Use of gold
nanoparticles functionalization experiment was conducted in a biotechnology summer course at UFOP as a model for the introduction
of the laboratory practice in nanotechnology for postgraduate courses in the areas of chemistry and biotechnology. The gold
nanorods were synthesized by the seed method and then, functionalized with anti-IL-6 antibodies using the reagents EDAC/NHS
and polyethyleneimine (PEI). This nanocompound was tested against the binding with the specific antigen (IL-6) and changes in the
longitudinal plasmon absorption spectrum showed the coupling efficiency, which was also verified by the decrease in zeta potential.
The experiment was satisfactory, with a positive feedback from participants, and could be implemented in nanotechnology practical
classes from postgraduate courses, as a way for improve education in the emergent area of nanobiotechnology
A new appraisal of sri lankan bb zircon as a reference material for LA-ICP-MS U-Pb geochronology and Lu-Hf isotope tracing.
A potential zircon reference material (BB zircon) for laser abla tion-in ductively coupled plasma-mass spectrometry (LA-ICP-M S)U-Pb geochronology and Hf isotope geochemistry is described. A batch of twenty zircon megacrysts (0.5?1.5 cm3) from SriLanka was studied. Within-grain rare earth element (REE) compositions are largely homogeneous, albeit with somevariation seen between fractured and homogeneous domains. Excluding fractured cathodoluminescence bright domains,the variation in U content for all analysed crystals ranged from 227 to 368 lgg-1and the average Th/U ratios werebetween 0.20 and 0.47. The Hf isotope composition (0.56?0.84 g/100 g Hf) is homogeneous within and between thegrains ? mean176Hf/177Hf of 0.281674 ? 0.000018 (2s). The calculated alpha dose of 0.59 3 1018g-1for a numberof BB grains falls within the trend of previously studied, untreated zircon samples from Sri Lanka. Aliquots of the samecrystal (analysed by ID-TIMS in four different laboratories) gave consistent U-Pb ages with excellent measurementreproducibility (0.1?0.4% RSD). Interlaboratory assessment (by LA-ICP-MS) from individual crystals returned results that arewithin uncertainty equivalent to the TIMS ages. Biodistribution and toxicological study of PEGylated single-wall carbon nanotubes in the zebrafish (Danio rerio) nervous system
Nanotechnology has been proven to be increasingly compatible with pharmacological and biomedical applications.
Therefore, we evaluated the biological interactions of single-wall carbon nanotubes functionalized with
polyethylene glycol (SWNT-PEG). For this purpose, we analyzed biochemical, histological, behavioral and
biodistribution parameters to understand how this material behaves in vitro and in vivo using the fish
Danio rerio (zebrafish) as a biological model. The in vitro results for fish brain homogenates indicated that
SWNT-PEG had an effect on lipid peroxidation and GSH (reduced glutathione) content. However, after intraperitoneal
exposure, SWNT-PEG proved to be less biocompatible and formed aggregates, suggesting that the PEG
used for the nanoparticle functionalization was of an inappropriate size for maintaining product stability in a biological
environment. This problem with functionalization may have contributed to the low or practically absent
biodistribution of SWNT-PEG in zebrafish tissues, as verified by Raman spectroscopy. There was an accumulation
of material in the abdominal cavity that led to inflammation and behavioral disturbances, as evaluated by a histological
analysis and an open field test, respectively. These results provide evidence of a lack of biocompatibility
of SWNTs modified with short chain PEGs, which leads to the accumulation of the material, tissue damage and
behavioral alterations in the tested subject
Delayed colorectal cancer care during covid-19 pandemic (decor-19). Global perspective from an international survey
Background
The widespread nature of coronavirus disease 2019 (COVID-19) has been unprecedented. We sought to analyze its global impact with a survey on colorectal cancer (CRC) care during the pandemic.
Methods
The impact of COVID-19 on preoperative assessment, elective surgery, and postoperative management of CRC patients was explored by a 35-item survey, which was distributed worldwide to members of surgical societies with an interest in CRC care. Respondents were divided into two comparator groups: 1) ‘delay’ group: CRC care affected by the pandemic; 2) ‘no delay’ group: unaltered CRC practice.
Results
A total of 1,051 respondents from 84 countries completed the survey. No substantial differences in demographics were found between the ‘delay’ (745, 70.9%) and ‘no delay’ (306, 29.1%) groups. Suspension of multidisciplinary team meetings, staff members quarantined or relocated to COVID-19 units, units fully dedicated to COVID-19 care, personal protective equipment not readily available were factors significantly associated to delays in endoscopy, radiology, surgery, histopathology and prolonged chemoradiation therapy-to-surgery intervals. In the ‘delay’ group, 48.9% of respondents reported a change in the initial surgical plan and 26.3% reported a shift from elective to urgent operations. Recovery of CRC care was associated with the status of the outbreak. Practicing in COVID-free units, no change in operative slots and staff members not relocated to COVID-19 units were statistically associated with unaltered CRC care in the ‘no delay’ group, while the geographical distribution was not.
Conclusions
Global changes in diagnostic and therapeutic CRC practices were evident. Changes were associated with differences in health-care delivery systems, hospital’s preparedness, resources availability, and local COVID-19 prevalence rather than geographical factors. Strategic planning is required to optimize CRC care
