Tuz Stresinin Bitkiler Üzerindeki Etkileri ve Geliştirilen Tolerans Mekanizmaları
Jeolojik, hidrolojik ve antropojenik birçok faktörün neden olduğu tuz stresi, bitkilerin hemen hemen tüm gelişme dönemlerinde olumsuz etkilere sebep olan oldukça karmaşık bir abiyotik strestir. Tuz stresinin neden olduğu ozmotik stres, bitki hücresindeki iyonik dengeyi ve genel homeostazı bozar. Bu abiyotik stres, osmotik strese neden olarak bitkilerin kullanabileceği su miktarını kısıtladığı gibi, iyonik stresi de tetikleyerek bitkilerde iyon miktarının toksik seviyelere ulaşmasına neden olmaktadır. Ayrıca bitkilerde reaktif oksijen türlerinin (ROT) artışını tetikleyen ikincil bir oksidatif stres oluşumuna neden olmaktadır. Genel olarak tuz stresine maruz kalan bitkilerde turgor kaybı, stoma iletkenliği ve fotosentez hızında azalma, besin dengesizliği, biyokütlenin azalması ve büyümenin yavaşlaması yaygın etkiler arasında görülür. Bitkiler bu olumsuz etkilerle baş edebilmek ve tuza tolerans oluşturabilmek için tuz alımı ve iyon taşınmasının kontrolü, ozmolitlerin ve antioksidanların sentezi veya birikimini içeren çeşitli savunma mekanizmaları geliştirmişlerdir. Farklı bitkiler, genotipe, adaptasyon kabiliyetine ve bitkinin diğer özelliklerine bağlı olarak tuzluluğa farklı tepkiler vermektedir. Endüstrileşmenin ve insan faaliyetlerinin hızla arttığı küreselleşen dünyada tuzluluğa dayanıklı transgenik bitkilerin geliştirilmesi verimliliği ve ürün kalitesi daha yüksek bitkiler elde etmek için oldukça önemlidir. Yüksek tuz konsantrasyonlarında yaşamlarını sürdürebilen halofitler ise, istenilen gen kaynağının sağlaması açısından tuza toleranslı glikofit bitkilerin yetiştirildiği çoğu transgenik çalışmada model organizmalar olarak kullanılmaktadır
Impact of Some Entomopathogenic Fungi on the Growth of Zea mays L. and Helianthus annuus L.
Entomopathogenic fungi, a promising alternative to conventional insecticides, have been widely used as biocontrol agents for the regulation of pest populations. Furthermore, recent studies have shown that entomopathogenic fungi also have growth-promoting properties in plants. The purpose of this investigation was to assess the effects of indigenous entomopathogenic fungi strains, Beauveria bassiana Pa4, Cordyceps fumosorosea KTU-42, and Metarhizium flavoviride As-18, on the germination and early development period of sunflower and maize plants. In this study, it was determined that EPF application did not affect the seed germination percentage in maize, but increased the germination percentage of sunflower seeds. It was determined that M. flavoviride application significantly increased root-stem weight, root length, stem diameter and biomass in sunflower samples compared to other EPF applications and control. In maize, it was determined that EPF applications had a positive effect only on root length. The most effective fungus to increase growth on maize plants was C. fumosorosea KTU-42. On the other hand, the same result was obtained with M. flavoviride As-18 on sunflower plants. The present study reported that these fungi promote plant development and should be considered an important factor in plant production besides pest management
Antimicrobial activity of leaf extracts of Bituminaria sp. genotypes at different growth stages
