    Membrane glucocorticoid receptors are localised in the extracellular matrix and signal through the MAPK pathway in mammalian skeletal muscle fibres

    A number of studies have previously proposed the existence of glucocorticoid receptors on the plasma membrane of many cell types including skeletal muscle fibres. However, their exact localisation and the cellular signalling pathway(s) they utilise to communicate with the rest of the cell are still poorly understood. In this study, we investigated the localisation and the mechanism(s) underlying the non-genomic physiological functions of these receptors in mouse skeletal muscle cells. The results show that the receptors were localised in the cytoplasm in myoblasts, in the nucleus in myotubes and in the extracellular matrix, in satellite cells and in the proximity of mitochondria in adult muscle fibres. Also, they bound laminin in a glucocorticoid-dependent manner. Treating small skeletal muscle fibre bundles with the synthetic glucocorticoid, beclomethasone dipropionate, increased the phosphorylation (=activation) of extracellular-signal regulated kinase 1&2, c-Jun N-terminal kinase and p38 mitogen-activated protein kinase. This occurred within 5min and depended on the fibre-type and the duration of the treatment. It was also abolished by the glucocorticoid receptor inhibitor, mifepristone, and a monoclonal antibody against the receptor. From these results we conclude that the non-genomic/non-canonical physiological functions of glucocorticoids, in adult skeletal muscle fibres are mediated by a glucocorticoid receptor localised in the extracellular matrix, in satellite cells and close to mitochondria and involve activation of the MAPK pathway

Our gains have not been made easily and the price has been paid in both men and equipment.     Road Dust Correlated with Decreased Reproduction of the Endangered Utah Shrub Hesperidanthus suffrutescens

    Roads associated with energy development have fragmented much of the Uinta Basin, the Colorado Plateau in general, and other areas of western North America. Beyond reducing available habitat, spreading exotic species, and creating barriers to dispersal, unpaved roads also increase dust loads on plants and pollinators, which may reduce plant growth and reproduction. We studied the effects of an unpaved road on reproduction of an endangered Utah endemic shrub. We measured the size and reproductive output of 156 plants and the dust deposition in plots at increasing distances from the road. We also hand outcrossed 240 flowers from 80 plants to help determine if any reduced reproduction was due to pre- or postpollination mechanisms. Additionally, we experimentally dusted 3 leaves on 30 plants (n = 90) and measured stomatal conductance pre-dust and post-dust. We also dusted 3 flowers on 10 plants (n = 30) prior to hand pollination and measured fruit set. Generalized linear mixed models were used to examine the relationship between reproduction and dust deposition. When controlling for plant size and distance from the road, fruit set was negatively correlated with increasing levels of dust deposition (F1, 15 = 5.26, P = 0.036). The number of seeds per plant, mean plant seed weight, and the proportion of hand-pollinated flowers that set fruit were also negatively correlated with dust, although not significantly. Dusting significantly reduced stomatal conductance (F1, 58 = 87.56, P \u3c 0.001). Eighty percent of hand pollinated flowers (24 of 30) set fruit after dusting. These results demonstrate that road dust reduces H. suffrutescens reproduction, although the mechanisms are not clear. Although dust negatively affected physiological processes, hand-pollination results suggest that dust might be disrupting pollination. This study documents the effects of road dust on the reproduction of an endangered shrub in Utah\u27s Uinta Basin and highlights the need for further research into the effects of roads and dust on nearby plants

    A Photogrammetric Analysis of Cuneiform Tablets for the purpose of Digital Reconstruction

    Despite the advances made in the recording and cataloguing of cuneiform tablets, there is still much work to be done in the field of cuneiform reconstruction. The processes employed to rebuild cuneiform fragments still rely on glue and putty, with manual matching of fragments from catalogues or individual collections. The reconstruction process is hindered by inadequate information about the size and shape of fragments, and the inaccessibility of the original fragments makes finding information difficult in some collections. Most catalogue data associated with cuneiform tablets concerns the content of the text, and not the physical appearance of complete or fragmented tablets. This paper shows how photogrammetric analysis of cuneiform tablets can be used to retrieve physical information directly from source materials without the risk of human error. An initial scan of 8000 images from the CDLI database has already revealed interesting new information about the tablets held in cuneiform archives, and offered new avenues for research within the cuneiform reconstruction process.IBM Visual and Spatial Technology Centre, Institute of Archaeology and Antiquity, University of Birmingham, Edgbaston, Birmingham, B15 2TT
