTx and significantly reduced in CXCR2−/− mice treated with the E-selectin blocking mAb. In thioglycollate-induced peritonitis, PTx treatment blocked neutrophil recruitment into the peritoneum of E-selectin−/− mice, but had only a partial effect in WT animals. These data show that E-selectin– and chemokine-mediated arrest mechanisms are overlapping in this model and identify CXCR2 as an important neutrophil arrest chemokine in vivo

How Mouse Macrophages Sense What Is Going On

    Get PDF
Macrophages are central to both innate and adaptive immunity. With few exceptions, macrophages are the first cells that sense trouble and respond to disturbances in almost all tissues and organs. They sense their environment, inhibit or kill pathogens, take up apoptotic and necrotic cells, heal tissue damage, and present antigens to T cells. Although the origins (yolk sac versus monocyte-derived) and phenotypes (functions, gene expression profiles, surface markers) of macrophages vary between tissues, they have many receptors in common that are specific to one or a few molecular species. Here, we review the expression and function of almost 200 key macrophage receptors that help the macrophages sense what is going on, including pathogen-derived molecules, the state of the surrounding tissue cells, apoptotic and necrotic cell death, antibodies and immune complexes, altered self molecules, extracellular matrix components, and cytokines, including chemokines