Plants have secondary metabolites that play a role in defense mechanism are known to have effects on microorganisms. These metabolites and their amounts vary depending upon various factors. Here, we studied the antimicrobial effect of secondary metabolites of Bituminaria sp. genotypes and explored whether this effect changes with different growing stages of the plant. The results show that extracts obtained from 12 B. bituminosa genotypes were not significantly effective on Gram-negative bacteria but highly effective on Gram-positive bacteria and eukaryotic yeast. Of these genotypes, 9 and 12 genotype were more effective than the rest. Observations suggest that the extracts from the plants are more effective during the beginning of flowering stage than the growth and budding stages for the bacteria studied. It could be due to the differences in the nature of the metabolites and their quantity

FARKLI KONSANTRASYONLARDA KALSİYUM KARBONAT (CaCO3) UYGULAMASININ CARTHAMUS TINCTORIUS L.’A (ASTERACEAE) AİT FARKLI VARYETELERİN TOHUM ÇİMLENMESİ ÜZERİNE ETKİLERİ
Bu çalışmada, biyodizel eldesi için yetiştirilmekte
olan iki farklı Carthamus tinctorius L. (Asteraceae) çeşidinin, Remzibey ve
Dinçer, tohumları çalışma materyali olarak kullanılmıştır. Her bir çeşide ait
tohumlar Petri kapları içinde, iki kat kurutma kâğıdı üzerinde, 16 saat
aydınlık/8 saat karanlık fotoperiyotta, 25±1˚C’ta, üçer tekrarlı olmak üzere,
saf sudan ibaret kontrol ve %2, %4, %8, %15, %50 ve %60 oranlarında hazırlanmış
kalsiyum karbonat solüsyonları içinde çimlenmeye bırakılmıştır. Elde edilen veriler
istatistiksel olarak değerlendirildiğinde, Dinçer çeşidinin Remzibey’e göre
kalsiyum karbonata karşı daha dayanıklı olduğu tespit edilmiştir. Ayrıca her

çeşidinin %8’lik kalsiyum karbonat solüsyonunda %4’lük kalsiyum karbonata göre
daha iyi çimlenme gösterdiği belirlenmiştir. Sonuç olarak Carthamus tinctorius’un her iki çeşidinin de nötr topraklarda ve
hafif alkali topraklarda daha iyi çimlenebileceği söylenebilir
PORSUK NEHRİ SUYUNUN CUCUMIS SATIVUS (L.) TOHUMLARININ FİDE GELİŞİMİ ÜZERİNE ETKİLERİNİN BELİRLENMESİ
Porsuk Çayı 448 km uzunluğu ile Sakarya Irmağı’nın en uzun koludur ve Bayatçık Deresi ile Kızıltaş Suyu’nun birleşmesi ile oluşur. Kütahya Ovası’ndan geçip Eskişehir’in güneybatısında yer alan Porsuk-I ve Porsuk-II barajlarında toplandıktan sonra Eskişehir Ovası’ndan ve Eskişehir ilinden geçer ve Sakarya Irmağı’na ulaşır. Porsuk Çayı, Eskişehir’de tarım arazilerinin sulama suyu olarak kullanılmaktadır. Tüm İç Anadolu’da olduğu gibi Eskişehir’de de tarımı yapılan en yaygın bahçe bitkileri arasında Cucumis sativus (L.) yer almaktadır. İnsan beslenmesinde ilk bakışta önemli gibi görünmese de Cucumis sativus; vitaminler, enzimler, mineral maddeler bakımından zengin bir bitkidir. Fakat protein, karbonhidrat ve yağ bakımından fakirdir. Bu çalışmada, tohumların çimlendirilmesinde şehir şebeke suyu ve farklı oranlarda seyreltilmiş, şehir içinde ve şehir dışında belirlenen istasyonlardan alınan Porsuk nehrine ait su örnekleri kullanılmıştır. Fidelerin kök-gövde uzunlukları, kök ve gövdeye ait yaş ve kuru ağırlıklar ölçülerek şebeke suyu ve Porsuk nehrine ait suyun fide gelişimi üzerine etkileri araştırılmıştır. Elde edilen verilere göre, kök uzunluğunun nehir suyunun seyreltme oranı ile doğru orantıda arttığı ve su içeriğinin seyreltme oranı arttıkça azaldığı tespit edilmişti
