    Development of high resolution melting analysis for the diagnosis of human malaria

    Molecular detection has overcome limitations of microscopic examination by providing greater sensitivity and specificity in Plasmodium species detection. The objective of the present study was to develop a quantitative real-time polymerase chain reaction coupled with high-resolution melting (qRT-PCR-HRM) assay for rapid, accurate and simultaneous detection of all five human Plasmodium spp. A pair of primers targeted the 18S SSU rRNA gene of the Plasmodium spp. was designed for qRT-PCR-HRM assay development. Analytical sensitivity and specificity of the assay were evaluated. Samples collected from 229 malaria suspected patients recruited from Sabah, Malaysia were screened using the assay and results were compared with data obtained using PlasmoNexTM, a hexaplex PCR system. The qRT-PCR-HRM assay was able to detect and discriminate the five Plasmodium spp. with lowest detection limits of 1–100 copy numbers without nonspecific amplifications. The detection of Plasmodium spp. in clinical samples using this assay also achieved 100% concordance with that obtained using PlasmoNexTM. This indicated that the diagnostic sensitivity and specificity of this assay in Plasmodium spp. detection is comparable with those of PlasmoNexTM. The qRT-PCR-HRM assay is simple, produces results in two hours and enables high-throughput screening. Thus, it is an alternative method for rapid and accurate malaria diagnosis

    a b s t r a c t A five-factor asset-pricing model is employed to estimate the systematic financial risk exposure of airlines in North America, Europe and Asia between 1990 and 2010. Our panel data reveal that the risk to North America airlines is positively related to operating leverage and profitability, but while European and Asian airlines also have risk positively related to operating leverage, their risks are significant negative related to earnings growth. The most important systematic risk determinant for Asian airlines however is their size. Looking at the effects of operating leases and government ownership on Asian airlines' risk, we find that leasing is equally important as size but acts in the opposite sign; operating leverage is not significant while earnings growth is significant only for government owned airlines

    Class algebra provides a natural framework for sharing of ISA hierarchies between users that may be unaware of each other's definitions. This permits data from relational databases, objectoriented databases, and tagged XML documents to be unioned into one distributed ontology, sharable by all users without the need for prior negotiation or the development of a "standard" ontology for each field. Moreover, class algebra produces a functional correspondence between a class's class algebraic definition (i.e. its "intent") and the set of all instances which satisfy the expression (i.e. its "extent"). The framework thus provides assistance in quickly locating examples and counterexamples of various definitions. This kind of information is very valuable when developing models of the real world, and serves as an invaluable tool assisting in the proof of theorems concerning these class algebra expressions.     Ultrasensitive cDNA detection of dengue virus RNA using electrochemical nanoporous membrane-based biosensor

    A nanoporous alumina membrane-based ultrasensitive DNA biosensor is constructed using 5′-aminated DNA probes immobilized onto the alumina channel walls. Alumina nanoporous membrane-like structure is carved over platinum wire electrode of 76 µm diameter dimension by electrochemical anodization. The hybridization of complementary target DNA with probe DNA molecules attached inside the pores influences the pore size and ionic conductivity. The biosensor demonstrates linear range over 6 order of magnitude with ultrasensitive detection limit of 9.55×10−12 M for the quantification of ss-31 mer DNA sequence. Its applicability is challenged against real time cDNA PCR sample of dengue virus serotype1 derived from asymmetric PCR. Excellent specificity down to one nucleotide mismatch in target DNA sample of DENV3 is also demonstrated.Published versio
