Immunoaffinity isolation of Na,K-ATPase Ξ±3 isoform from pig kidney
AbstractThe Na,K-ATPase Ξ±3 isoform of the catalytic subunit has been isolated from pig kidney microsomes. The procedure employs immunoaffinity chromatography on Sepharose 4B covalently coupled with monospecific antibodies a-II against the synthetic peptide including the putative Ξ±3 N-terminus. The structural analysis provides unambiguous proof that the isolated protein corresponds to the third transcript for the Ξ±3 isoform. The N-terminal amino acid sequence determined, Met-Gly-Asp-Lys-Lys-Asp-Asp, shows that unlike the Ξ±1 and Ξ±2 proteins, the mature Na,K-ATPase Ξ±3 isoform lacks post-translational proteolytic processing.ATPase isozyme, (Na+ + K+)-; Immunoaffinity chromatography; N-terminal sequence analysi
The effect of intense external influences on the structure and properties of alloys
For the production of special high-wire thin and very thin sections austenitic steel has been widely adopted.Developed carbon-free highly alloyed Fe-Cr-Ni based steel in the quenched condition has a high plasticity,manufacturability and low strength. High adaptability of these steels allowed conducting extensive plasticdeformation as shear under pressure and drawing. Prerequisite for a high plasticity and adaptability of thedeveloped steels are: their doping with low carbon, 0.03% C, as well as cobalt and nickel content, whichincrease the plasticity of steels. Secondly, the presence of strain-metastable austenite, which during severeplastic deformation is almost completely transformed into deformation martensite and the related TRIP-effect.Especially important is the fact that thanks to high steels manufacturability, the effect of severe plasticdeformation leads to the formation of submicro- and nanocrystalline structure (mainly with high-anglemisorientations at grain boundaries with high strength) in long workpieces. The aging of the deformed steelscauses an additional increase of mechanical properties, which is associated with the occurrence of asupersaturated BCC solid solution (strain martensite). The resulting allocation of intermetallic phase NiAl isnanocrystalline, which is especially important in obtaining the finest wire diameters. The urgency of the paper is determined by the continuous information development of all spheres of education: integration of new knowledge, accessibility of information technologies and computer facility aids, professionalization and computerization of educational activities. The purpose of the research is to develop the structure and content of learning information environment in a higher education institute on the basis of geo-information technologies. The authors show the possibilities of using geo-information technologies in teaching outside the scope of their typical application (geographic, geodetic, geological education). The principles of designing the information environment for training on the basis of geo-information technologies are developed, which is built into the general information environment of higher education institute. The peculiarities of using geo-information technologies in non-core training are revealed, and the structure of learning environment modules based on geo-information technologies is developed and their content is described. The authors adapted the system of criteria evaluating the the effectiveness of training information environment, carried out an empirical study of the quality of education information environment in a higher education institute on the basis of geo-information technologies. Cognitive simulation as integrated innovative technology in teaching of social and humanitarian disciplines
Currently, teachers are searching for innovative educational technologies that enhance the effectiveness of educational activities. The purpose of the paper is the development and approbation of cognitive simulation technology in teaching of social and humanitarian disciplines. Based on the integration of sources of innovation, the authors developed the technology of cognitive simulation aimed at the formation of a structured system of knowledge for social and humanitarian disciplines' subject area. Various forms of cognitive map methods used in the learning process (analysis, synthesis, projection, and simulation) allow developing the analytical and predictive abilities of students, and strengthening their educational motivation. The technology use is proposed for the first time for all socio-humanitarian disciplines in the process of vocational training, which contributes to the formation of inter-subject communications, spatial understanding of events, and transformations of reality. The empirical study carried out by the authors on the implementation of this technology in the educational process (the students of 3 universities, totaling 315 people participated in the study) confirmed the productivity of the technology and the possibility of its adaptation to various disciplines of social and humanitarian sphere. Background: Endothelial dysfunction is recognized as one of the early markers of cardiovascular disorders. It is supposed to be a potential predictor of cardiovascular complications in patients receiving adjuvant and neoadjuvant chemotherapy.
Aim: To estimate the prevalence of endothelial dysfunction in solid cancer patients compared to that in individuals without any malignancies.
Materials and methods: This observational study included 74 patients with solid malignancies, mostly gastrointestinal. Prior to polychemotherapy, all patients were examined for endothelial function of small and large arteries (AngioScan-01, Fiton, Russia) and peripheral artery stiffness (pulse wave contour analysis and occlusion test). The results were compared with those of the Russian population trials of endothelial dysfunction Meridian-RO (Ryazan region) and trial in the rural population of the Krasnodar region.
Results: Compromised vasodilation and smaller arteries tone were found in 64.9% (48/74) of the cancer patients, while impaired vasodilation of larger muscular arteries was present in 94.6% (70/74) of the patients. According to the Meridian-RO trial in the Ryazan region, endothelial dysfunction had been found in 51.2% (n = 341) of women and 52.4% (n = 300) of men, whereas the Krasnodar regional population data had shown it in 68.4% (n = 353) of women and 71.7% (n = 253) of men.
Conclusion: The prevalence of endothelial dysfunction in the patients with solid malignancies, who have not undergone any chemo- or radiation therapy, is significantly higher than in the population of comparable age, conventional cardiovascular risk factors, and comorbidities.
