    Some conceptual and empirical issues in linguistic theory : an illustration with pronominal clitics

    I would like to discuss a few general conceptual issues in linguistic theory, and see how they bear on some empirical facts about pronominal clitics. In particular, I would like to show that the conception of linguistic theory, justified on independent grounds, limits the class of issues and possible explanations for grammatical properties of specific linguistic expressions. I argue that this is not simply a consequence of a specific conception of grammar, conceived of as a system of principles and rules governing language, but has non-trivial empirical ramifications. Pronominal clitics are a good case study, since their grammatical properties bear on a wide range of facts falling under the purview of principles of phonology, morphology, syntax and semantics

    The impact of High Performance Workplace Systems (HPWS) on workers and unions is a contentious area for debate in the fields of industrial relations and social science in general. Proponents of HPWS claim that one of the benefits for workers is that they enable workers to develop and raise their skill levels. This paper offers a preliminary evaluation of that claim by sketching an updated map of the territory. It concludes that the HPWS literature contains significant weaknesses concerning the definition of skill in explaining what skill development means for workers, individually and collectively

    Honesty, social presence, and self-service in retail

    Retail self-service checkouts (SCOs) can benefit consumers and retailers, providing control and autonomy to shoppers independent from staff. Recent research indicates that the lack of presence of staff may provide the opportunity for consumers to behave dishonestly. This study examined whether a social presence in the form of visual, humanlike SCO interface agents had an effect on dishonest user behaviour. Using a simulated SCO scenario, participants experienced various dilemmas in which they could financially benefit themselves undeservedly. We hypothesised that a humanlike social presence integrated within the checkout screen would receive more attention and result in fewer instances of dishonesty compared to a less humanlike agent. Our hypotheses were partially supported by the results. We conclude that companies adopting self-service technology may consider the implementation of social presence to support ethical consumer behaviour, but that more research is required to explore the mixed findings in the current study

    Biocatalytic conversion of lignin to aromatic dicarboxylic acids in Rhodococcus jostii RHA1 by re-routing aromatic degradation pathways

    The heteropolymer lignin represents an untapped resource for production of renewable aromatic chemicals, if efficient depolymerisation methods can be developed. In this work, the metabolic pathways in Rhodococcus jostii RHA1 for degradation of aromatic lignin breakdown products are re-routed, in order to generate an aromatic dicarboxylic acid product that could be used for bioplastic synthesis. Protocatechuic acid is normally metabolised via ortho-cleavage to the ÎČ-keto-adipate pathway. Insertion of recombinant genes for protocatechuate 4,5-dioxygenase or protocatechuate 2,3-dioxygenase into R. jostii RHA1, followed by ammonia cyclisation of the extradiol cleavage products, generates pyridine 2,4-dicarboxylic acid or pyridine 2,5-dicarboxylic acid bioproducts in yields of 80–125 mg L−1 when grown on minimal media containing 1% wheat straw lignocellulose

    Biochemical characterization of the retinoid isomerase system of the eye

    Journal ArticleWe have previously shown that membranes from the retinal pigment epithelium can transform added all-trans-retinol into a mixture of 11-cis-retinoids, demonstrating the "missing reaction" in the visual cycle for the first time (Bernstein, P. S., Law, W. C., and Rando, R. R. (1987) Proc. Natl. Acad. Sci. U.S.A. 84, 1849-1853). In this article, this isomerase activity is further characterized. Double-label experiments with [15-3H]- and [15-14C]all-trans-retinol as the substrate show that the tritium label is retained in the 11-cis-retinol and 11-cis-retinyl palmitate products. This requires that isomerization occur at the alcohol level of oxidation. All-trans-retinyl esters, such as the palmitate, acetate, butyrate, and hexanoate esters, are not directly transformed into their 11-cis counterparts by the membranes. The data are consistent with the presence of an all-trans-retinol isomerase enzyme system or enzyme complex, which produces 11-cis-retinol. Other isomeric retinols were tested for substrate activity. Neither 9-cis-retinol(al) nor 13-cis-retinol were processed by the isomerase. Since the membranes containing the isomerase possess other retinol metabolizing activities, such as retinyl ester synthetase and dehydrogenase activities, further purification was attempted. Appreciable quantities of all detergents tested led to the disappearance of isomerase activity, and high salt or EDTA did not dissociate isomerase activity from the membranes. However, extensive sonication of the membranes did produce a 100,000 x g supernatant fraction of light membranes depleted of other all-trans-retinol processing activities. The isomerase activity in these membranes was saturable with all-trans-retinol, as required for a biologically significant process, and showed a Vmax of 5 pmol/h/mg of protein, a KM of 0.8 microM, and a pH optimum of 8. The isomerase was destroyed by proteinase K, by phospholipase C, by heating, or by ethanol at concentrations greater than 1%. The addition of high energy compounds, such as MgATP, MgGTP, or palmitoyl-CoA, did not appear to stimulate isomerase activity in the 100,000 x g supernatant
