    Multiple crack propagation in friction stir welded aluminium joints

    This paper is concerned with the simulation of crack propagation in friction stir welded butt joints, in order to assess the influence of process induced microstructural alterations and residual stresses on the fatigue behaviour of the assembly. The approach employed is based on the coupled use of the finite element method and the dual boundary element method in order to take advantage of the main capabilities of the two methods. The distribution of the process induced residual stresses has been mapped by means of the contour method. Then, the computed residual stresses field has been superimposed, in a dual boundary element environment, to the stress field as a result of a remote fatigue traction load and the crack growth is simulated. A two-parameter crack growth law, based on the evaluation of two thresholds, for the material being analysed, is used for the crack propagation rate assessment. The stress intensity factors are evaluated using the J-integral technique. Computational results have been compared with experimental data, provided from constant amplitude crack propagation tests on welded samples, showing the subdivision of the overall fatigue life in the two periods of crack initiation and crack propagation

    PIGSPro: prediction of immunoGlobulin structures v2

    PIGSpro is a significant upgrade of the popular PIGS server for the prediction of the structure of immunoglobulins. The software has been completely rewritten in python following a similar pipeline as in the original method, but including, at various steps, relevant modifications found to improve its prediction accuracy, as demonstrated here. The steps of the pipeline include the selection of the appropriate framework for predicting the conserved regions of the molecule by homology; the target template alignment for this portion of the molecule; the selection of the main chain conformation of the hypervariable loops according to the canonical structure model, the prediction of the third loop of the heavy chain (H3) for which complete canonical structures are not available and the packing of the light and heavy chain if derived from different templates. Each of these steps has been improved including updated methods developed along the years. Last but not least, the user interface has been completely redesigned and an automatic monthly update of the underlying database has been implemented.     Sperm ultrastructure and spermiogenesis in two Exogone species (Polychaeta, Syllidae, Exogoninae)

    The spermatozoa of Exogone naidina and E. dispar are characterized by a prominent bell-shaped acrosome, a spheroidal nucleus, and a conventional flagellum. During spermiogenesis, the acrosomal vesicle undergoes conspicuous modifications leading to its final bell shape with a posterior opening. The subacrosomal material initially shows radiating filaments but in mature sperms it appears as a meshwork of electron-opaque material. The acrosomal axis is oblique with respect to the main longitudinal sperm axis. The chromatin is arranged in electronopaque strands in the early spermatids, then becomes amorphous, and is finally organized in filaments in mature sperms. Centrioles are orthogonally arranged beneath the nucleus and fibers radiate from the distal centriole to contact the plasma membrane and the single mitochondrion. The latter is located eccentrically on the side of the nucleus opposite the acrosome. A disk-shaped structure is evident beneath the distal centriole. The flagellar axoneme has a 9+2 microtubule pattern. A conspicuous glycocalyx surrounds the flagellar plasma membrane, and an electron-lucent space is present between these two structures at the distal tip of the flagellum. We compare the sperm morphology of these two species of Exogone with that described in other members of the subfamily Exogoninae. The fine structure of these two species supports the occurrence of an ent-aquasperm type within Exogoninae, in accordance with the brood strategy present within this subfamily. The mode of reproduction is of taxonomic importance for defining subfamilies within Syllidae, and is likely also of phylogenetic significance. Because epitoky is probably plesiomorphic, the ent-aquasperm type found in Exogoninae can be considered a derived feature within Syllidae

    The analysis of groundwater resources is a particularly significant aspect of the economic, social and environmental development of the national territory. This is particularly relevant for the Campania region which, although characterized by the most significant aquifer systems of southern Italy, suffers from critical issues related to the progressive increase in demand and climatic variability at different time scales. In this framework, the lack of data concerning the survey of springs, including the minor ones, and of historical discharge easurements represents the main limitation to a more comprehensive regional hydrogeological characterization. The only source of historical data regarding the systematic and comprehensive survey of springs and discharge measurement is the Publication No. 14 of the National Hydrographic Service of the Ministry of Public Works “The Italian springs. List and description” reporting measures made between the 1920s and 1940s which was published in distinct volumes for each compartment. Despite its potential relevance, this source has so far been little used in regional hydrogeological studies. In this paper, a comparative analysis among data of springs derived from the Publication No. 14 and from measurement campaigns made by the Cassa per il Mezzogiorno (Special Project 26), between the 1960s and 1980s for main springs, was carried out for the Campania region. The information available from each source was validated through a cross-check, by means of a comparison of coordinates and a statistical analysis of the characterizing parameters. The new dataset allowed to expand the hydrogeological regional characterization with a higher number of springs, including the minor ones. The results obtained recognize the Publication No. 14 of the National Hydrographic Service as an important source of data to not be overlooked, especially in a condition of historical data shortage, by which can be both carried out regional hydrogeological and temporal analyses as well as identified integrative groundwater resources

    CryoPlasty therapy of the superficial femoral and popliteal arteries: A reappraisal after 44 months' experience

