Cytotoxicity and genotoxicity assessment of the extract and lectins from Moringa oleifera Lam. Seeds / Avaliação da citotoxicidade e genotoxicidade do extrato e lectinas das sementes de Moringa oleifera Lam

Moringa oleifera seeds are used globally as a treatment for water and contain the lectins cMoL and WSMoL, which display coagulant activity. In this study, we sought to determine the cytotoxicity and genotoxicity of the M. oleifera seed extract (SE), prepared with the same procedure that people use for treating water, as well as cMoL and WSMoL, in human peripheral blood mononuclear cells (PBMCs). Cell viability was assessed using the 3-(4,5-dimethylthiazol-2-yl)2,5-diphenyltetrazolium bromide (MTT) assay, while genotoxicity was evaluated using the comet assay, with cell nucleoids categorized in classes ranging from 0 (without damage) to 4 (maximum damage). The PBMCs treated with SE, cMoL, and WSMoL displayed viability higher than 60% in treatments with concentrations up to 100 µg/mL. In addition, SE and cMoL displayed low genotoxicity owing to the detection of nucleoids in class 1. However, the number of nucleoids in all classes increased when 50 and 100 µg/mL of WSMoL was administered, reaching a damage frequency of 50.0%. Although M. oleifera SE, cMoL, and WSMoL were not cytotoxic to PBMCs after 24 h of exposure, dose-dependent genotoxic effects were observed, especially with WSMoL. These findings indicate that caution must be exercised when selecting a lectin/extract concentration for water treatment

Analgesia pós-operatoria com Paracetamol injetável em pacientes oncológicos submetidos a tireoidectomia: Postoperative analgesia with injectable Paracetamol in cancer patients undergoing thyroidectomy

A dor pós-operatória de tireoidectomias totais é a principal queixa relatada pelos pacientes. É um sintoma importante que, se não modulado corretamente, pode se tornar uma complicação frequente que prolonga o tempo de internação hospitalar. Dessa forma, é essencial o manejo álgico eficiente no perioperatório. Nesse contexto, o paracetamol injetável se tornou recentemente uma alternativa adicional na analgesia multimodal, com a disponibilização da sua formulação endovenosa no Brasil (2020). O objetivo inicial desse estudo é avaliar a eficácia do paracetamol injetável e secundariamente o seu efeito poupador de opioides em pacientes oncológicos submetidos a tireoidectomia total em uma fundação que trata do câncer. Trata-se de um estudo longitudinal, experimental, observacional e descritivo, no qual pelo menos 20 pacientes submetidos à tireoidectomia total receberão paracetamol injetável e serão submetidos a um questionário, visando obter dados que auxiliem na identificação e na contribuição do paracetamol injetável na analgesia pós-operatória multimodal

Hexane extract from Spondias tuberosa (Anacardiaceae) leaves has antioxidant activity and is an anti-Candida agent by causing mitochondrial and lysosomal damages

Abstract Background Spondias tuberosa is a plant that produces a fruit crop with high economic relevance at Brazilian Caatinga. Its roots and leaves are used in folk medicine. Methods Chemical composition of a hexane extract from S. tuberosa leaves was evaluated by thin-layer chromatography (TLC), high-performance liquid chromatography (HPLC) and 1H nuclear magnetic resonance (NMR). Antioxidant potential was investigated by DPPH and ABTS assays. Antifungal action on Candida species was evaluated determining the minimal inhibitory concentration (MIC50) and putative mechanisms were determined by flow cytometry analysis. In addition, hemolytic activity on human erythrocytes was assessed and the concentration required to promote 50% hemolysis (EC50) was determined. Results Phytochemical analysis by TLC showed the presence of flavonoids, hydrolysable tannins, saponins and terpenes. The HPLC profile of the extract suggested the presence of gallic acid (0.28 ± 0.01 g%) and hyperoside (1.27 ± 0.01 g%). The representative 1H NMR spectrum showed saturated and unsaturated fatty acids among the main components. The extract showed weak and moderate antioxidant activity in DPPH (IC50: 234.00 μg/mL) and ABTS (IC50: 123.33 μg/mL) assays, respectively. It was able to inhibit the growth of C. albicans and C. glabrata with MIC50 of 2.0 and 0.078 mg/mL, respectively. The treatment of C. glabrata cells with the extract increased levels of mitochondrial superoxide anion, caused hyperpolarization of mitochondrial membrane, and compromised the lysosomal membrane. Weak hemolytic activity (EC50: 740.8 μg/mL) was detected. Conclusion The results demonstrate the pharmacological potential of the extract as antioxidant and antifungal agent, aggregating biotechnological value to this plant and stimulating its conservation