    ObjectivesLong-term patency remains a significant hurdle in the minimally invasive treatment of arteriosclerosis in the superficial femoral (SFA) and popliteal arteries. CryoPlasty therapy (PolarCath, Boston Scientific Corp, Natick, Mass) is a novel approach designed to significantly reduce injury, elastic recoil, neointimal hyperplasia, and constrictive remodeling. The technique combines the dilatation forces of percutaneous transluminal angioplasty (PTA) with cold thermal energy applied to the plaque and vessel wall. We previously reported a technical success rate of 96% and a 12-month freedom from restenosis rate of 82.2%. However, a review of the original cohort supplemented by experience with a further 47 lesions has demonstrated less desirable results.MethodsFrom December 2003 through July 2007, 92 lesions in 64 consecutive patients were treated and followed up for a median of 16 months with statistically significant follow-up at 24 months.ResultsThe immediate technical success rate was 88%. Nine stents were immediately required after unsuccessful CryoPlasty (9.8%) five of which were as a result of a dissection. No unanticipated adverse events occurred, specifically, no thrombus, acute occlusions, distal embolizations, aneurysms, or groin complications. Vascular calcification was responsible for technical failure in six of the 11 immediately unsuccessful procedures. Freedom from restenosis for successfully treated lesions was 57% and 49% at 12 and 24 months, respectively. CryoPlasty of heavily calcified lesions, vein graft lesions, and in-stent stenosis faired poorly. Excluding these lesions from analysis would have resulted in an immediate success of 94% (81 of 86) and freedom from restenosis of 61% and 52% at 12 and 24 months, respectively. However, on an intention-to-treat basis, freedom from restenosis was 47% and 38% at 12 and 24 months, and CryoPlasty added approximately $1700 to the cost of each procedure.ConclusionAnalysis of this expanded, longer-term data suggests that our earlier, smaller study provided an overly optimistic appraisal of the benefits of CryoPlasty. It is possible that a larger analysis might have identified a subset of patients or lesions that would benefit from CryoPlasty, but considering the additional cost, we no longer use this technique in our practice

    Combined TRPC3 and TRPC6 blockade by selective small-molecule or genetic deletion inhibits pathological cardiac hypertrophy

    Chronic neurohormonal and mechanical stresses are central fea-tures of heart disease. Increasing evidence supports a role forthe transient receptor potential canonical channels TRPC3 andTRPC6 in this pathophysiology. Channel expression for both is nor-mally very low but is increased by cardiac disease, and geneticgain- or loss-of-function studies support contributions to hypertro-phy and dysfunction. Selective small-molecule inhibitors remainscarce, and none target both channels, which may be useful giventhe high homology among them and evidence of redundant sig-naling. Here we tested selective TRPC3/6 antagonists (GSK2332255Band GSK2833503A; IC50,3–21 nM against TRPC3 and TRPC6) andfound dose-dependent blockade of cell hypertrophy signaling trig-gered by angiotensin II or endothelin-1 in HEK293T cells as well as inneonatal and adult cardiac myocytes. In vivo efficacy in mice andrats was greatly limited by rapid metabolism and high protein bind-ing, although antifibrotic effects with pressure overload were ob-served. Intriguingly, although gene deletion of TRPC3 or TRPC6alone did not protect against hypertrophy or dysfunction frompressure overload, combined deletion was protective, support-ing the value of dual inhibition.     Mass assembly and AGN activity at z1.5z\gtrsim1.5 in the dense environment of XDCPJ0044.0-2033

    XDCP0044.0-2033 is the most massive galaxy cluster known at z>1.5 and its core shows a high density of galaxies which are experiencing mergers and hosting nuclear activity. We present a multi-wavelength study of a region located 157 kpc from the center of this galaxy cluster, for which we have photometric and spectroscopic multi-wavelength observations (high resolution HST images in F105W, F140W and F160W bands, NIR KMOS data in H and YJ bands and Chandra ACIS-S X-ray data). Our main goal is to investigate the environmental effects acting on the galaxies inhabiting this high density region. We find that the analyzed region hosts at least nine different sources, six of them confirmed to be cluster members within a narrow redshift range 1.5728<z<1.5762. These sources form two different complexes at a projected distance of \sim13 kpc, which are undergoing merging on an estimated timescale off 10-30 Myr. One of the sources shows the presence of a broad H alpha emission line and is classified as Type 1 AGN. This AGN is associated to an X-ray point-like source, whose emission appears moderately obscured (with intrinsic absorption NH1022cm2N_{H} \sim 10^{22} cm^{-2}) and hosts a relatively massive black hole with mass MBH107MM_{BH} \sim 10^{7} M_{\odot}, which is accreting with an Eddington ratio of \sim0.2. We conclude that the analyzed region is consistent with being the formation site of a secondary BCG. These findings, together with an in-depth analysis the X-ray morphology of the cluster, suggest a merging scenario of the entire cluster, with two massive halos both harbouring two rapidly evolving BCGs on the verge of being assembled. Our results are also consistent with the scenario in which the AGN phase in member galaxies is triggered by gas-rich mergers, playing a relevant role in the formation of the red sequence of elliptical galaxies observed in the center of local galaxy clusters
